[chemical reaction: see text]. Three new ent-trachylobane diterpenoids (1-3) were isolated and structures elucidated from Mitrephora glabra Scheff. (Annonaceae). Mitrephorone A (1) possesses a hexacyclic ring system with adjacent ketone moieties and an oxetane ring, both of which are unprecedented among trachylobanes. All compounds were evaluated for cytotoxicity against a panel of cancer cells, where 1 displayed the most potent and broadest activity, and against a battery of antimicrobial assays, where all compounds were approximately equipotent. 相似文献

The coulometric electrolytic principle is utilized for the measurement of moisture sorbed on sintered thorium dioxide. Moisture is removed from the sample by heating and is swept by dry argon gas through a hygrometer. The resulting hygrometer output is converted into frequency pulses and counted cumulatively. The instrument is calibrated by use of standard hydrated chemicals. The number of counts/mu;g of water obtained from standards compares favourably with the theoretical number of counts/mu;g calculated from performance specifications of the components employed. The error is <5% for 1-350mu;g of water. A high sensitivity and low operating blank recommends application of this method to materials other than thorium dioxide, having very low moisture contents. 相似文献

[reaction: see text] OSW saponins, featuring a 16beta,17alpha-dihydroxycholest-22-one aglycon and an acylated beta-D-xylopyranosyl-(1-->3)-alpha-L-arabinopyranosyl residue attached to the 16-hydroxyl group, have recently been discovered from a group of lily plants, which show potent antitumor activities with a novel mechanism of action. This paper describes an aldol approach to the stereoselective construction of the 16alpha,17alpha-dihydroxycholest-22-one structure from 16alpha-hydroxy-5-androsten-17-ones and propionates. Elaboration of the aldol adducts toward OSW-1, involving installation of the isoamyl ketone side chain, inversion of the 16-hydroxyl configuration, and selective protection of the C22-oxy function, has been explored and accomplished. In particular, the present route was found convenient for the synthesis of OSW saponin analogues with a C22-ester side chain. Thus, the 23-oxa-analogue of OSW-1 (40) was prepared starting from the industrial dehydroisoandrosterone (1) in a linear eight-step sequence and in 26% overall yield. Analogues with a variety of modified side chains were prepared, via aldol condensation with propionates of varying length, thiopropionate, and acetate (for preparation of 68-75) or via aminolysis of the 22,16-lactone 26 (for preparation of the 23-N-analogues). Cross metathesis (CM) reaction was also found feasible for modification at the final stage from C22-allyl ester 70. Valuable structure-activity relationships (SAR), together with the practical synthetic approach, have thus been provided to set a new stage for further studies on this new type of antitumor structures. 相似文献

A method for the identification of double bond locations in polyunsaturated long chain alkenones adapted to nanogram amounts
as currently analyzed by gas chromatography coupled to mass spectrometry (GC-MS) has been developed. The method is based on
interpretation of the electron impact mass spectra of the imino derivatives of the carbonyl groups using either cyclopentyl
or phenyl substitutents. Other complementary derivatization methods such as elaidization and hydrogenation have also been
used for structural characterization of these compounds. This application has led to the identification of a novel homologous
series of di-, tri-, and tetraunsaturated ketones with carbon number chain lengths between 37 and 40 in coastal hypersaline
sediments. The novel series identified shows a distribution in which the double bond position between different homologs is
established by reference to the distance from the carbonyl group whereas the previously known alkenones were constituted by
unsaturated homologs with double bonds located at defined distances of the terminal methyl. This difference points out to
a dissimilar, but still unknown, biogenic precursor of these novel alkenones. 相似文献

Direct injection of the vascular endothelial growth factor (VEGF) gene plasmid DNA into the myocardium was shown to induce development of new blood vessels to increase the circulation in the heart of patients with coronary artery diseases. However, such angiogenic gene therapy (via naked DNA) was limited by low level of gene expression. Furthermore, the temporal and spatial characteristics of VEGF gene transfer in the heart are not known. In this study, we demonstrated that a plasmid vector, containing the human cytomegalovirus immediate early (HCMV IE) promoter and enhancer, induces greater expression of gene in the rat heart monitored by gene fused to the chloramphenicol acetyl transferase (CAT) reporter, than four different viral and cellular promoters. Interestingly, expression of VEGF121 protein showed an earlier peak, a shorter duration, and a wider distribution than that of CAT only. Therefore, a plasmid vector with an HCMV IE promoter/enhancer provides clear advantages over other previously developed plasmids. Furthermore, expression profile of VEGF121 gene may provide useful information in the design of angiogenic gene therapy in the heart. 相似文献

Biotin is a vitamin of the B-complex, which plays an important biochemical role in every living cell. In the recent years, the interest in this vitamin has been rekindled, mainly due to its association with serious human disorders, such as the inherited syndrome multiple carboxylase deficiency, which can be successfully treated with biotin administration. Diagnosis of biotin deficiency as well as monitoring of biotin levels in biological fluids of patients receiving biotin treatment is crucial. Equally important is the determination of biotin levels in pharmaceutical preparations as well as in food and food supplement products, which constitute the main source of biotin in humans. Several analytical methods for measuring biotin in various samples, e.g. human fluids, pharmaceutical formulations, food material etc., have been reported in the literature. A method was developed for the recovery and determination of cyanide ion in organic sample matrices. To facilitate the solubilization of cyanide ions, cetyltrimethylammonium bromide (CTAB) was added at concentrations above the critical micelle concentration. Sample cyanation reaction products consisted of solvent mixtures of a hydroxynitrile in DMF-toluene or DMF-isopropylacetate (IPAC). Spectrophotometric determination of cyanide ion at 578 nm by the pyridine-barbituric acid method was automated by flow injection analysis. Recovery of cyanide ion from spiked samples was 93.2% in DMF-IPAC solvent matrix and 93.9% in DMF-toluene. Low alkali concentration was observed to favor solubilization of cyanide ion in the micellar solution. 相似文献
