Interactions of polyamidoamine dendrimers, termed Gn, where n indicates the generation (=number of amidoamine layers), at different protonation levels with selected amino acids and proteins have been investigated by means of continuous wave electron paramagnetic resonance (cw-EPR) and pulsed-EPR (electron spin-echo = ESE) analyses. A low-generation dendrimer (G2) and a high-generation one (G6) were labeled with nitroxides for the EPR measurements. Gly, Glu, Arg, and Leu were selected as representative of neutral(zwitterionic)-polar, acidic, basic, and low-polar amino acids, respectively. The water-soluble proteins alpha-chymotrypsin and albumin were selected on the basis of a basic and an acidic isoelectric point, respectively. The cw-EPR spectra were analyzed by computing the line shapes to extract information about the dendrimer-biomolecule interactions. In general, dendrimers at a high protonation level interact stronger with amino acids than those at a low level of protonation. However, even for highly protonated dendrimers, a synergistic effect between hydrophilic and hydrophobic interactions promoted the formation of stable Gn-amino acid adducts, as demonstrated by the enhanced interactions with Leu. As expected from acid-base interactions, stable adducts were formed between Arg and highly protonated dendrimers and between Glu and low level protonated dendrimers. The relatively strong dendrimer interactions with the protein chymotrypsin and the poor interactions of dendrimers with albumin demonstrated that the protonated amino groups of the dendrimers are predominantly involved in the interactions with these proteins and indicated a significant role in the interactions with the dendrimers of the hydrophobic external residues of chymotrypsin. Computer-aided analysis of the ESE experiments was consistent with the cw-EPR results and supported the conclusion of a partial complexation of the nitroxides of the dendrimer with Leu and alpha-chymotrypsin.  相似文献

This work describes the syntheses of a new poly(amidoamine) (PAMAM) dendrimer family possessing a disulfide function (cystamine) in its core. Traditional redox-chemistry associated with the disulfide core in these dendrimer structures, provides a versatile strategy for designing unique sizes, shapes and controlling the regio-disposition of chemical groups on the surface of these dendrimers. Various single site, sulfhydryl functionalized dendron reactants may be generated in situ, under standard reducing conditions (i.e. dithiothreitol (DTT)). Facile control of size, shape and chemical functionality placement involves covalent hybridization of these single point, sulfhydryl reactive dendron components. This is accomplished by re-oxidation in the presence of air, to yield generation/surface chemistry differentiated cross-over products which may be isolated by preparative thin layer or column chromatography. Differentiated cystamine core dendrimers derived from combination and permutation of lower generation (i.e. Gen.=0-3) sulfhydryl functionalized dendrons possessing amino, hydroxyl, acetamido or dansyl surface groups, were synthesized and isolated. They were characterized by a variety of methods including; 13C NMR, capillary electrophoresis (CE), gel electrophoresis (PAGE), thin layer chromatography (TLC) and electrospray (ES) or matrix assisted laser desorption ionization (MALDI-TOF) mass spectrometry. This general strategy has broad implications for the systematic size, shape and regio-chemical control of a wide range of dendritic nanostructures, many of which may be designed to mimic the sizes, shapes and regio specific chemo-domains observed for globular proteins.  相似文献

2-Aryl- and 2-alkyl-2-oxazolines have been polymerized to poly-(N-aroyl)aziridines and poly(N-acyl)aziridines, respectively, in the presence of boron trifluoride. The polymers obtained were glassy, light yellow resins with molecular weights ranging from 3500 to 7500 (35–50 oxazoline units per chain). The polymerization rates have been determined for several of these monomers. A polymerization mechanism is proposed.  相似文献

Polyamidoamine dendrimers have been studied extensively for their potential applications in nanomedicine. Their uses as imaging, drug, and nucleic acid delivery agents are nearing clinical trials. As such, characterization of polyamidoamine dendrimers and their nano-devices is of immense importance for monitoring the efficiency of their synthesis, purity, and quality control of manufactured products as well as their in vivo behavior. We report here the analysis of polyamidoamine dendrimers possessing various cores and surface groups with a simple and inexpensive isoelectric focusing method. The isoelectric points of the dendrimers were readily determined from a calibration plot generated by running proteins with known pI values. The isoelectric points for various surface-modified polyamidoamine dendrimers ranged from 4 to 9. Polyamidoamine dendrimers possessing terminal hydroxyl groups gave a pI?>?7, while those with terminal carboxyl groups exhibit a pI?<?7. Generation number and cores of the dendrimers did not significantly affect their isoelectric points. Isoelectric focusing thus offers another important tool for characterizing these nanomolecules.

