Porous zirconia particles are very robust material and have received considerable attention as a stationary phase support for HPLC. We prepared cellulose dimethylphenylcarbamate-bonded carbon-clad zirconia (CDMPCCZ) as a chiral stationary phase (CSP) for separation of enantiomers of a set of 14 racemic compounds in normal phase (NP) and reversed-phase (RP) liquid chromatography. Retention and enantioselectivity on CDMPCCZ were compared to those on CDMPC-coated zirconia (CDMPCZ) to see how the change in immobilization method of the chiral selector affects the retention and chiral selectivity. In NPLC, retention was longer and the number of resolved racemates was smaller on CDMPCCZ than on CDMPCZ. However, chiral selectivity factors for some resolved racemates were better on CDMPCCZ than on CDMPCZ. The longer retention on CDMPCCZ is likely due to strong, non-chiral discriminating interactions with the carbon layer on CDMPCZ. In RPLC only two racemates were resolved on CDMPCCZ, but retention times were shorter than, and resolutions were comparable to, those in NPLC, indicating a potential for improving chromatographic performance of the CDMPCCZ column in RPLC with optimized column preparation and separation conditions.  相似文献

The formation of mixed ligand complexes in Ti(IV)-xylenol orange (XO)-H2O2 and Ti(IV)-chromazurol S (CAS)-H2O2 systems was studied by spectrophotometry. The former system gave constant absorbance (λmax = 562 nm) under the condition of [XO]/[Ti(IV)] = 1 in the pH 2–4 region. In the latter system, a distinct maximum at 557 nm was observed when [CAS]/[Ti(IV)] = 4 in the pH range of 4.5–5.2. In both cases, the absorbance at λmax was stable for a long time and proportional to the concentration of hydrogen peroxide. From those facts, the usefulness of the mixtures of Ti(IV)-XO and Ti(IV)-CAS as the colorimetric reagents for the determination of hydrogen peroxide can be expected. The conditions for the use of the Ti(IV)-XO and the Ti(IV)-CAS reagents were examined in detail, and both reagents were found to be available for trace analysis of hydrogen peroxide with high sensitivity.  相似文献

3-Alkylamido-3-deoxy-betulinic acids were synthesized and evaluated for anti-HIV activity as part of the structure-activity relationship study of the potent anti-HIV agent 3-O-(3',3'-dimethyl)-succinyl-betulinic acid (DSB) (2). 3Alpha-diglycorylamide-3-deoxy-betulinic acid demonstrated relatively potent anti-HIV activity (EC50 0.24 microm, TI 728). However, replacing the ester group at C-3 in 2 and its analogues with an amido group yielded inactive or much less potent compounds against HIV replication, indicating that the ester group at C-3 in 2-4 is essential for potent anti-HIV activity.  相似文献

The structural analysis of amylose tris(3,5-dimethylphenylcarbamate) (ADMPC) was performed by NMR spectroscopy using a sample with a lower degree of polymerization in order to understand the chiral recognition mechanism when it was used as a chiral stationary phase (CSP) in high-performance liquid chromatography (HPLC). ADMPC exhibited chiral discrimination for many enantiomers, including 1-(9-anthryl)-2,2,2-trifluoroethanol (1) and 1,1'-bi-2-naphthol (2) in both NMR and HPLC. A good agreement was observed between the HPLC and NMR results when chloroform was employed as the common solvent. The structure of ADMPC in solution was investigated by NMR using the 2D NOESY technique coupled with computer modeling, and a left-handed 4/3 helical structure was obtained as the most probable one. The binding geometry between ADMPC and the enantiomers of 1 was also investigated by (1)H NMR titration. On the basis of these results combined with molecular modeling, a rational model to explain the chiral discrimination mechanism of 1 on ADMPC was proposed.  相似文献

