Flow-injection determination of 3-hdroxybutyrate in serum with an immobilized 3-hydroxybutyrate dehydrogenase reactor and chemiluminescence detection

A flow-injection system for the determination of 3-hydroxybutyrate in serum is described. 3-Hydroxybutyrate dehydrogenase is immobilized on poly(vinyl alcohol) beads and incorporated in a flow-injection system. 1-Methoxy-5-methylphenazinium methylsulphate reacts with enzymatically generated NADH to give H₂O₂, which is detected chemiluminometrically with the reaction of luminol and hexacynoferrate(III). Serum is diluted and filtered through an ultrafiltration membrane. The system responds linearly to injected samples (80 μl) in the concentration range 0.5–300 μM; the detection limit is 0.1 μM. The within-day relative standard deviation (n = 90) for 58 μM 3-hydroxybutyrate in serum is 0.8%. The maximum throughout is 20 samples per hour. The immobilized enzyme is stable for at least 1 month.     

Optically active 1,2-bis(1-arylhydroxymethyl) ferrocene: a new, efficient chiral ligand for scandium-catalyzed asymmetric Diels-Alder reaction

[reaction: see text] The Sc(OTf)3/FERRODIOL (2) complex was prepared at -78 degrees C in CH2Cl2 in the presence of 2,6-lutidine and MS 4A. The chiral scandium Lewis acid-catalyzed asymmetric Diels-Alder reaction of cyclopentadiene (3) with 3-acyloxazolidin-2-ones (4) effectively produced the adduct (5) in a high yield with good selectivity, i.e., endo/exo = 90:10 up to 91% ee (endo).     

Flow-injection determination of d-mannitol with immobilized mannitol dehydrogenase

A flow-injection system is described for the determination of d-mannitol. Mannitol dehydrogenase is immobilized on poly(vinyl alcohol) beads and packed in a column (5 cm × 4 mm i.d.). The NADH formed is detected fluorimetrically. The response is linear between 5 × 10^?7 and 1 × 10^?4 M mannitol and the detection limit is 1 × 10^?7 M. The throughput is 30 samples per hour. The reactor is stable for at least 8 weeks.     

Analysis of Salmonella lipopolysaccharides by sodium deoxycholate-polyacrylamide gel electrophoresis

Lipopolysaccharides (LPSs) were analyzed by polyacrylamide gel electrophoresis (PAGE) in the presence of sodium deoxycholate (DOC) or sodium dodecyl sulphate (SDS) and the results obtained compared. Two-dimensional PAGE of the S form of LPS revealed that, with SDS, bands which appeared to be single after the one-dimensional experiment were resolved into several bands after two-dimensional electrophoresis. In the presence of DOC, however, a diagonal of single bands was obtained in one-dimensional electrophoresis indicating optimum resolution. The high quality of resolution by DOC-PAGE was constant for amounts of LPS up to 20 micrograms. Finally, DOC-PAGE does not require boiling of the samples in DOC, which may be an advantage over SDS-PAGE.     

Synthesis and central nervous system stimulant activity of 5,8-methanoquinazolines fused with imidazole,thiadiazole, pyrimidine and 1,3,5-triazine

5,8-Methanoquinazolines fused with imidazoles 4a-4b , thiadiazoles 5–6 , pyrimidines 7, 9, 11 and 12 , and 1,3,5-triazine 13 were prepared starting from (5R,8S)-2-amino-8,9,9-trimethyl-5,6,7,8-tetrahydro-5,8-methanoquinazoline 3 . Most compounds possessed central nervous system stimulant activities.     

Synthesis and structural characterization of silanethiolato complexes having tert-butyldimethylsilyl and trimethylsilyl groups

Treatment of cyclotrisilathiane (Me2SiS)3 with 3 equiv. of RLi (R = Me, But) in hexane-Et2O afforded the lithium silanethiolates LiSSiMe2R, and the tmeda adduct [(tmeda)LiSSiMe2But]2 1 (tmeda =N,N,N',N'-tetramethylethylenediamine) was isolated in the case of R = But. Reaction of Fe(CH3CN)2(CF3SO3)2, CoCl2, and [Cu(CH3CN)4](PF6) with 1 gave rise to the silanethiolato complexes M(SSiMe2But)2(tmeda)(M = Fe 2, Co 3), and [Cu(SSiMe2But)]4 4, respectively. Complexes (C5H5)2Ti(SSiMe2R)2(R = Me 5, But 6) and Ni(SSiMe2R)2(dppe)[R = Me 7, But 8; dppe = 1,2-bis(diphenylphosphino)ethane] were prepared from treatments of (C5H5)2TiCl2 and NiCl2(dppe) with the corresponding lithium silanethiolates. Complex 7 readily reacted with (C5H5)TiCl3 to produce the Ti-Ni heterobimetallic compound (C5H5)TiCl(mu-S)2Ni(dppe) 9, in which silicon-sulfur bond cleavage took place. Characterization of all compounds through spectroscopic techniques and elemental analyses are also described. X-Ray structural data for compounds 1 and 3-9 are reported.     

Application of solvent extraction and synchronous spectrofluorimetry to the determination of the formation constant for a 1:1 complex of carbazole withβ-cyclodextrin in water-dimethyl sulfoxide medium

Extraction of carbazole in heptane was performed at 25±1°C with an aqueous dimethyl sulfoxide (DMSO) medium containing -cyclodextrin (CD) at consecutive concentrations in the range of 0–10 mM. The fluorescence intensity of carbazole remaining in the heptane phase was measured by synchronous scanning fluorimetry. The apparent formation constant (K _f) for a 1:1 carbazole: CD inclusion complex in water-DMSO medium was determined by using a linear plot of the distribution ratio calculated from the fluorescence intensities vs. the -CD concentration. The values thus obtained ranged from 477 M^–1 in a 10% v/v DMSO medium to 12.1 M^–1 in a 60% v/v medium. Good linear relationships were observed between logK _f and the DMSO concentration ([DMSO]), and also between logK _f and the logarithm of the distribution coefficient (K _d) for carbazole. The formation constant in 100% water was estimated to be approximately 1.0×10³ M^–1 on the basis of the logK _f vs. [DMSO] and the logK _f vs. logK _d correlations.