Small-angle x-ray scattering was used to characterize the single-particle scattering factors produced by poly(amidoamine) dendrimers, poly(propleneimine) dendrimers, and polyol hyperbranched polymers in dilute solutions with methanol as solvent. Fits from electron density modeling reveal similar overall densities of the dendrimers as a function of dendrimer generation. The seventh through tenth generation poly(amidoamine) dendrimers exhibit higher order scattering features that require nearly monodisperse, spherical particles with essentially uniform internal segment densities. Dilute hyperbranched polymer solutions exhibit scattering more indicative of the inherent irregularity of internal segment densities and overall sizes to be expected within these systems. Radii of gyration estimated from electron density modeling agree reasonably well with those estimated by standard Guinier methods used in previous studies. © 1997 John Wiley & Sons, Inc. Silica nanoparticles suspended in an organic solvent (nanosuspension) have been imaged and characterized via terahertz nanoscanning reconstructive three-dimension (3D) imaging technique. The size of individual silica nanoparticles in the suspension was quantified. In addition, the presence of nanoclusters along with their distribution in the suspension was visualized in 3D. It has also been qualitatively demonstrated that the volume fraction of solvent is significantly higher than that of the silica nanoparticles; an observation consistent with the composition of the nanosuspension in the present investigation. The measured size range of individual nanoparticles was found to be 10–12 nm, while the manufacturer’s specification indicates a nanoparticle size distribution in the range of 10 to 15 nm. However, a typical nanocluster size was determined to be 17.5 nm, thus indicating the presence of nanoparticles less than 10 nm. The nanometrology instrument used in this investigation was based on a dendrimer dipole excitation-based continuous wave terahertz source generating >?200 mW stable terahertz power.  相似文献

The nanoscopic domain of structural complexity, which ranges from 1 to 100 run on a particle size scale includes a relatively unexplored area of science which resides between classical chemistry and molecular biology. This rapidly growing area of science is referred to as nanoscopic chemistry and architecture. Concepts evolving in this area lead to a rich variety of precise structures, architecture and properties. These concepts are based on dendritic macromolecules in general and on Starburst® dendrimers in particular. They envision dendrimers as fundamental building blocks which may be used to synthesize nanoscopic compounds, clusters, polymers, etc. Accordingly, dendrimers are regarded architecturally as functional analogues of atoms; therefore, their potential role in nanoscopic chemistry may be compared to that of the atoms in classical chemistry.  相似文献

Recently developed multifunctional cancer therapeutic nano-device production is based on poly(amidoamine) PAMAM generation 5 (G5) dendrimer as a carrier 1-5. Scale up synthesis of this nano-device is limited because of long reaction sequence (12 reaction steps) and long and not easy work up of the products after each reaction step. Combination of poly(propyle-imine) and poly(amidoamine) synthesis can improve the production of the drug carrier.In this paper we give a general overview of the synthesis and characterization of a series of novel hybrid dendrimers which we coined as novel POMAM hybrid dendrimers, constructed from poly(propylene-imine) (PPI or POPAM) core and poly(amidoamine) PAMAM shells. The synthesis was accomplished by a divergent reiterating method involving repeating subsequent Michael addition and amidation reactions. Each generation of the newly synthesized dendrimer was characterized by using HPLC, GPC, NMR and AFM.  相似文献

Gd(III)-containing dendrimers are promising contrast agents for magnetic resonance imaging (MRI). An important issue in the effectiveness and toxicity of a Gd(III) based MRI contrast agent is knowledge of the relative locations and concentrations of Gd(III) in dendrimer drug delivery hosts. In order to provide experimental information on this issue, we have investigated the electron paramagnetic resonance (EPR) of a stable Gd(III) complex with diethylenetriaminepentaacetic acid (DTPA) in various polyammidoamine (PAMAM) dendrimers as a function of dendrimer generation (G2, G4, and G6), dendrimer core (ethylenediamine = EDA, and cystamine = cys), and dendrimer surface functionality (NH(2), 5-oxo-3-pyrrolidinecarboxylic acid methyl ester = pyr, and tris(hydroxymethyl) methylamine = tris). The dendrimer systems were investigated in the presence and absence of paramagnetic probes, that is, Cu(II) and nitroxide radicals (4-(trimethylammonium and dodecyl-dimethylammonium) 2,2,6,6-tetramethylpiperidine 1-oxyl bromide = CAT1 and CAT12, respectively). The analysis of the EPR spectra revealed anisotropic locations of Gd-DTPA inside the dendrimer. Computer analysis of the EPR spectra of the probes identified the interactions of the Gd-dendrimers with ions and organic molecules. The interaction between the probes and the dendrimer internal and external surface depends on the type of core, the composition of the external surface and the generation of the dendrimer. The negatively charged Gd-DTPA complex attracts the positively charged species and this provokes spin-spin interactions between Gd and the probes, which increases with a decrease in generation, mainly from G6 to G4, and with an increase in both the Gd-dendrimer concentration and the probe concentration. The cys core increases the internal volume and decreases the packing of the branches.  相似文献
