The convergent syntheses of pyrazinyl and quinoxalinyl phenylmethanones from 2-lithio pyrazines and quinoxalines

Pyrazines and quinoxalines bearing 2-substituents that direct ortho metalation reacted with lithium 2,2,6,6-tetramethylpiperidide to produce 2-substituted-3-lithiopyrazines and quinoxalines. These lithio reagents reacted with N-methoxy-N-methylbenzamide to give good to moderate yields of 3-substituted pyrazinyl or quinoxalinylphenylmethanones. The 3-methylthio substituents of some ketone products were oxidized to methylsulfonyl groups that were susceptible to nucleophilic displacement.     

Transient absorption spectra of 2-hydroxybenzophenone photostabilizers

Transient absorption spectra (400–600 nm) of 2-hydroxybenzophenone and the methyl methacrylate copolymer of 2-hydroxy, 3-allyl, 4,4'-dimethoxybenzophenone following 355 nm excitation (7–480 ps delay) are reported. A short-lived, 435 nm transient (τ ≈ 10 ps in CH₂Cl₂) for both molecules is assigned to the lowest excited singlet before internal proton transfer. Weaker, broad T-T absorption is observed after 480 ps.     

The Combination of Gas Chromatography With Mass Spectrometry

Abstract

The coupling of the techniques of gas chromatography and mass spectrometry has provided the chemist with one of his most powerful analytical methods. The analysis of such diverse substances as fuels, air pollutants, food flavors, smokes, protein hydrolyzates, pesticides, lipid extracts, etc. may all require the separation and identification of the components of extremely complex mixtures.     

Assessment of angiogenesis: implications for ultrasound imaging

In this paper, the fundamentals of tumor angiogenesis and the implications for ultrasound imaging will be described. Twenty-eight athymic nude mice were implanted with the human melanoma cell lines DB-1 or MW-9 (14 mice/group). Ultrasound contrast agents were injected in the tail veins. Power Doppler and pulse inversion harmonic imaging (PI-HI) was performed (in real time and intermittently). Ultrasound results were compared to immunohistochemical stains for endothelial cells (CD31), vascular endothelial growth factor (VEGF), and cyclooxygenase-2 (COX-2). Linear regression analysis indicated statistically significant correlations between percent area stained with COX-2 and with VEGF relative to power Doppler (p<0.05) and intermittent PI-HI (p<0.05) measures of tumor neovascularity in the MW-9 and the DB-1 mice, respectively. Preliminary results from a human trial of the anti-angiogenic drug Angiostatin (Entremed, Rockville, MD) showed tumor volumes increased in two patients, while the vascularity remained virtually unchanged. Conversely, in three patients with diminished tumor volumes vascularity increased by 38%. In conclusion, contrast enhanced ultrasound imaging of tumor neovascularity may provide noninvasive markers of angiogenesis and may become a useful tool for monitoring anti-angiogenic therapies in vivo.     

Synthesis and X-ray structural analysis of (2<Emphasis Type="Italic">Z</Emphasis>)-3(4-hydroxyphenyl)-2-pyridin-4-ylacrylonitrile and (2<Emphasis Type="Italic">Z</Emphasis>)-3-(3,5-di-<Emphasis Type="Italic">tert</Emphasis>-butyl-4-hydroxyphenyl)2-pyridin-4-ylacrylonitrile

Synthesis and X-ray structural investigations have been carried out for the two title compounds C₁₄H₁₀N₂O (3a) and C₂₂H₂₆N₂O (3b). Compound 3a crystallizes in the monoclinic space group P2₁/c, with a = 3.843(1) Å, b = 24.618(5) Å, c = 11.318(2) Å, = 92.61(3), V = 1069.7(4) Å,³ and Z = 4. Compound 3b crystallizes in the triclinic space group P-1, with a = 9.004(2) Å, b = 9.447(2) Å, c = 11.713(2) Å, = 76.70(3), = 83.12(3), = 82.16(3), V = 956.5(3) Å,³ and Z = 2. Both stilbazole derivatives have Z-geometry about the ethylene bridge which links the heterocyclic and aromatic rings. The molecular skeleton of 3a is slightly non-planar: the dihedral angles between the acrylonitrile linkage and the pyridine ring, and between this linkage and the p-hydroxyphenyl ring are 7.2(2) and 4.1(2), respectively. The molecular skeleton of 3b is less planar: the values of similar dihedral angles are 17.0(2) and 20.8(2), respectively. In the crystal of 3a, the molecules are packed in stacks along the a axis with head-to-head orientation. Intermolecular hydrogen bonds O=H s N and C=H s N link molecules into sheets parallel to (100) plane. In the crystals of 3b, the molecules have a head-to-tail orientation and intermolecular hydrogen bonds O=H s N link the molecules into infinite chains along [01-1] direction.
