The use of quantitative carbon nuclear magnetic resonance spectroscopy ((13)C NMR) for the determination of resin loadings has been investigated. Magic angle spinning (MAS) NMR spectra have been obtained for solvent-swollen resins on a conventional 7 mm CP/MAS probe using the two pulse phase modulation (TPPM) proton decoupling sequence. Loadings of resin-bound organic compounds were evaluated via addition of tetrakis(trimethylsilyl)silane as reference or using the carbon resonances of the polymeric resin material as an internal standard. Results for several functionalized Wang and trityl resins are consistent with those obtained using well-established analytical methods. The (13)C NMR method has interesting applications in the field of solid-phase organic synthesis (SPOS), since no functional group acting as a support for the attachment of a quantifiable chromophore must be available in the material of interest.  相似文献

Combinatorial diversity in hypervariable β‐hairpin loops is exploited by the immune system to select binding sites on antibodies for a wide variety of different protein antigens. In a first step towards mimicking this strategy in vitro, for the selection of novel protein ligands, an approach is described here for the parallel synthesis of small libraries of conformationally defined β‐hairpin protein epitope mimetics. Starting from a protruding hairpin loop in platelet‐derived growth factor B (PDGF‐B), 8 and 12 residues were first transplanted from the protein to a D ‐Pro‐L ‐Pro template, to afford the cyclic peptide‐loop mimetics 1 and 2 , respectively. NMR and MD studies in aqueous solution show that both mimetics populate conformations which closely mimic the β‐hairpin in the crystal structure of the native protein (Fig. 5). Based on 1 as a scaffold, a library of 24 mimetics was synthesized in which the four residues at the tip of the loop (VRKK) were held constant, and flanking residues at positions 1, 2, 7, and 8 in the hairpin were varied (Fig. 7). The library was prepared by parallel synthesis in a two‐stage solid‐phase assembly/solution‐phase cyclization process. The products were analyzed by MS, NMR, and CD. 2D‐NOESY revealed for most library members characteristic long‐range NOEs that show that the hairpin conformation is stably maintained. The results suggest that this approach may be useful for the synthesis of much larger libraries of peptide and protein mimetics based on a β‐hairpin scaffold.  相似文献

A novel electrochemical sensor on ZIF-8 nanocomposites (Ag/ZnO/ZIF-8) was developed to analyze the mercury ions (Hg2+). The ZIF-8 materials are one of the 3-dimensional porous metal-organic frameworks with highly accessible pores and great surface area. The ZIF-8 nanocomposites were prepared through simple sol-gel methods and their physio-chemical properties were characterized via different analytical analyses. As a result of cyclic voltammetry, Ag/ZnO/ZIF-8 exhibited a better electrocatalytic behavior towards the detection of mercury ions (Hg2+). Furthermore, the composite modified electrode was then inspected as a sensor for DPV detection of mercury ions. The nanocomposite sensor performed a wide linear range from 0.5 μM to 140 μM with a low detection limit of 40 nM, and high sensitivity of 56.06 μA μM−1 cm−2. Moreover, the ZIF-8 composite sensor showed a higher selectivity toward the detection of mercury ions (Hg2+). The real-time applications of the ZIF-8 composites sensor were inspected in various samples with good sensitivity.  相似文献

In the crystal structure of the title compound, C10H11N3O2, the pyrrolidine ring adopts a puckered envelope conformation. The supramolecular architecture is dictated by the cooperative H-bonded network of centrosymmetric dimers of N–H···N and C–H···O interactions. Two edge to face C–H···π interactions involving the centroid of the pyrrolidine ring contribute to the supramolecular aggregation.  相似文献

The present work investigates the improvement in mechanical properties observed for commercially available diglycidyl ethers of bisphenol-A (DGEBA) with the incorporation of a new type of skeletal modified tetra glycidyl epoxy resin TGBAPB as modifier. Varying weight percentages of TGBAPB have been blended with DGEBA and cured with diaminodiphenylmethane (DDM). The chemical structure of TGBAPB was confirmed by FTIR, NMR, and molecular weight determination was carried out by ESI-MS spectroscopic techniques. The thermal properties were characterized by differential scanning calorimetry (DSC), thermogravimetric analysis (TGA) and mechanical properties like tensile strength, flexural strength, impact strength were also studied by universal testing machine (UTM). Scanning electron microscopy (SEM) investigates the morphological behavior of the neat and blend epoxy resins. The results from different studies indicate that the blend epoxy resin system “B” comprising 75% DGEBA/25% TGBAPB has shown improvements in both toughness and stiffness, despite the fact that it is often found that the enhancement of these two properties together in a material cannot be simultaneously achieved. These aspects of this work are novel.  相似文献
