The novel NAD+-linked opine dehydrogenase from a soil isolate Arthrobacter sp. strain 1C belongs to an enzyme superfamily whose members exhibit quite diverse substrate specificites. Crystals of this opine dehydrogenase, obtained in the presence or absence of co-factor and substrates, have been shown to diffract to beyond 1.8 ? resolution. X-ray precession photographs have established that the crystals belong to space group P21212, with cell parameters a = 104.9, b = 80.0, c = 45.5 ? and a single subunit in the asymmetric unit. The elucidation of the three-dimensional structure of this enzyme will provide a structural framework for this novel class of dehydrogenases to enable a comparison to be made with other enzyme families and also as the basis for mutagenesis experiments directed towards the production of natural and synthetic opine-type compounds containing two chiral centres.  相似文献

Here we present a system of coupled phase oscillators with nearest neighbors coupling, which we study for different boundary conditions. We concentrate at the transition to the total synchronization. We are able to develop exact solutions for the value of the coupling parameter when the system becomes completely synchronized, for the case of periodic boundary conditions as well as for a chain with fixed ends. Two methods are proposed for aspherical surface evaluation — scanning and interferometry. Each of these methods enables the determination of the radius of curvature at the lens vertex, the asphericity, astigmatism, rotation asymmetry, surface topography and the deviation of the surface from following the theoretically stated shape. While the scanning technique is low cost and simple, the interferometric technique (which needs optical arrangements) gives more information about the surface quality. These methods, combined with a computer, establish powerful instruments for controlling lens production.  相似文献

Tyrosinase is a multifunctional copper-containing oxidase enzyme that initiates melanin synthesis in humans. Excessive accumulation of melanin pigments or the overexpression of tyrosinase may result in skin-related disorders such as aging spots, wrinkles, melasma, freckles, lentigo, ephelides, nevus, browning and melanoma. Nature expresses itself through the plants as a source of phytochemicals with diverse biological properties. Among these bioactive compounds, flavonoids represent a huge natural class with different categories such as flavones, flavonols, isoflavones, flavan-3-ols, flavanones and chalcones that display antioxidant and tyrosinase inhibitor activities with a diversity of mechanistic approaches. In this review, we explore the role of novel or known flavonoids isolated from different plant species and their participation as tyrosinase inhibitors reported in the last five years from 2016 to 2021. We also discuss the mechanistic approaches through the different studies carried out on these compounds, including in vitro, in vivo and in silico computational research. Information was obtained from Google Scholar, PubMed, and Science Direct. We hope that the updated comprehensive data presented in this review will help researchers to develop new safe, efficacious, and effective drug or skin care products for the prevention of and/or protection against skin-aging disorders.  相似文献

New plastic membrane electrodes for amitriptyline hydrochloride (AmCl) based on amitriptylinium phosphotungstate (Am-PTA); amitriptylinium phosphomolybdate (Am-PMA) and a mixture of both (Am-PTA/PMA) were prepared. The electrodes were fully characterized in terms of their composition, life span, pH and temperature and then were applied to the potentiometric determination of the amitriptylinium ion in its pure state and pharmaceutical preparations and flow injection conditions. The selectivity of the electrodes towards many inorganic cations, sugars and amino acids was also tested.  相似文献

The design and operations of inpatient care facilities are typically largely historically shaped. A better match with the changing environment is often possible, and even inevitable due to the pressure on hospital budgets. Effectively organizing inpatient care requires simultaneous consideration of several interrelated planning issues. Also, coordination with upstream departments like the operating theatre and the emergency department is much-needed. We present a generic analytical approach to predict bed census on nursing wards by hour, as a function of the Master Surgical Schedule and arrival patterns of emergency patients. Along these predictions, insight is gained on the impact of strategic (ie, case mix, care unit size, care unit partitioning), tactical (ie, allocation of operating room time, misplacement rules), and operational decisions (ie, time of admission/discharge). The method is used in the Academic Medical Center Amsterdam as a decision-support tool in a complete redesign of the inpatient care operations.  相似文献

Syzygium cumini (Pomposia) is a well-known aromatic plant belonging to the family Myrtaceae, and has been reported for its various traditional and pharmacological potentials, such as its antioxidant, antimicrobial, anti-inflammatory, and antidiarrheal properties. The chemical composition of the leaf essential oil via gas chromatography–mass spectrometry (GC/MS) analysis revealed the identification of fifty-three compounds representing about 91.22% of the total oil. The identified oil was predominated by α-pinene (21.09%), followed by β-(E)-ocimene (11.80%), D-limonene (8.08%), β-pinene (7.33%), and α-terpineol (5.38%). The tested oil revealed a moderate cytotoxic effect against human liver cancer cells (HepG2) with an IC50 value of 38.15 ± 2.09 µg/mL. In addition, it effectively inhibited acetylcholinesterase with an IC50 value of 32.9 ± 2.1 µg/mL. Furthermore, it showed inhibitory properties against α-amylase and α-glucosidase with IC50 values of 57.80 ± 3.30 and 274.03 ± 12.37 µg/mL, respectively. The molecular docking studies revealed that (E)-β-caryophyllene, one of the major compounds, achieved the best docking scores of −6.75, −5.61, and −7.75 for acetylcholinesterase, α-amylase, and α-glucosidase, respectively. Thus, it is concluded that S. cumini oil should be considered as a food supplement for the elderly to enhance memory performance and for diabetic patients to control blood glucose.  相似文献

Reproterol hydrochloride (RpCl), selective PVC membranes based on ion associates of reproterolium-phosphotungstate (Rp-PTA); reproterolium-phospho-molybdate (Rp-PMA) or a mixture of both (Rp-PTA/PMA) were prepared. The electrodes displayed a linear response over the concentration range of 6.3×10−6–1.0×10−1 mol dm−3 RpCl. The working pH ranges of the above electrodes were 2.5–9.0, 2.5–8.5 and 2.0–9.0 and their isothermal temperature coefficients were 0.00014, 0.00090 and 0.00103 V/°C, respectively. The electrodes showed good selectivity to the reproterolium ion with respect to many inorganic cations, sugars and amino acids. The standard additions and potentiometric titration methods were used to determine RpCl in pure solutions and in its pharmaceutical preparations with high accuracy and precision.  相似文献
