Oxidation of guanosine derivatives by a platinum(IV) complex: internal electron transfer through cyclization

Many transition-metal complexes mediate DNA oxidation in the presence of oxidizing radiation, photosensitizers, or oxidants. The DNA oxidation products depend on the nature of the metal complex and the structure of the DNA. Earlier we reported trans-d,l-1,2-diaminocyclohexanetetrachloroplatinum (trans-Pt(d,l)(1,2-(NH(2))(2)C(6)H(10))Cl(4), [Pt(IV)Cl(4)(dach)]; dach = diaminocyclohexane) oxidizes 2'-deoxyguanosine 5'-monophosphate (5'-dGMP) to 7,8-dihydro-8-oxo-2'-deoxyguanosine 5'-monophosphate (8-oxo-5'-dGMP) stoichiometrically. In this paper we report that [Pt(IV)Cl(4)(dach)] also oxidizes 2'-deoxyguanosine 3'-monophosphate (3'-dGMP) stoichiometrically. The final oxidation product is not 8-oxo-3'-dGMP, but cyclic (5'-O-C8)-3'-dGMP. The reaction was studied by high-performance liquid chromatography, (1)H and (31)P nuclear magnetic resonance, and matrix-assisted laser desorption ionization time-of-flight mass spectrometry. The proposed mechanism involves Pt(IV) binding to N7 of 3'-dGMP followed by nucleophilic attack of a 5'-hydroxyl oxygen to C8 of G and an inner-sphere, 2e(-) transfer to produce cyclic (5'-O-C8)-3'-dGMP and [Pt(II)Cl(2)(dach)]. The same mechanism applies to 5'-d[GTTTT]-3', where the 5'-dG is oxidized to cyclic (5'-O-C8)-dG. The Pt(IV) complex binds to N7 of guanine in cGMP, 9-Mxan, 5'-d[TTGTT]-3', and 5'-d[TTTTG]-3', but no subsequent transfer of electrons occurs in these. The results indicate that a good nucleophilic group at the 5' position is required for the redox reaction between guanosine and the Pt(IV) complex.     

Improved Stabilities of Labeling Probes for the Selective Modification of Endogenous Proteins in Living Cells and In Vivo

To date, various affinity-based protein labeling probes have been developed and applied in biological research to modify endogenous proteins in cell lysates and on the cell surface. However, the reactive groups on the labeling probes are also the cause of probe instability and nonselective labeling in a more complex environment, e. g., intracellular and in vivo. Here, we show that labeling probes composed of a sterically stabilized difluorophenyl pivalate can achieve efficient and selective labeling of endogenous proteins on the cell surface, inside living cells and in vivo. As compared with the existing protein labeling probes, probes with the difluorophenyl pivalate exhibit several advantages, including long-term stability in stock solutions, resistance to enzymatic hydrolysis and can be customized easily with diverse fluorophores and protein ligands. With this probe design, endogenous hypoxia biomarker in living cells and nude mice were successfully labeled and validated by in vivo, ex vivo, and immunohistochemistry imaging.     

The electronic band structure analysis of OLED device by means of in situ LEIPS and UPS combined with GCIB

Low-energy inverse photoelectron spectroscopy (LEIPS) and ultraviolet photoelectron spectroscopy (UPS) incorporated into the multitechnique XPS system were used to probe the ionization potential and the electron affinity of organic materials, respectively. By utilizing gas cluster ion beam (GCIB), in situ analyses and depth profiling of LEIPS and UPS were also demonstrated. The band structures of the 10-nm-thick buckminsterfullerene (C₆₀) thin film on Au (100 nm)/indium tin oxide (100 nm)/glass substrate were successfully evaluated in depth direction.     

Nonexistence of abelian difference sets: Lander's conjecture for prime power orders

In 1963 Ryser conjectured that there are no circulant Hadamard matrices of order 4$"> and no cyclic difference sets whose order is not coprime to the group order. These conjectures are special cases of Lander's conjecture which asserts that there is no abelian group with a cyclic Sylow -subgroup containing a difference set of order divisible by . We verify Lander's conjecture for all difference sets whose order is a power of a prime greater than 3.

     

Enhanced optical absorption in nanopatterned silicon thin films with a nano-cone-hole structure for photovoltaic applications

In this paper, the optical properties of the silicon nano-cone-hole (NCH) structure array are studied. The ultimate efficiency of the optimized NCH array is enhanced by 23.11% compared to an optimized nanohole array of the same thickness. The absorptance enhancement of the NCH arrays is attributed to its lowered reflectance, more supported resonant modes, and enhanced mode interaction. The angular dependence of ultimate efficiency is also investigated.     

Finite quotients of the automorphism group of a free group

The automorphism group AutF_n of a free group F_n of rank n acts on the product of n copies of a group G by substituting n elements of G into the words defining an automorphism of the free group. This gives rise to an antihomomorphism from AutF_nto a permutation group. We determine this antihomomorphic image of AutF_n when G is the semidirect product Z_p x Z_q     

Finiteness of circulant weighing matrices of fixed weight

Let n be a fixed positive integer. Every circulant weighing matrix of weight n arises from what we call an irreducible orthogonal family of weight n. We show that the number of irreducible orthogonal families of weight n is finite and thus obtain a finite algorithm for classifying all circulant weighing matrices of weight n. We also show that, for every odd prime power q, there are at most finitely many proper circulant weighing matrices of weight q.     

Soulamarin, a new coumarin from stem bark of Calophyllum soulattri

The extracts of the stem bark of Calophyllum soulattri gave a new pyranocoumarin, soulamarin (1), together with five other xanthones caloxanthone B (2), caloxanthone C (3), macluraxanthone (4), trapezifolixanthone (5) and brasixanthone B (6) one common triterpene, friedelin (7), and the steroidal triterpene stigmasterol (8). The structures of these compounds were established based on spectral evidence (1D and 2D NMR).
