The binding constants of camptothecin, topotecan and its lactone ring-opened carboxylate derivative to DNA octamers were measured by UV and NMR spectroscopy. The self-association of topotecan (TPT) was also measured. The carboxylate form of TPT binds in the same way as the lactone, but more weakly. Titration of TPT into d(GCGATCGC)2 shows a preferred location stacked onto the terminal G1 base. However, the intermolecular NOEs cannot be reconciled with a single conformation of the complex, and suggest a model of a limited number of conformations in fast exchange. MD calculations on four pairs of starting structures with TPT stacked onto the G1-C8 base pair in different orientations were therefore performed. The use of selected experimental "docking" restraints yielded ten MD trajectories covering a wide conformational space. From a combination of calculated free energies, NOEs and chemical shifts, some of the structures produced could be eliminated, and it is concluded that the data are consistent with two major families of conformations in fast exchange. One of these is the conformation found in a crystal of a TPT/DNA/topoisomerase I ternary complex [Proc. Natl. Acad. Sci. USA 2002, 99, 15 387-15 392].  相似文献

During the first two years of operation, the TCV tokamak has produced a large variety of plasma shapes and magnetic configurations, with 1.0B tor1.46T,I p800kA,k2.05, –0.71. A new shape control algorithm, based on a finite element reconstruction of the plasma current in real time, has been implemented. Vertical growth rates up to 1000s–1 have been stabilized using the external coil system. Ohmic H-modes with Troyon factors ( tor aB/I p) up to two and densities up to 2.25×1020m–3, corresponding to the Greenwald limit, have been obtained in diverted discharges. Limiter H-modes with line averaged electron densities up to 1.7×1020m–3 have been obtained in elongated D-shaped plasmas with 360 kAI P600 kA.Presented at 17th Symposium Plasma Physics and Technology, Prague, June 13–16, 1995.This work was partly supported by the Fonds National Suisse de la Recherche Scientifique.  相似文献

A fused-silica capillary that is wall-modified via chemically bonding a sulfonated polymer to the capillary wall has a uniform negative charge density on its surface and produces an electroosmotic flow (EOF) greater than 4 x 10(-4) cm2 V(-1) s(-1) The EOF is nearly independent of buffer pH over the pH range of 2 to 10 and is lower than the EOF obtained for the bare fused-silica capillary at the more basic pH but is higher at the more acidic buffer pH. Optimization of buffer pH can be based on analyte pKa values to improve the overall quality of the capillary zone electrophoresis (CZE) separation of complex mixtures of weak acid and base analytes. Because of the high EOF in an acidic buffer, the capillary is useful for the separation of weak organic bases which are in their cation forms in the acidic buffer. EOF for the sulfonic acid bonded phase capillary can be adjusted via buffer additives such as organic solvent, tetraalkylammonium salts, multivalent cations and alkylsulfonic acids. The advantages of utilizing buffer pH and the EOF buffer modifiers to enhance migration time, selectivity, and resolution in CZE separations with this capillary are illustrated using a series of test analyte mixtures of inorganic anions, carboxylic acids, alkylsulfonic acids, benzenesulfonic acids, sulfas, pyridines, anilines or small-chain peptides.  相似文献

The sol–gel process is widely used for the production of powders, coatings and bulk materials. However, being a wet-chemical technique, it has certain limitations related to properties of aqueous colloidal solution, especially when applied as a coating. The most frequently used methods, such as dip- and spin-coating, are difficult to apply onto more complex substrates. In these cases, the aerosol–gel deposition method can be regarded as the solution of this problem. In the present article, a novel plasma enhanced aerosol–gel method of coatings production is presented. A novelty of this method is based on an integration of the aerosol–gel deposition of thin films and their low temperature plasma treatment. Owing to the above, all stages of the coatings production process—substrate preparation, film deposition, and its plasma treatment, can be carried out in a single reactor. The design and operational scheme of such device is presented in this work. Using this device, thin coatings were first deposited on substrates and then plasma treated. The effect of deposition and plasma discharge conditions on morphology and chemical structure of the films has been studied. It was found that plasma treatment had a substantial influence on all the examined properties of the aerosol–gel deposited coatings.  相似文献

A double discharge TEA CO2 laser of simple design has been tested. The linearly polarized beam delivers a total energy of 14 Joules in 25 ns. The influence of the different gas mixtures on the problem of arcing, delay between pumping and lasing, and evolution of the light pulse, has been investigated. Using 0.1% xylene in the gas mixture, 200 Joules per liter were fed in the glow discharge without arcing. The maximum efficiency is 6.5%.
Résumé Le comportement d'un laser CO2TEA à deux décharges est étudié. Il délivre des impulsions de lumière polarisée d'une énergie de 14 Joules et durant 25 ns. L'influence de la composition du mélange gazeux sur des phénomènes tels que l'arcage, le dálai entre le pompage et l'émission lumineuse ainsi que sur la forme de l'impulsion, a été étudiée. L'introduction de 0.1% de xylène dans le mélange gazeux a permis de dissiper une énergie de 200 Joules par litre de décharge sans produire d'arc. Le rendement maximum a été de 6.5%.

