D-3-Hydroxybutyrate dehydrogenase (BDH) is an NAD(+)-dependent dehydrogenase of the mitochondrial inner membrane involved in the energetic balance between the liver and peripheral organs in mammals. It allows the conversion of ketone bodies (acetoacetate and D-3-hydroxybutyrate) and it is one of the best documented lipid-requiring enzymes with a dependence on lecithins. After release of proteins from the membrane by phospholipase A2 treatment of salt-treated mitochondria, the rat liver enzyme is absorbed on controlled-pore glass beads. After batch washing, the enzyme, devoid of lipids (apoBDH), is specifically eluted at pH 8.05-8.15 with a 0.1 M Tris-1 M LiBr buffer under reducing conditions (5 mM dithiothreitol). It appears that during BDH absorption, the glass beads mimic the phospholipid surface of biomembranes. 相似文献

The influence of the loading path on the failure locus of a composite lamina subjected to transverse compression and out-of-plane shear is analyzed through computational micromechanics. This is carried out using the finite element simulation of a representative volume element of the microstructure, which takes into account explicitly fiber and matrix spatial distribution within the lamina. In addition, the actual failure mechanisms (plastic deformation of the matrix and interface decohesion) are included in the simulations through the corresponding constitutive models. Two different interface strength values were chosen to explore the limiting cases of composites with strong or weak interfaces. It was found that failure locus was independent of the loading path for the three cases analyzed (pseudo-radial, compression followed by shear and shear followed by compression) in the composites with strong and weak interfaces. This result was attributed to the fact that the dominant failure mechanism in each material was the same in transverse compression and in shear. Failure is also controlled by the same mechanisms under a combination of both stresses and the failure locus depended mainly on the magnitude of the stresses that trigger fracture rather than in the loading path to reach the critical condition. 相似文献

A series of Emivirine and GCA-186 analogues substituted at N-1 with indan-1-yloxymethyl (6a–6c) and indan-2-yloxymethyl (6d–6f) were synthesized by reaction of the corresponding bis(indanyloxy)methans with uracils having 5-ethyl or 5-isopropyl and 6-benzyl or 6-(3,5-dimethylbenzyl) substituents. A route to the corresponding N-1 substituted 4-hydroxybut-2-enyloxymethyl analogue was also devised. All newly synthesized compounds showed potent activity against wild-type HIV-1, the most active compound being 5-ethyl-1-(indan-1-yloxymethyl)-6-(3,5-dimethylbenzyl)uracil (6b), which was 50-fold more active than Emivirine. 相似文献

The methanolic extract of the bark of the medicinal plant Qualea parviflora (Vochysiaceae) contains new nor-seco-triterpene and pentacyclic triterpenoids. They were separated in a preparative scale using droplet counter-current chromatography. The optimum solvent used was composed of a mixture of CHCl3/MeOH/H2O (43:37:20, v/v) in the descending mode and led to a successful separation of the new compound 28-nor-17, 22-seco-2α, 3β, 19, 22, 23-pentahydroxy-Δ12-oleanane, besides the known triterpenoids bellericagenin B, bellericaside B and arjunglucoside I .
