Ekeland’s principle for vector equilibrium problems

In this paper, the authors deal with bifunctions defined on complete metric spaces and with values in locally convex spaces ordered by closed convex cones. The aim is to provide a vector version of Ekeland’s theorem related to equilibrium problems. To prove this principle, a weak notion of continuity of a vector-valued function is considered, and some of its properties are presented. Via the vector Ekeland’s principle, existence results for vector equilibria are proved in both compact and noncompact domains.     

Normal human dermal fibroblasts: proteomic analysis of cell layer and culture medium.

Proteins present within the cell layer and those released in the cell medium from in vitro cultured normal human dermal fibroblasts were separated and characterized in terms of their isoelectric point and molecular weight, by two-dimensional (2-D) gel electrophoresis. All spots in the synthetic gel were firstly analyzed by the Melanie 3 software and compared with those of breast cancer cells, colorectal epithelial cells, HL60, lymphoma cells, and platelets, already available on-line. From the identification of 144 spots from both the cell layer and the medium, we were able to recognize 89 different proteins, since a certain number of spots represented different isoforms of the same molecule. Identifications were performed by matching with on-line 2-D databases, and by matrix assisted laser-desorption/ionization-time of flight-mass spectrometry (MALDI-TOF-MS), in order to confirm the identification by matching, or to identify new proteins. The procedure we used allows (i) to design a highly reproducible reference map of the proteome of adult human normal fibroblasts in culture, (ii) to evaluate protein species produced in the cell layer as well as those released in the culture medium, and (iii) to compare data from gel matching with those obtained by MS. This work represents an essential step for a better knowledge of mesenchymal cells, given the widespread use of this cell type in both clinical and experimental investigations.     

Focusing-defocusing phenomena of radio echo signal after passage of the morning solar terminator

Summary Campaign results of high-repetition ionospheric soundings, performed at the ionospheric station of Rome using a Digisonde 128P, are presented. We introduce an index derived from run rapid ionograms that can be used to detect oscillations of isodensity surfaces ofF ionospheric region started by morning solar terminator.     

The behaviour of alkylcobalt carbonyls under “oxo” conditions

Both normal and secondary alkylcobalt carbonyls are shown not to undergo isomerization under catalytic “oxo” conditions, and the relevance of this obervation for the mechanism of the “oxo” reaction is discussed.     

Structure factors calculated from molecular wavefunctions. II. Analysis of the molecular charge distribution in diborane

The electron density distribution in the crystal of diborane has been determined using structure factors based on molecular densities. The libration-correction and third-cumulant terms have also been included in the expression for the temperature factor. The equilibrium r_e values for BH bond lengths obtained by this treatment are about 0.02Åshorter than the spectroscopic ones. A total librational motion of the diborane molecule is determined: the root-mean-square oscillations about inertial axis are 11.0° (8), 9.6° (5) and 7.2° (5), respectively. The dynamic theoretical deformation density shows a three-center BHB bond picture for the bridge structure.     

Asymmetric synthesis by chiral ruthenium complexes : VI. Enantiomer-discrimination in the hydrogen transfer from racemic alcohols to prochiral ketones in the presence of H4Ru4(CO)8[(−)-diop]2

Hydrogen transfer from racemic alcohols to prochiral ketones in the presence of H₄Ru₄(CO)₈[(?)-DIOP]₂ has been examined. The enantiomer-discrimination is influenced by the structure of the reactants, temperature and the excess of phosphine present.     

Structure-based optimization of a non-beta-lactam lead results in inhibitors that do not up-regulate beta-lactamase expression in cell culture

Bacterial expression of beta-lactamases is the most widespread resistance mechanism to beta-lactam antibiotics, such as penicillins and cephalosporins. There is a pressing need for novel, non-beta-lactam inhibitors of these enzymes. One previously discovered novel inhibitor of the beta-lactamase AmpC, compound 1, has several favorable properties: it is chemically dissimilar to beta-lactams and is a noncovalent, competitive inhibitor of the enzyme. However, at 26 microM its activity is modest. Using the X-ray structure of the AmpC/1 complex as a template, 14 analogues were designed and synthesized. The most active of these, compound 10, had a K(i) of 1 microM, 26-fold better than the lead. To understand the origins of this improved activity, the structures of AmpC in complex with compound 10 and an analogue, compound 11, were determined by X-ray crystallography to 1.97 and 1.96 A, respectively. Compound 10 was active in cell culture, reversing resistance to the third generation cephalosporin ceftazidime in bacterial pathogens expressing AmpC. In contrast to beta-lactam-based inhibitors clavulanate and cefoxitin, compound 10 did not up-regulate beta-lactamase expression in cell culture but simply inhibited the enzyme expressed by the resistant bacteria. Its escape from this resistance mechanism derives from its dissimilarity to beta-lactam antibiotics.     

New developments in the synthesis of pyrrolizidinone-based dipeptide isosteres

1,3-Dipolar cycloaddition of acrylamide with the cyclic nitrone derived from proline tert-butyl ester has been employed in the synthesis of bicyclic Gly-(s-cis)Pro isosteres suitably protected for the Fmoc-based solid phase peptide synthesis. (R)-1-Phenylethylamine was introduced as chiral auxiliary to resolve racemic intermediates and obtain enantiopure compounds. Using methacrylamide as dipolarophile, the analogous Ala-Pro mimetics have been prepared in racemic form, whereas the same strategy applied to methyl itaconate failed to give the corresponding Asp-Pro mimetic.
