Excitation energy transfer from phycobilisomes to photosystem II in high-light adapted cells of Anabaena cylindrica was studied by fluorescence spectroscopy and compared to that of low-light adapted cells. Measurements were made on membrane fragments containing phycobilisomes, photosystem I and II, isolated in 0.75 M K-phosphate. Relative efficiency of 430 to 590 nm light in the excitation of F680 chlorophyll fluorescence was compared in low and high light adapted cells, respectively. The values indicate that light energy absorbed by phycobilisomes is transferred to photosystem II antenna chlorophylls with higher efficiency in high-light adapted cells than in low-light adapted cells. Partial dissociation and uncoupling of energy transfer caused by low ion concentration were different in the membrane fragments isolated from the two kinds of cells and indicated a higher aggregation state of pigment-protein complexes of phycobilisomes in high-light adapted A. cylindrica cells. 相似文献

Two rapid and popular methods—capillary electrophoresis (CE) and high-performance liquid chromatography (HPLC) have been compared
for analysis of cotinine in human urine. Cotinine was analyzed in less than 7 min, with detection limits of 5 and 3.2 ng mL−1 for CE and HPLC, respectively. The performance of the methods was evaluated in terms of sensitivity, specificity, precision,
accuracy, and limits of detection and quantification. Calibration plots were linear in the range 50–4,000 ng mL−1, at least, and mean recoveries were satisfactory for both techniques. The methods were successfully used for quantification

By the interaction of heterocyclic thiols with sulfoxides of 6,6-dihydro- and 6-chloropenicillanates, derivatives of 4-heteryldithio-2-azetidinones have been synthesized, as well as products of their cyclization to form 2-heterylthiomethyl-and 2-halomethyl-substituted penicillanates and an ester of 3-chloro-3-methyl-7-chlorocepham-4-carboxylic acid. Summary A semi-preparative low-pressure liquid-chromatographic procedure using tribenzoylcellulose has been developed for the separation and isolation of the four -bisabolol stereoisomers. Stereochemical assignments were made by HPLC-polarimeter coupling and by using the authentical isomers. This method can be applied to the quality control of chamomile preparations, as it allows the detection of adulterations resulting from admixtures of synthetic -bisabolol. Trimethylsilyl ethers of various hydroxyl-containing thiazole derivatives have been synthesized. The psychotropic activity (in vivo) and the cytotoxicity (in vitro on tumor cell lines HT-1080 and MG-22A) of these ethers and of their unsilylated precursors have been studied. It was discovered that the obtained compounds possess a sedative action. Mass action type deterministic kinetic models of ion channels are usually constructed in such a way as to obey the principle of detailed balance (or, microscopic reversibility) for two reasons: first, the authors aspire to have models harmonizing with thermodynamics, second, the conditions to ensure detailed balance reduce the number of reaction rate coefficients to be measured. We investigate a series of ion channel models which are asserted to obey detailed balance, however, these models violate mass conservation and in their case only the necessary conditions (the so-called circuit conditions) are taken into account. We show that ion channel models have a very specific structure which makes the consequences true in spite of the imprecise arguments. First, we transform the models into mass conserving ones, second, we show that the full set of conditions ensuring detailed balance (formulated by Feinberg) leads to the same relations for the reaction rate constants in these special cases, both for the original models and the transformed ones. 相似文献

Water-soluble double-coated magnetic nanoparticles (NPs) containing cytotoxic decyldimethyl(β-dimethylaminoethoxy)silane methiodide (AA) molecule sorbed at biocompatible magnetic particles, which consist of magnetite pre-coated with oleic acid (OA), have been prepared. X-ray line profile broadening analysis was used for crystallite size determination. The method of magnetogranulometry has been used for determination of diameter of iron oxide magnetic core and magnetic properties of NPs prepared. In vitro cytotoxicity on monolayer tumor cell lines HT-1080 (human fibrosarcoma), MG-22A (mouse hepatoma) and normal mouse fibroblasts (NIH 3T3) has been studied. Sirtuins are NAD+‐dependent deacetylases acting as sensors in metabolic pathways and stress response. In mammals there are seven isoforms. The mitochondrial sirtuin 5 is a weak deacetylase but a very efficient demalonylase and desuccinylase; however, its substrate acyl specificity has not been systematically analyzed. Herein, we investigated a carbamoyl phosphate synthetase 1 derived peptide substrate and modified the lysine side chain systematically to determine the acyl specificity of Sirt5. From that point we designed six potent peptide‐based inhibitors that interact with the NAD+ binding pocket. To characterize the interaction details causing the different substrate and inhibition properties we report several X‐ray crystal structures of Sirt5 complexed with these peptides. Our results reveal the Sirt5 acyl selectivity and its molecular basis and enable the design of inhibitors for Sirt5. 相似文献
