Abstract— Recently, a new technique of 1O2 generation has been described by Midden and Wang. The sensitizer is immobilized on silica gel particles which are attached on one surface of a glass plate which is placed, sensitizer-side down, above a solution leaving a small air space between the sensitizer and the solution. Applying the principle of the technique, i.e. generation of 1O2 in the gas phase above the solution, we first improved the long-term stability of the dye (rose bengal) by using double stock transparent tape instead of rubber cement. Under these conditions, although the initial rate of production was higher in rubber cement, the rate of production reached a plateau after 30 min, whereas with double stick transparent tape, the rate of generation was linear up to 100 min. This result was explained by the fact that silica glass beads are embedded in the cement which could lead to the bleaching reaction of the dye. Different reactions have been investigated, the bleaching of N,N-dimethyl-4-nitrosoaniline (RNO) in the presence of imidazole, destruction of anthracenedipropionic acid, formation of a stable nitroxide from a sterically hindered secondary amine and the influence of pH on the nitroxide yield. The yield was measured to be 5 × 1012 molecules of singlet oxygen produced per second per cm2 from the surface of the plate using the RNO bleaching reaction. A thin chip polymer-based microsprayer has been coupled to a hybrid quadrupole time-of-flight mass spectrometer (QTOF MS) and introduced in carbohydrate research. The feasibility of the approach is demonstrated for mapping, sequencing and structural elucidation of glycoconjugates originating from human body fluids and tissues such as a glycopeptide mixture from normal human urine and an isolated and purified GT1 ganglioside fraction from normal adult human brain. The optimization procedure required by each glycoconjugate category is described and the advantages of the system in terms of flexibility and adaptability to QTOF MS, stability of the ESI MS signal, carbohydrate ionization and sequencing, sensitivity, speed of analysis and sample consumption are discussed. 相似文献

Pathogen reduction technologies (PRT) are photochemical processes that use a combination of photosensitizers and UV-light to inactivate pathogens in platelet concentrates (PCs), a blood-derived product used to prevent hemorrhage. However, different studies have questioned the impact of PRT on platelet function and transfusion efficacy, and several proteomic analyses revealed possible oxidative damages to proteins. The present work focused on the oxidative damages produced by the two main PRT on peptides. Model peptides containing residues prone to oxidation (tyrosine, histidine, tryptophane, and cysteine) were irradiated with a combination of amotosalen/UVA (Intercept process) or riboflavin/UVB (Mirasol-like process). Modifications were identified and quantified by liquid chromatography coupled to tandem mass spectrometry. Cysteine-containing peptides formed disulfide bridges (R-SS-R, ?2 Da; favored following amotosalen/UVA), sulfenic and sulfonic acids (R-SOH, +16 Da, R-SO3H, +48 Da, favored following riboflavin/UVB) upon treatment and the other amino acids exhibited different oxidations revealed by mass shifts from +4 to +34 Da involving different mechanisms; no photoadducts were detected. These amino acids were not equally affected by the PRT and the combination riboflavin/UVB generated more oxidation than amotosalen/UVA. This work identifies the different types and sites of peptide oxidations under the photochemical treatments and demonstrates that the two PRT may behave differently. A simple desalting procedure for the coupling of a polymer microchip injector to mass spectrometry is proposed. The overall process is based on the adsorption of proteins on a poly(vinylidene difluoride) (PVDF) membrane, which are then directly eluted in the spraying solution. This microchip-based approach has been successfully applied to small drugs, peptides and proteins originally diluted in phosphate-buffered saline (PBS). Moreover, when eluting the retained proteins in small volumes, a preconcentration is obtained. The combination of single-use, mass-produceable, low-sample-consumption, easy-to-automate, miniaturized polymer injectors with easy-to-handle solution-exchange membranes makes this system particularly amenable to screening applications. 相似文献
