Now it is well known, antioxidant may affect the interaction of anticancer drugs and DNA. This study aims to investigate the interaction between Idarubicin and DNA and the effect of caffeic acid on this interaction. Disposable, inexpensive, easy handle electrodes were used in this study to investigate the interaction of idarubicin and DNA electrochemically. Idarubicin (IDR) is an anthracyline antitumor antibiotic, which used against one or more types of leukemia. Electrochemical behaviour of IDR was investigated by using cyclic voltammetry. The interaction between anticancer drug, IDR and calf thymus double‐stranded DNA (ctdsDNA) was investigated by using differential pulse voltammetry (DPV) technique in the absence and presence of caffeic acid.  相似文献

Present knowledge suggests that in glioblastoma multiforme the value of the apparent diffusion coefficient (ADC) is elevated in the solid part and hyperintense in T1, in spite of the elevated cellularity, and also in areas where peritumoral vasogenic edema is present. The purpose of our study has been to verify in vivo if the ADC increases in areas of solid tumor because of an increased presence of edema, like it happens in areas surrounding the tumor. Sixteen patients with histologically verified glioblastoma multiforme underwent a magnetic resonance (MR) examination with sequences: T1-weighted pre and post contrast, diffusion-weighted at b = 0 and b = 1000 s/mm(2), perfusion-weighted. One hundred sixty-five regions of interest (ROI) have been obtained for all set of patients. In each ROI we have estimated 4 parameters: ADC, intensity of T2-signal normalised to the white matter (SI(T2W)(n)), regional cerebral blood volume (rCBV), T1-signal enhancement (E%). With the SI(T2W)(n) the presence of edema was estimated. For each pair of measured parameters a statistical test of linear regression on the set of all ROI was made. The overall aim of this paper was to compare the multielement composition and antioxidant capacity of two Montenegrin Merlot wines obtained from specific vine clones (VCR1 and VCR 101) along with commercial Merlot wine throughout the consecutive vintages in 2010 and 2011. Elemental composition was analysed using inductively coupled plasma optical emission spectrometry (ICP-OES) and inductively coupled plasma mass spectrometry (ICP-MS). Additionally, antioxidant capacity was assessed by cyclic voltammetry. VCR 1 wine from 2011 stood out for its elemental composition. On the other hand, antioxidant capacity of VCR 101 wines was the highest one for the both vintages. According to the experimental data obtained, all three wines are good source of essential elements and products with a significant antioxidant activity and specific geographical origin.  相似文献

In this study, methylene blue (MB) imprinted microbeads were synthesized and characterized by Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM).Differential pulse voltammetric responses of carbon paste electrodes modified with MB imprinted polymer were used to evaluate the adsorption and selectivity features of the polymer. For selectivity studies two basic dyestuffs (thionine blue and toluidine blue) which have similar structure to MB were chosen. Comparison of the voltammetric responses obtained with pure carbon paste and carbon paste modified with either imprinted or nonimprinted polymer electrodes revealed that MB imprinted polymer presented a higher selectivity to the template molecule MB in contrast to structurally similar molecules, thionine blue and toluidine blue.  相似文献

We present a graphene oxide (GO) integrated disposable electrochemical sensor for the enhanced detection of nucleic acids and the sensitive monitoring of the surface-confined interactions between the anticancer drug mitomycin C (MC) and DNA. Interfacial interactions between immobilized calf thymus double-stranded (dsDNA) and anticancer drug MC were investigated using differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS) techniques. Based on three repetitive voltammetric measurements of 120 μg mL(-1) DNA immobilized on GO-modified electrodes, the RSD % (n = 3) was calculated as 10.47% and the detection limit (DL) for dsDNA was found to be 9.06 μg mL(-1). EIS studies revealed that the binding of the drug MC to dsDNA leads to a gradual decrease of its negative charge. As a consequence of this interaction, the negative redox species were allowed to approach the electrode, and thus increase the charge transfer kinetics. On the other hand, DPV studies exploited the decrease of the guanine signal due to drug binding as the basis for specifically probing the biointeraction process between MC and dsDNA.  相似文献

In this study, SnO2 nanoparticles (SNPs)-poly(vinylferrocenium) (PVF+) modified single-use graphite electrodes were developed for electrochemical monitoring of DNA hybridization. The surfaces of polymer modified and polymer-SNP modified pencil graphite electrodes (PGEs) were firstly characterized by using SEM analysis. The electrochemical behaviours of these electrodes were also investigated using the differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS) techniques. The polymer-SNP modified PGEs were then tested for the electrochemical sensing of DNA based on the changes at the guanine oxidation signals. Experimental parameters, such as; different modifications in DNA oligonucleotides, DNA probe concentrations were examined to obtain more sensitive and selective electrochemical signals for nucleic acid hybridization. After optimization studies, DNA hybridization was investigated in the case of complementary of hepatitis B virus (HBV) probe, mismatch (MM), and noncomplementary (NC) sequences.  相似文献

The effect of the SnO(2) nanoparticles (SNPs) on the behaviour of voltammetric carbon paste electrodes were studied for possible use of this material in biosensor development. The electrochemical behaviour of SNP modified carbon paste electrodes (CPE) was first investigated by using cyclic voltammetry (CV), differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS) techniques. The performance of the SNP modified electrodes were compared to those of unmodified ones and the parameters affecting the response of the modified electrode were optimized. The SNP modified electrodes were then tested for the electrochemical sensing of DNA purine base adenine to explore their further development in biosensor applications.  相似文献

The interaction of PLGA-chitosan Nanoplexes with ocular mucosa was investigated ex vivo and in vivo to assess their potential as ocular delivery system. Fluorescent Rhodamine Nanoplexes (Rd-Nanoplexes) were prepared by ionotropic gelation method. The size and morphology of Nanoplexes was investigated by TEM, SEM and PCS. The corneal retention, uptake and penetration of Nanoplexes were analyzed by spectrofluorimetry and confocal microscopy. Corneas from Rd-Nanoplexes-treated rabbits were evaluated for the in vivo uptake and ocular tolerance. The Nanoplexes prepared were round with a mean diameter of 115.6±17nm and the encapsulation efficiency of Rd was 59.4±2.5%. Data from ex vivo and in vivo studies showed that the amounts of Rd in the cornea were significantly higher for Nanoplexes than for a control Rd solution, these amounts being fairly constant for up to 24h. Confocal microscopy of the corneas revealed paracellular and transcellular uptake of the Nanoplexes. The uptake mechanism postulated was adsorptive-mediated endocytosis and opening of the tight junctions between epithelial cells. No alteration was microscopically observed after ocular surface exposure to Nanoplexes. Taken together, these data demonstrate that Nanoplexes are potentially useful as ocular drug carriers.  相似文献

In the present study a chitosan/ionic liquid modified pencil graphite electrode (CHIT‐IL‐PGEs) was developed for the first time for enhanced electrochemical monitoring of nucleic acid, and the interaction of the anticancer drug Mitomycin C (MC) and calf thymus double stranded DNA (dsDNA) by measuring the oxidation signals of MC and guanine in the same voltammetric scale. Differential pulse voltammetry, cyclic voltammetry and electrochemical impedance spectroscopy techniques were used to evaluate the performance of the CHIT‐IL based biosensor on electrochemical monitoring of DNA, and drug‐DNA interaction. The experimental parameters, IL, dsDNA and MC concentration and the interaction time were then optimized.  相似文献