The 3,5-dimethylphenylcarbamates of cellulose and amylose bearing a small amount of 3-(triethoxysilyl)propyl residues were synthesized by a simple process and efficiently immobilized onto a silica gel support by intermolecular polycondensation of the triethoxysilyl groups. The obtained chiral packing materials (CPMs) were evaluated by high-performance liquid chromatography. The polysaccharide derivatives containing about 1-2% of the 3-(triethoxysilyl)propyl residue were efficiently immobilized with a high chiral recognition. The immobilized CPMs could be used with the eluents containing chloroform and tetrahydrofuran, which cannot be used with the conventional coated-type CPMs. By using these eluents, the chiral recognitions for many racemates could be improved.  相似文献

Technological advances have facilitated the generation of artificial proteins that possess the capabilities of recognizing and binding to inorganic solids and/or controlling nucleation processes that form inorganic solids. However, very little is known regarding the structure of these interesting polypeptides and how their structure contributes to functionality. To address this deficiency, we report structural investigations of an artificial protein, p288, that self-assembles and controls the nucleation of simple salts and organic compounds into dendrite-like crystals. Under aqueous conditions at low pH and in the presence of high salt, p288 is conformationally labile and exists primarily as a random coil conformer in equilibrium with other undefined secondary structures, including polyproline type II and beta turn. We note that p288 can fold into either a partial beta strand (at neutral pH) or a predominantly alpha helical (in the presence of TFE) conformation. Solid-state 13C-15N NMR experiments also reveal that p288 in the lyophilized, hydrated state possesses some degree of nonrandom coil structure. These results indicate that p288 is conformationally labile but can undergo conformational transitions to a more stable structure when water solvent loss/displacement occurs and protein concentrations increase. We believe that conformational instability and the ability to adopt different structures as a function of different environmental conditions represent important molecular features that impact p288 supramolecular assembly and crystal nucleation processes.  相似文献

It is commonly thought that UV or visible-light luminescence imaging of biological subjects during X-ray irradiation at the energy below 120 keV is impossible because the secondary electrons produced in this energy range do not emit Cerenkov light. Contrary to this consensus, we found UV or visible-light luminescence imaging of the subjects were possible with X-ray irradiations of this energy range. We placed one of the biological subjects in a black box; visible-light luminescence images were measured with a high-sensitivity, cooled charge coupled device (CCD) camera during X-ray irradiation at energy below 120 keV. We also conducted the imaging of air without subjects during irradiation of the same X-ray. The biological subjects emitted visible-light luminescence, and the imaging was possible with the irradiation of the X-ray below 120 keV. The luminescence images were observed in only the X-ray irradiated areas. Also air luminescence images could be obtained and the intensity of the luminescence measured from the images was proportionally increased with the exposure dose. UV or visible-light luminescence imaging of biological subjects was possible during X-ray irradiations lower energy than the Cerenkov-light threshold. The phenomenon was different from general X-ray fluorescence because wavelength of the luminescence is UV or visible-light. The luminescence imaging method is promising for estimating the irradiated area with X-ray, which could be used for interventional radiology (IVR). Also air luminescence imaging would be applied to the exposure dose distribution measurements for X-ray of diagnostic X-ray systems.  相似文献

Cellulose 3,5-dimethylphenylcarbamates bearing a low content of a vinyl group at the 6-position on the glucose units were synthesized by a previously developed regioselective method and chemically immobilized onto a vinylized silica gel as chiral stationary phases (CSPs) for high-performance liquid chromatography (HPLC). The immobilization of the derivatives was performed through a radical polymerization reaction with AIBN as the initiator in the presence of toluene. The effects of vinyl monomers, such as isoprene, 2,3-dimethyl-1,3-butadiene (DMBD), ethylene glycol dimethacrylate (EDMA) and 1,5-hexadiene, on the immobilization and enantioselectivities of the derivatives were investigated. The effect of the temperature used for the radical polymerization reaction on the immobilization was also examined. In addition, the direct comparison of the chiral recognition abilities of the laboratory-made and commercially available columns was discussed.  相似文献
