With the combination of the the stoichiometric displacement model for retention (SDM-R) in reversed phase liquid chromatography (RPLC) and the stoichiometric displacement model for adsorption (SDM-A) in physical chemistry,the total number of moles of the re-solvated methanol of stationary phase side.nr,and that of solute side in the mobile phase,q,corresponding the one mole of the desorbing solute,were separately determined and referred as the characterization parameters of the contributions of the adsorption mechanism and partition mechanism to the solute retention,respectively.A chromatographic system of insulin,using mobile phase consisting of the pseudo-homologue of alcohols(methanol,ethanol and 2-propanol)-water and trifluoroacetic acid was employed.The maximum number of the methanol layers on the stationary phase surface was found to be 10.6,only 3 of which being valid in usual RPLC,traditionally referred as a volume process in partition mechanism.However,it still follows the SDM-R.Both of q and nr of insulin were found not to be zero,indicating that the retention mechanism of insulin is a mixed mode of partition mechanism and adsorption mechanism.When methanol is used as the organic modifier,the ratio of q/nr was 1.13,indicating the contribution to insulin retention due to partition mechanism being a bit greater than that due to adsorption mechanism.A linear relationship between q,or nr and the carbon number of the pseudo-homologue in the mobile phase was also found.As a methodology for investigating the retention mechanism retention and behavior of biopolymers.a homologue of organic solvents as the organic modifier in mobile phase has also been explored. 相似文献

Abnormal Fluorescence Behavior of Pyrene Functionalized Chitosan Films to Some Quenchers 总被引:4,自引:0,他引:4
Investigation of the fluorescence behavior of a pyrene functionalized chitosan film (PSC-CS) to some quenchers,including KI,CH3NO2 and Cu(NO3)2,has revealed that the monomer emission from the PSC-CS film increased rather than decreased,whereas the excimer emission decreased with the increase of time of duration after addition of KI or Cu(NO3)2.Both the increasing and decreasing processes could last as long as 20min to 40min,Unlike that observed in the system containing KI or Cu(NO3)2,for CH3NO2 system,both the monomer emission and the excimer emission increased with introduction of the quencher,but the excimer emission only started to increase after an hour-long induction period.The position of the excimer emission of the PSC-CS film did not change very much with addition of KI or Cu(NO3)2.However,addition of CH3NO2 made the excimer emission blue shifted for at least 20nm indicating formation of distorted excimers opr partially overlapped excimers.The fluorescence response of the film to external additions is sensitive but is low in selectivity.This property may make the film use as a novel sensing material for monitoring the purity of water. 相似文献

Spontaneous vesicles from the mixed queous solution of poly-talied cationic surfactant PTA and anionic surfactant AOT are firstly obtained.Vesicle formation and characterizations are demonstrated by negative-staining TEM and dynamic light scattering.A monodisperse vesicle system is obtained with a polydispersity of 0.082.Ultrasonication can promote the vesicle formation,Mechanism of vesicle formation is discussed from the viewpoint of molecular interaction. 相似文献

The contributions of tetrahydrofurfuryl alcohol(THFA) and polyethylene glyco(PEG) to the renature efficiency of α-chymotrypsin were investigated and compared with each other,The maximum increments of bioactivity recovery of α-Chy were found to be 25.1% for THFA,10.4% for PEG,respectively.The experimental results indicated that the denaturant solution containing THFA contributed more to the renaturation of α-Chy in high performance hydrophobic interaction chromatography(HPHIC) than that containing PEG,when the concentration of THFA was 3.2%,the bioactivity recovery of α-Chy is the highest. 相似文献
