The microcalorimetric method was used to study the antibacterial activity of two newly synthesized Schiff basecompounds(H_2L~(3')and H_2L~3)on Escherichia coli,trying to obtain the action on both of multiplying bacteria andnon-multiplying bacteria at one experiment.The metabolic power-time curves of the bacteria treated with the com-pounds were obtained,and the thermokinetic parameters were analyzed,from which the antibacterial activities ofthese compounds were evaluated.The results showed that both of the two compounds have good activity on aerobicmultiplying metabolism of E.coli,with the value of IC_(50)75.8 and 168.8 mg/L respectively,but have not effectiveaction on fermentation metabolism of E.coli.The action of the compounds on the non-multiplying metabolism wasinvestigated by taking the heat output of E.coli in the stationary phase as the guideline of the activity.The value ofMSC_(50)(minimum stationary-cidal concentration 50)of them is 118 and 187.5 mg/L,respectively.So,H_2L~(3')hasstronger antibacterial action on E.coli than H_2L~3 either for multiplying bacteria or non-multiplying bacteria,andtheir activity on the aerobic multiplying bacteria of E.coli is mainly shown.It does strongly suggest that the calo-rimetric method should play an important role in the fight against the drug-resistant bacteria.  相似文献

以曙红Y(Eosin Y)为探针,采用竞争结合法考查了非甾体抗炎药吲哚美辛和舒林酸及其相关的金属配合物与BSA的结合。在中性环境中,曙红Y的吸收光谱随着BSA的加入发生明显变化,结果表明,曙红Y与BSA之间的结合以静电作用为主,两者之间可能发生电子转移。曙红Y与BSA结合后,抗炎药吲哚美辛,舒林酸的加入可以破坏这种结合,曙红Y部分游离出来,其吸收值恢复,说明药物可以同样的方式结合到BSA上,抢占曙红Y在BSA上的结合位点。将药物转化为金属配合物后,与血清蛋白的亲和程度更强,铜(Ⅱ)的配合物最为突出,推测将非甾体抗炎药转化为铜配合物后,有更好的药理活性。  相似文献

采用稀释法与胺5倍过量合成了一种新型的含吡啶环的开链二胺1a(N,N′-双(2-氨基乙基)-2,6-吡啶二甲酰胺)。此外,合成了六个新型多齿希夫碱配体N,N′-双(β-R-苯甲醛亚胺基乙基)-2,6-吡啶二甲酰胺[其中,R=H (2a),o-OH (2b),p-OH (2c),m-NO2 (2d),p- N(CH3)2 (2e)]及N,N′-双[γ-水杨醛亚胺基正丙基]-2,6-吡啶二甲酰胺2f。通过元素分析,紫外-可见光谱,红外光谱,氢核磁共振谱及质谱对化合物进行了表征。通过化合物2e的单晶结构X-射线单晶衍射分析表明该晶体属于立方晶系P-1空间群,其晶胞参数为:a=11.010(2) nm,b=13.865(3) nm,c=9.6537(19) nm,α=102.77(2)º,β=92.07(3)º,γ=87.98(3)º,V=1435.7(5) nm3,Z=2,Dc=1.230 mg•cm-3,Mr=531.66。微量热法检测了化合物对大肠杆菌的抑制作用,并初步分析了化合物结构与抗菌活性之间的关系。实验结果表明,所有化合物都对大肠杆菌有抑制作用,其中水杨醛希夫碱的抑菌活性最好。  相似文献

Metabolic thermogenic curves of mitochondria isolated from liver tissue of Carassius auratus and the effect of different concentration of NaAsO2 on it were investigated by TAM air isothermal microcalorimeter, ampoule method, at 28.00 ℃. From the thermogenic curves, activity recovery rate constants k, the maximum heat production rate (Pmax) and the total heat produced (Q) were obtained. The values of k and Pmax decline gradually with the increase of the concentration of NaAsO2, and both of the values of Pmax and k are highly correlated to the concentration of NaAsO2. When concentration of NaAsO2 reached 16.0 μg/mL, the maximum heat production rate dropped to 70.8% of the control group, and the corresponding percentage of k was 71.4% of the control group. This experimental result indicates that the addition of NaAsO2 has restrained the metabolic activities of mitochondria in vitro.  相似文献

用微量热法研究一种新型希夫碱及其3d,4f配合物(2L, 2LZnYb)对大肠杆菌和金黄色葡萄球菌的抗菌活性, 得到了在它们作用下大肠杆菌和金黄色葡萄球菌生长代谢的产热曲线, 并且基于分析生长代谢和非生长代谢的产热曲线建立的热动力学方程, 获得了它们的抗菌活性. 结果表明, 两种化合物(ZL, 2LZnYb)对大肠杆菌的生长代谢有强的活性(IC50分别为6.1 和5.1 mg·L-1), 但对金黄色葡萄球菌的生长代谢的活性弱得多(IC50分别为310.1 和595.5 mg·L-1). Zn 和Yb的导入使化合物对大肠杆菌生长代谢的抑制作用稍微增加, 但大大降低了对金黄色葡萄球菌的抑制作用. 对于非生长代谢, 两种化合物的活性有很大的差别. 无论对大肠杆菌还是金黄色葡萄球菌, 由于配体2L的导入, 表现出显著的抑制作用, 2L的MSC50为6.4和209.7 mg·L-1. 配体2L可能成为新的抗菌先导化合物.  相似文献

Several new transition metal complexes using Schiff base containing pyridine and amide moieties (N, N′-bis(β-salicylaliminoethyl)-2, 6-pyridinedicarboxylic amide, H4L) as the ligand have been prepared. Their compositions and structures are corroborated by elemental analysis, IR, UV, ^1H NMR, DTA-TG and molar conductivity data.Their anti-bacterial activities have been studied by microcalorimetry. The result shows that the ligand and all complexes are potential anti-bacteria reagent and their inhibitory capacities are concentration-depended. The Mn complex has the strongest inhibitory capacity.  相似文献

用TAM air等温微量热检测系统,测定鲫鱼(Carassius auratus)肝脏线粒体体外代谢热及不同浓度联苯胺(1.0,5.0,10.0,20.0,40.0mg/L)对其代谢活性的影响.结果表明。线粒体体外代谢活性恢复期速率常数k1,活性衰竭期速率常数k2以及最大放热功率Pmax均受到联苯胺的影响,且随着联苯胺浓度的升高,线粒体代谢总产热量Q依次下降。  相似文献
