微柱高效液相色谱法测定大鼠脑微透析液中 的乙酰胆碱和胆碱

用所建立的微柱高效液相色谱分离、柱后固定化酶反应器酶解、电化学检测器检测的方法, 对用不同浓度异氟醚麻醉前后的大鼠脑微透析液中的乙酰胆碱和胆碱的浓度进行了测定。在 乙酰胆碱和胆碱的浓度为10～ 2 000 nmol/L 时,其浓度分别与各自相应峰高的线性 关系良好,两者的检测限(以信噪比为3计)均可达5 nmol/L 。微透析液中乙酰胆碱和胆碱的含量在大鼠清醒时最高,随着异氟醚吸入浓度的增加,乙酰胆碱和胆碱含量明显降低。在大鼠海马和脑皮层中乙酰胆碱浓度的降低与大鼠吸入异氟醚的浓度相关( P ＜     

STUDIES ON THE MECHANISM OF INDIRUBIN ACTION IN THE TREATMENT OF CHRONIC GRANULOCYTIC LEUKEMIA——V.BINDING BETWEEN INDIRUBIN AND DNA AND IDENTIFICATION OF THE TYPE OF BINDING

The binding between indirubin and calf thymus DNA in vitro has been verified by meansof the isotope labelling method, spectrophotometric method and thermal denaturation meas-urements. The λ_max 207 nm of indirubin shifted toward longer wave length with decrease ofabsorbance after the incubation of indirubin with DNA. The escalation of Tm value of DNAinduced by indirubin was about 2.4°C and it was reproducible. The binding force between themwas rather weak, as indirubin molecules were easily released during the precipitation withalcohol or the gel filtration. The binding was not affected by sodium chloride even at high con-centration but greatly decreased (to 20-30% of the control) in the presence of 8 M urea.These results showed that the binding between indirubin and DNA might be of hydrogen bondrather than ionic. The amount of bound ~3H-indirubin was directly proportional to the con-centration of indirubin. However, it increased abruptly when the concentration of indirubinreached 1.5×10~(-4) M. This     

靛玉红治疗慢性粒细胞白血病疗效原理的研究——靛玉红与DNA的结合作用及其结合键型的鉴别

本文应用吸收光谱分析法、同位素结合法、DNA融解曲线法及其它一些方法,证明了靛玉红与DNA之间具有结合作用。二者结合的最大分子比例(以靛玉红:脱氧核苷一磷酸计)为46:1000,结合反应的平衡常数K=5.7×10~6;结合作用发生迅速,无碱基特异性;热变性DNA能结合更多的药物分子;靛玉红与DNA结合的主要键型为氢键,但当靛玉红浓度大于一定限度时,还出现另一种形式的结合。     

中国人Bcll/St14 RFLPs研究与甲型血友病基因诊断

本文报道了St14(DXS52)上的一组BclI限制酶片段长度多态性(RFLPs)。该RFLPs由4.0kb,3.3kb,3.0kb和2.3kb DNA片段组成。在中国人中这组多态性片段出现的频率分别为0.09,0.12,0.44和0.35,提供的多态性信息量为0.66。甲型血友病家系基因连锁分析结果表明,这组RFLPs的DNA杂交图谱较目前国际上广泛使用的TaqI/St14 RFLPs更易于分析,在甲型血友病基因连锁分析中具有较高的应用价值。     

STUDY ON BclI/St14 RFLPs IN CHINESE FOR DNA DIAGNOSIS FOR HEMOPHILIA A

The BclI polymorphism within DXS52 (St14) was reported. It was composed of 4 allelic fragments 4.0 kb, 3.3 kb, 3.0 kb and 2.3 kb, The frequency of these fragments were 0.09, 0.12, 0.44 and 0.35 respectively in the Chinese. The polymorphism provided the PIC of 0.66. DNA analysis of families, with hemophilia A showed that the confidence of the RFLPs was the same as the TaqI/St14 RFLPs and for carrier detection the former is much better than that of the TaqI/St14 RFLPs.