A lepton-pair trigger was used to select a hadronic system in different flavour and colour states: properties of the transformation of this system into hadrons were measured in terms of charge flow, multiplicity distribution, and fragmentation function. It is observed that, while the effects of flavour are clearly seen, the colour degrees of freedom are not observed in the region of low pT.  相似文献

NH3 temperature-programmed desorption (NH3-TPD) is frequently used for probing the nature of the active sites in CuSSZ-13 zeolite for selective catalytic reduction (SCR) of NOx. Herein, we propose an interpretation of NH3-TPD results, which takes into account the temperature-induced dynamics of NH3 interaction with the active centers. It is based on a comprehensive DFT/GGA+D and first-principles thermodynamic (FPT) modeling of NH3 adsorption on single Cu2+, Cu+, [CuOH]+ centers, dimeric [Cu-O-Cu]2+, [Cu-O22−-Cu]2 species, segregated CuO nanocrystals and Brønsted acid sites (BAS). Theoretical TPD profiles are compared with the experimental data measured for samples of various Si/Al ratios and distribution of Al within the zeolite framework. Copper reduction, its relocation, followed by the intrazeolite olation/oxolation processes, which are concomitant with NH3 desorption, were revealed by electron paramagnetic resonance (EPR) and IR. DFT/FPT results show that the peaks in the desorption profiles cannot be assigned univocally to the particular Cu and BAS centers, since the observed low-, medium- and high-temperature desorption bands have contributions coming from several species, which dynamically change their speciation and redox states during NH3-TPD experiment. Thus, a rigorous interpretation of the NH3-TPD profiles of CuSSZ-13 in terms of the strength and concentration of the active centers of a particular type is problematic. Nonetheless, useful connections for molecular interpretation of TPD profiles can be established between the individual component peaks and the corresponding ensembles of the adsorption centers.  相似文献

为提高磁体系统安装精度,在 HL-2M 集成大厅建立 63 个基准点构成测量基准网,并利用激光跟 踪仪等高精度测量设备建立每个磁体的局部坐标系,测量特征点的局部坐标;基于测量基准网和公共测量点,采 用最佳拟合得到坐标转换矩阵,以此得到特征点在测量基准网的位置,指导磁体安装。完成安装后的中心柱同支 撑基础的同轴度为∅2.03mm;PF1~PF4 线圈安装标高偏差为±0.5mm,与中心柱的同轴度为∅2.60mm;PF5/6/7/8 线圈与中心柱的同轴度偏差小于∅3.00mm,标高偏差在[−1mm, 1mm]区间内。基于以上方法所得到的线圈安装精 度都满足设计需求。  相似文献

Molecular structures of the isolated tetrahedral oxovanadium(IV) and bridged μ-oxo-divanadium(IV) complexes hosted by the clusters mimicking surfaces of amorphous silica-based materials were investigated using density functional theory (DFT) calculations. Principal values of the g and A tensors for the monomer vanadyl species were obtained using the coupled-perturbed DFT level of theory and the spin–orbit mean-field approximation (SOMF). Magnetic exchange interaction for the μ-oxo bridged vanadium(IV) dimer was investigated within the broken symmetry approach. An antiferromagnetic coupling of the individual magnetic moments of the vanadium(IV) centers in the [VO–O–VO]2+ bridges was revealed and discussed in detail. The coupling explains pronounced decrease of the electron paramagnetic resonance signal (EPR) intensity, observed for the reduced VO x /SiO2 samples with the increasing coverage of vanadia, in terms of transformation of the paramagnetic monomer species into the dimers with S = 0 ground state.  相似文献

Cross sections for the production of hypernuclei were measured in the reaction A(K, π) ΛA. on light and heavy nuclear targets, using a separated K beam at the CERN Proton Synchrotron. The pions were detected in the forward direction. The results are compared with cross sections calculated under the assumption that the reaction took place on a single neutron. The good agreement between the measured and the calculated cross sections justifies the use of the (K, π) reaction in order to obtain spectroscopic information on hypernuclei.  相似文献
