This paper describes khellin hydrolysis mechanism using semiemperical PM3 implemented in Gaussian 03 package. The calculations show that in the presence of an acidic media, an enolate molecular ion leads directly to ω-acetokhellinone while in the basic media it leads to khellinone. 相似文献
A liquid membrane electrode prepared with moclobemide-dipicrylamine ion-pair complex, dissolved in nitrobenzene as solvent, was studied for analytical performance. The linear response covers the range 10(-3)-10(-6) M moclobemide solution, with a slope of 50.7 mV decade(-1) (pH range 3.5-8). The detection limit is 3 x 10(-7) M. The electrode shows stability, good reproducibility and fast response. The selectivity of the electrode is good. There are two important interfering ions: mianserin and thiamine (Vitamin B(1)). The compression excipients (such as Mg(2+), starch, talcum powder) do not interfere. These characteristics of the electrode enabled it to be used for the determination of moclobemide in drugs and as an active substance, via indirect and direct potentiometric methods. Via an indirect potentiometric method moclobemide, as an active substance, can be determined with an average recovery of 99.96% and a relative standard deviation of 0.85%, and this method can also be used for its determination in drugs with a relative standard deviation of < 2%. The electrode is useful for the determination of the dissolution rate of moclobemide tablets. The physical processes are numerically simulated by typical equations. The apparent first-order rate constants for disintegration and dissolution were calculated. 相似文献
Reliable, traceable and comparable measurements provide the rational basis for evaluation of the quality of a result and
the starting point for recognized laboratory accreditation in any national area. Modern medical diagnostics and treatment
involve rapidly rising numbers and types of clinical laboratory measurements, that are reliable. Therefore, the basic principles
to be followed to assure the traceability of clinical measurements as required by the Romanian Laws of Metrology are reviewed.
Main sources affecting the quality of the unbroken chain of calibrations that relate the measurements back to appropriate
measurement standards are discussed. Examples of how to achieve traceable measurements in clinical laboratories are presented.
Details of specific uses of reference materials, measuring instruments and standard measurement methods are also discussed.

Enantioselective, potentiometric membrane electrodes (EPMEs) based on carbon paste impregnated with different maltodextrins {dextrose equivalent (DE) 4.0-7.0 (I), 13.0-17.0 (II) and 16.5-19.5 (III)} as chiral selectors for the assay of S-perindopril is described. The proposed electrodes could be reliably employed in the assay of S-perindopril raw material and from its pharmaceutical formulation, Coversyl® tablets. The electrode based on maltodextrin (I) showed the best enantioselectivity and time-stability. The surfaces of the electrodes are easily renewable by simply polishing on an alumina paper. 相似文献
The interaction of rutin and venoruton (troxerutin), with alpha-, beta- and gamma-cyclodextrin (CD), hydroxypropyl-beta-cyclodextrin (HP-beta-CD) and methyl-beta-cyclodextrin (M-beta-CD) was investigated by reversed-phase thin layer chromatography on polyamide plates. A mobile phase consisted of NH(4)OH; NH(4)Cl buffer solution containing various CD concentrations (pH = 9.7, 20 degrees C) was used as mobile phase. The equilibrium constants (K(f)) and the retention factor (R(f)) were determined and used to study the inclusion process. The in fluence of CDs on the solubility of rutin and venoruton was characterized by R(M) values and the increasing hydrophilicity of drugs. The results show that the inclusion capacity of cyclodextrins follows the order HP-beta-CD > M-beta-CD > beta-CD > gamma-CD, and rutin is more easily included by the studied cyclodextrins than venoruton. In addition, the thermodynamic parameters (Delta H, Delta S) for the formation of complexes were obtained from the van't Hoff equation, displaying the enthalpy-entropy compensation effect. 相似文献
The present study investigates the antioxidant activities of some Romanian plants, using different spectrophotometric methods (FRAP I, FRAP II, and CUPRAC). The plants investigated are hawthorn (Crataegus oxyacantha), bilberry (Vaccinium myrtillus L.), rosehip (Rosa canina), and chokeberries (Aronia melanocarpa). Hawthorn is used to treat a wide variety of inflammatory conditions, but the primary use is generally restricted for treating hypertension, ischemic heart disease, congestive heart failure, and arrhythmia. Investigations have proved the safe and reliable use of plant and plant extracts for treatment of cardiovascular disorders. 相似文献
Following administration of the acidic drug tolmetin (TOL) anaphylactic reactions occurred, which have been hypothesized to be related to the formation of reactive acyl glucuronides. Recently, glutathione adducts have been detected upon incubation of TOL with human liver microsomal preparations, which proved that oxidative activation might also be a pathway of formation of reactive—possibly toxic—glutathione metabolites of TOL. The aim of this work was to develop a new and robust HPLC method to investigate the in vivo effect of 2 coadministered drugs/nutritional supplements on the kinetics of TOL in rats (cimetidine; CIM) known to be a potent inhibitor of CYP3A4, an enzyme that catalyzes the oxidative metabolism and Quercetin; and QUE which induces UGT1A6, an enzyme involved in glucuronidation of acidic drugs. DryLab®, a computer simulation software package, was used to assist in the development and optimization of the HPLC method used for separation of TOL and the two potential kinetic modulators together with three potential internal standards (zomepirac, carvedilol and fexofenadine). The method was validated in biological samples obtained from rats. Non-compartmental pharmacokinetic analysis of data obtained from plasma and rat liver tissue showed significantly higher concentrations of TOL in the presence of CIM; and significantly longer elimination half-life lives in presence of QUE, which implies that drugs or food components interacting with CYP3A4 cause alteration in the metabolic oxidative biotransformation of TOL in vivo leading to accumulation of TOL in the body through a decrease of its clearance. These findings might account for to the side-effects associated with TOL when co-administered with such kinetic modulators.
A simple solid phase extraction (SPE) method coupled with high performance liquid chromatography (HPLC) using UV detector and microemulsion electrokinetic chromatography (MEEKC) has been developed and compared for the quantitative determination of miconazole nitrate in pharmaceutical formulation. For HPLC method, two parameters were optimized, namely, the wavelength and the mobile phases. The optimized condition was at the 225 nm wavelength and the mobile phase of ACN:MeOH (90:10 v/v). There are seven MEEKC parameters that were optimized, in this research, which were applied to voltage, temperature, wavelength, sodium dodecyl sulfate (SDS) concentration, buffer pH, buffer concentration and butan-1-ol concentration. The optimum MEEKC condition was obtained using 86.35 % (w/w) 2.5 mM borate buffer pH 9, 0.25 % (w/w) SDS, 0.8 % (w/w) ethyl acetate, 6.6 % w/w butan-1-ol and 6.0 % (w/w) acetonitrile. The combination of SPE using a diol column with HPLC–UV and the MEEKC methods were successfully applied for the determination of miconazole nitrate in a pharmaceutical formulation with the recovery percentage of 98.35 and 92.50 %, respectively. 相似文献
A new amperometric biosensor based on D-aminoacid oxidase is described for the assay of R-perindopril. R-perindopril can be determined in the 400-20 nmol/L concentration range; the detection limit is 10 nmol/L. The selectivity was checked with S-perindopril, D- and L-proline, and polyvinylpyrrolidone. The main interfering species was D-proline. An automated system for the assay of R-perindopril based on the concept of flow injection with an amperometric biosensor (based on D-aminoacid oxidase) as detector is also described. The system is suitable for the on-line monitoring of R-perindopril at a sampling rate of 72 samples/h, in the linear range: 100 nmol/L -20 nmol/L with an RSD better than 0.09% (n = 10). 相似文献
