A new HPLC method for simultaneous determination of neopterin, creatinine, kynurenine and tryptophan in human serum was developed and validated. Monolithic stationary phase's technology (two monolithic columns RP-18e were connected with guard monolithic cartridge 4.6 mm × 50 mm + 3.0 mm × 100 mm and 4.6 × 10 mm) and special auto sampler for micro titration plates (samples are storage in dark cooled place protected against evaporation) were combined with easy sample preparation step. As mobile phase 15 mmol/L phosphate buffer at pH 4.50 was used. Neopterin and tryptophan were detected using fluorescent detection and kynurenine and creatinine were detected by diode-array detection. This work describes a novel methodology for the recognition of brandies based on direct injection of a raw sample followed by GC-MS analysis. Direct injection was chosen for its simplicity and the fact that the composition of the samples analysed remains unchanged compared to original brandy. The repeatability of the analytical procedure was evaluated by a comparison of the peak areas for randomly selected compounds obtained from 10 parallel measurements. A novel chemometric procedure was investigated in order to separate the samples studied on the basis of their geographical origin, processing technology or maturation time. In this procedure, a principal component analysis was applied to full chromatograms to select the time interval that shows the significant differences between the samples studied. It was shown that the chromatogram recorded at 36–39 min bore the maximal differences, hence it could be used to classify the brandy samples. The chromatographic peaks found within this time interval were identified and their peak areas determined. These compounds could be used as specific markers for determining geographical origin or processing technology. 相似文献

This study was aimed at testing the possibility of using a biotrickling filter for the treatment of waste air resulting from animal farming. For this purpose, a laboratory and pilot biotrickling filter with a moving bed were constructed. In the first case, the removal of ammonia emissions from waste air, by means of ammonia nitrogen utilization by present micro-organisms under laboratory conditions, was tested. The mass load of the biotrickling filter was gradually increased by slowly increasing the ammonia concentration. In the second case, the pilot biotrickling filter with a moving bed was installed inside a pig farm and it was tested under real operation conditions. In the first case, the highest value of the laboratory biotrickling filter’s elimination capacity was 6.2 gNH3 m−3 of filling h−1. The elimination capacity of the pilot filter was 1.6 gNH3 m−3 of filling h−1. The source of carbon for the present micro-organisms was a 1% sucrose solution, which simulated waste water with high content of CODCR. A novel kind of fluorescent surfactant having 7-hydroxylcoumarin group in the long alkyl chain was synthesized. The critical micelle concentration (CMC), surface tension (γcmc) at CMC and absorption, fluorescence properties of this product were determined. From the variations of fluorescence spectra in different solvents, it is observed that the polarity and dielectric constant of the solvents play important roles in the maximum fluorescence intensity and wavelength. Moreover, the surprised exhibition of two fluorescence bands in neutral and alkaline solutions has been attributed to the superexciplex formation of the product molecules. Also, the lower product concentration measuring the fluorescence properties as well as the supposed configuration of hydrogen bond of the product indicate that the larger aggregations cannot exist in alkaline solutions. Biomarkers, 25‐hydroxyvitamin D3 and 25‐hydroxyvitamin D2, are important indicators of the vitamin D general status and are monitored in several pathophysiological disorders, such as osteoporosis, diabetes, heart disease, etc. A novel ultra‐HPLC with MS/MS methodology for the analysis of 25‐hydroxyvitamin D derivatives coupled with a very simple and highly rapid sample preparation step was developed. Analytical parameters obtained showed linearity (R2) above 0.999 for both vitamins with accuracies between 95.8 and 102%. The LODs were as low as 0.22 and 0.67 nmol/L for 25‐hydroxyvitamin D3 and 25‐hydroxyvitamin D2, respectively. Intra‐assay precision (%RSD) was lower than 4.5%, and inter‐assay precision (%RSD) was lower than 6.5%. The feasibility of the developed methodology to be applied in clinical routine analysis has been proved by its application in blood samples from non‐agenarian patients, patients with familial hypercholesterolemia and patients suffering from age‐related macular degeneration. 相似文献
