Interleukin-1beta stimulates matrix metalloproteinase-2 expression via a prostaglandin E2-dependent mechanism in human chondrocytes

IL-1beta is known promote cyclooxygenase-2 (COX- 2) and matrix metalloproteinase-2 (MMP-2) expression. This study focuses on the characterization of the signaling cascade associated with IL-1beta-induced matrix metalloproteinase-2 (MMP-2) regulation in human chondrocytes. The decrease in collagen levels in the conditioned media was prevented by a broad spectrum MMP inhibitor, suggesting that IL-1beta promotes the proteolytic process leading to MMP-2 activation. IL-1beta-related MMP-2 expression was found to be dependent on prostaglandin E2 (PGE2) production. In addition, the induction of COX-2 and MMP-2 was inhibited by the pretreatment of chondrocytes with a SB203580 or Ro 31-8220, indicating the involvement of protein kinase C (PKC) or p38 mitogen-activated protein kinase (MAPK). However, there is no cross-talk between PKC and p38 MAPK in the IL-1beta-induced MMP-2 activation. Taken together, these results demonstrated that IL-1beta induces MMP-2 expression through the PGE2-dependent mechanism in human chondrocytes.     

Fabrication and physical properties of lanthanide oxide glass wasteform for the immobilization of lanthanide oxide wastes generated from pyrochemical process

The pyrochemical process, which uses a dry method to recycle used nuclear fuel generates waste LiCl–KCl salt containing radioactive lanthanide elements. To reuse LiCl–KCl salt, the lanthanide elements are separated through a precipitation method promoted by oxygen sparging and the separated fission product of lanthanide oxide should be fabricated into durable wasteforms sustainable for several 1,000 years to store in a final geological repository. Herein, we report the fabrication of a borosilicate glass based wasteform with a glass matrix of SiO₂–Al₂O₃–B₂O₃ having a high waste loading of 50 wt% lanthanide oxide. Th physical properties of four kinds of wasteforms having a different lanthanide oxide waste composition were evaluated. To investigate the long-term physical stability of each sample having 50 wt% lanthanide oxide waste loading, time–temperature–transformation (TTT) test was conducted at 500 and 700 °C for 60 and 180 h, and the physical properties were evaluated after each TTT test.     

Localizability of the embedding problem with symplectic kernel

In this paper we consider the question of how much information is supplied by local solutions to a global embedding problem for the special case in which the normal subgroup belonging to the given group extension is the projective symplectic group PSp(2m, q). It is proved that for suitable Galois extensions K of a given number field k (which constitute part of the data of the embedding problem), the local solutions in a sense determine whether or not an extension K ? K, Galois over k, with $\text{G(}\text{L}\text{K}\text{) ≈ PSp(2m, q)}$, represents a global solution to the embedding problem.

Let G be a finite abelian group, and F a global field of characteristic prime to the order of G. Then there exists a finite extension of F whose class group has a direct summand isomorphic to G.

5-(4-Hydroxy-2,3,5-trimethylbenzylidene) thiazolidine-2,4-dione attenuates atherosclerosis possibly by reducing monocyte recruitment to the lesion

A variety of benzylidenethiazole analogs have been demonstrated to inhibit 5-lipoxygenase (5-LOX). Here we report the anti-atherogenic potential of 5-(4-hydroxy- 2,3,5-trimethylbenzylidene) thiazolidin-2,4-dione (HMB-TZD), a benzylidenethiazole analog, and its potential mechanism of action in LDL receptor-deficient (Ldlr-/-) mice. HMB-TZD Treatment reduced leukotriene B4 (LTB4) production significantly in RAW264.7 macrophages and SVEC4-10 endothelial cells. Macrophages or endothelial cells pre-incubated with HMB-TZD for 2 h and then stimulated with lipopolysaccharide or tumor necrosis factor-alpha (TNF-α) displayed reduced cytokine production. Also, HMB-TZD reduced cell migration and adhesion in accordance with decreased proinflammatory molecule production in vitro and ex vivo. HMB-TZD treatment of 8-week-old male Ldlr-/- mice resulted in significantly reduced atherosclerotic lesions without a change to plasma lipid profiles. Moreover, aortic expression of pro-atherogenic molecules involved in the recruitment of monocytes to the aortic wall, including TNF-α , MCP-1, and VCAM-1, was downregulated. HMB-TZD also reduced macrophage infiltration into atherosclerotic lesions. In conclusion, HMB-TZD ameliorates atherosclerotic lesion formation possibly by reducing the expression of proinflammatory molecules and monocyte/macrophage recruitment to the lesion. These results suggest that HMB-TZD, and benzylidenethiazole analogs in general, may have therapeutic potential as treatments for atherosclerosis.
