Effect of aeration on lignin peroxidase production by Streptomyces viridosporus T7A

The effect of aeration on lignin peroxidase production by Streptomyces viridosporus T7A was studied in a bench-scale bioreactor using a previously optimized growth medium (0.65% yeast extract and 0.1% corn oil, pH7.0) at 37°C and natural pH. Airflow rates of 0.3, 1.0, and 1.5 vvm and a fixed agitation of 200 rpm were initially studied followed by 1.0 vvm and 200, 300, 400, and 500 rpm. The use of 1.0 vvm and 400 rpm increased enzyme concentration 1.8-fold (100–180 U/L) and process productivity 4.8-fold (1.4–6.7 U/[L·h]) in comparison with the use of 200 rpm and 0.3 vvm. The inexpensive corn oil, used as carbon source, besides its antifoam properties, proved to be nonrepressive for enzyme production.

Zusammenfassung Der Bleigehalt von pflanzlichem Material lÄ\t sich nach einer modifizierten photometrischen Bestimmungsmethode schnell und zuverlÄssig erfassen. Das extraktive Dithizonverfahren für Blei wurde hierzu systematisch getestet und hinsichtlich Schwierigkeitsgrad und Zeitbedarf der Arbeitsoperationen entscheidend vereinfacht. Absolutmengen bis zu einer unteren Grenze von 2 g bzw. 1 g Pb können in 2 Arbeitsbereichen mit einer Standardabweichung von ± 0,5 g bzw. ± 0,35 g Pb sicher bestimmt werden. Die Arbeit zeigt darüber hinaus den Gebrauch und den Wert einfacher mathematisch-statistischer Hilfsmittel für den Vergleich und die Beurteilung von Test- und Analysendaten.

Summary A modified photometric method is recommended for the rapid and reliable determination of lead in plant material. The dithizone extraction has been tested systematically and has been essentially simplified with regard to difficult operations and time consumption. Quantities down to a lower limit of 2 g or 1 g of Pb can be safely determined within two working ranges with a standard deviation of ±0.5 g or 0.35 g of Pb, respectively. Furthermore, the use of simple mathematicalstatistical means for comparison and evaluation of analytical data is shown.

     

Grundlagen photometrischer Analysenverfahren

Zusammenfassung Der violettrote Pyrophosphatkomplex des Mn^III in 2 n Perchlorsäure ermöglicht eine schnelle, zuverlässige und selektive photometrische Bestimmung von Haupt- und Nebenbestandteilen von Mangan in Verbindungen und Legierungen. Zusammensetzung und Struktur des Komplexes sowie die optimalen Bildungsbedingungen wurden systematisch untersucht und sind die Basis der entwickelten Arbeitsvorschrift. Die Ergebnisse von über 400 Testanalysen für die Arbeitsbereiche Makro und Halbmikro (55-0,5 mg Mn) wurden nach mathematisch-statistischen Methoden ausgewertet und lieferten objektive Daten über Extinktionskoeffizienten, Verfahrenskonstanten, Fehlergrößen und Selektivitätsverhalten gegenüber mehr als 50 Fremdionen, die in einer Bewertungstabelle zusammengefaßt sind. Als Beispiele wurden die Mn-Bestimmungen in Braunstein und Ferromangan ausführlicher beschrieben.

Summary The violet-red diphosphate complex of Mn^III in 2 n perchloric acid has been employed for a rapid, reliable and selective photometric determination of manganese in compounds and alloys. The complex and the conditions for its formation have been examined systematically. The results of more than 400 test analyses in the macro and semimicro range have been evaluated by mathematical-statistical methods and the analytical data obtained are presented in a table. More than 50 foreign ions have been considered. The determination of manganese in pyrolusite and ferromanganese is described as an example.

Elektronische Auswertung von Me\daten photometrischer Eichmessungen

Zusammenfassung Es wird ein Rechenmodell zur Auswertung photometrischer Eichmessungen skizziert, das die HomogenitÄt der Leerwerte überprüft und berücksichtigt, den optimalen funktionellen Zusammenhang zwischen Konzentration und Me\wert (Extinktion oder DurchlÄssigkeit) feststellt und anschlie\end das gesamte Datenmaterial auf Ausrei\er-Werte testet. Der Rechenablauf und die erforderlichen zahlreichen logischen Entscheidungen wurden in der problemorientierten Computer-Sprache ALGOL programmiert, so da\ sich die Auswertungen auf reine input/ output-Prozesse beschrÄnken können, die auch von Mitarbeitern ohne mathematische Kenntnisse durchführbar sind. An einem Anwendungs- beispiel mit zwei Eichme\reihen wird gezeigt, da\ die elektronische Auswertung weniger als 6 min beansprucht, wÄhrend der Zeitbedarf bei konventioneller Rechentechnik mehr als 200 fach grö\er ist.

Electronic evaluation of the data of photometric calibration measurements

A calculation model for the evaluation of photometric calibration readings ist described. The model checks and takes into account the homogeneity of the blank data, determines the optimum functional correlation between concentration and measured values (optical density or transmittance) and finally tests the whole data material with respect to runaway data. The course of calculation and the numerous logical decisions required were programmed in the problem-oriented computer language ALGOL. The evaluation can, therefore, be restricted to simple input/output processes which can be handled by people without mathematical background. An example of application with two series of calibration readings shows that the electronic evaluation takes less than 6 min, whereas the time consumption for conventional calculation technique is 200 times greater.

RESONANCE-ENHANCED CARS SPECTROSCOPY OF BILIPROTEINS. INFLUENCE OF AGGREGATION and LINKER PROTEINS ON CHROMOPHORE STRUCTURE IN ALLOPHYCOCYANIN (Mastigocladus laminosus)

Abstract— Resonance-enhanced coherent anti-Stokes Raman spectra (CARS) are reported for monomers and for trimers with and without linker proteins of allophycocyanin isolated from Mastigocladus laminosus. The CARS spectrum of the monomer is independent of the presence of linker proteins and is very similar to that of phycocyanin monomers indicating that the equivalent chromophores exhibit like structures in both biliproteins. Large differences are, however, observed between the spectra of phycocyanin trimers and those of allophycocyanin trimers with or without linker proteins (L_c^8,9). The observed differences between monomer and trimer spectra are consistent with a change of the α-chromophore-protein arrangement upon aggregation without linker. If linker proteins are present in the trimer, then additional geometry changes of the β-chromophores are induced; these could relate to a transition from the 15Z- anti to 15Z- syn conformation.
