理想的坚守与人性的洞察—迟子建小说创作论

迟子建以《北极村童话》为代表的早期小说基本属于故乡题材小说,从90年代初开始,迟子建的小说题材日趋扩大,由从前单纯描写故乡风情逐渐转向关注都市社会,风格也由单纯明净趋向浑厚成熟;题材的扩大没有改变迟子建的文学理想,在小说中建构温情与爱意的家园是她对理想的执着坚守;迟子建的小说包含对人性最深刻的洞察,她笔下人物浓郁的忏悔意识体现了最丰富、最真实的人性。     

《蚀》和《莎菲女士的日记》女性形象比较

茅盾在他的小说处女作《蚀》三部曲中刻画了一群个性鲜明的“新女性”形象,文学史上称她们为“时代女性”。丁玲的早期小说《莎菲女士的日记》为中国现代文学创造了一个“莎菲性格”,她后来的多篇小说人物都是“莎菲型”形象的拓展和延伸。比较分析茅盾的“时代女性”和丁玲的“莎菲型”女性形象,可知：在人生理想的追求上,“时代女性”是革命的理想主义者,而莎菲女士是启蒙的个人主义者;在生活的逆境面前,“时代女性”在绝望中求生,莎菲女士却在颓废中耗尽生命;在对待爱情的态度上,“时代女性”狂热地追逐爱情只为填补内心的空虚,而莎菲女士执着地将灵肉一致的爱情作为自己矢志不渝的追求。     

B链羧端去三肽(B28～B30)胰岛素晶体结构

采用悬滴气相扩散法，获得了Ｂ链羧端去三肽胰岛素３种晶型的单晶体，其中 晶型１属于Ｐ２_1空间群，晶胞参数为ａ＝４．７７ ｎｍ，ｂ＝６．１９ ｎｍ，ｃ＝６．１２ ｎｍ，β＝１１０．３°；晶 型２属于Ｐ４_１２２或Ｐ４_３２２空间群，晶胞参数为ａ＝ ６．４５ ｎｍ， ｃ＝ １２．０７ ｎｍ；晶型３属于 Ｐ２_１２_１２_１空间群，晶胞参数为 ａ＝ ４．９８ ｎｍ， ｂ＝ ５．１６ ｎｍ， ｃ＝ １０．０６ ｎｍ．采用分子置换法， 测定并精化了 Ｂ链羧端去三肽胰岛素晶型１的 ０．２３ ｎｍ分辨率晶体结构．最终模型的 Ｒ 因子为１８．８％，键长和键角与标准键长和键角的均方根偏差分别为０．００１５ ｎｍ和３．３°． 晶体结构的结果表明， Ｂ链羧瑞去三肽胰岛素失去了对缔合有重要作用的 Ｂ２８ Ｐｒｏ后， 虽然仍能形成六聚体，但表现出比天然胰岛素更易解聚的结构变化。     

正交晶型江浙蝮蛇毒碱性磷脂酶A2初步结晶学研究

The basic phospholipase A2 from the venom of Agkistrodon halys pallas possesses the ability to cause hemolysis in contrast to the other two phospholipase A2 from the same venom. A new form of crystals of this enzyme was grown. The crystals belong to space group of P212121 with unit cell parameters of a=9.175nm, b=10.080nm , c=2.287nm.The crystals diffract to high resolution, and are suitable for detailed structural studies. The data were collected up to 0.25nm resolution using synchrotron radiation-imaging plate-Weissenberg camera system. Preliminary analysis reveals the presence of two molecules in the asymmetric uint and the molecule may pack with the smallest dimension approximately parallel to the c axis. The new crystal form is more attractive than the monoclinic one previously reported for crystallographic structure determination as it contains fewer molecules in the asymmetric unit.     

微重力下的蛋白质晶体生长

空间微重力环境是研究复杂的蛋白质结晶过程和生长高质量蛋白质晶体的理想地方。于1992年8月应用国内研制的装置,在返回式卫星FSW-2上完成了空间蛋白质晶体生长实验。有6种蛋白质在空间长出了晶体,其中蛇毒酸性磷脂酶A_2和鸡蛋清溶菌酶的晶体明显优于地面对照实验长出的晶体,表明空间的微重力利于改进蛋白质晶体的大小、完整性、形态及内部有序性。     

Preliminary crystallographic analysis of recombinant chicken liver fructose-2,6-bisphosphatase

The bisphosphatase domain of chicken liver 6-phosphofructo-2-kinase/fructose-2,6-bisphos-phosphatase was expressed in high yield Escherichia coli and purified to homogeneity. Single crystals of chicken liver bisphophatase domain suitable for X-ray diffraction were obtained by the hanging drop vapor diffusion method. The crystals belong to tetragonal space group P41212 or P43213 with two molecules per asymmetric unit. The determined cell dimensions are: a=b= 10.02 nm, c= 13.98 nm, α=β=г=90°. EHffraction data were collected on Weissenberg camera with synchrotron radiation at 0.32nm resolution.     

Crystal structural studies of destripeptide (B28-B30) insulin

Single crystals of destripeptide (B28-B30) insulin (DTRI) in three forms were obtained by hanging-drop vapor diffusion method. Form 1 belongs to P21 space group with cell parameters a-4.77 nm, b=6.19 nm, c=6.12 nm, β=110.3°. Form 2 belongs to P4122 or P4322 space group with cell parameters a= 6.45 nm, c=12.07 nm. Form 3 belongs to P212121 space group with cell parameters a=4.98 nm, b=5.16 nm, c=10.06 nm. The structure of form 1 crystal was determined by molecular replacement method and refined at 0.23 nm resolution. The R-factor of the final model is 18.8% with r.m.s. deviations of 0.001 5 nm and 3.3?for the bond lengths and the bond angles, respectively. Studies on the crystal structure show that the removal of B28 Pro has brought DTRI structural changes which made it dissociate more easily than native insulin although DTRI can still form a hexamer.
