A recently characterized oviposition-deterring pheromone (ODP, structure 1) of the European cherry fruit fly was used as a test case for probing the potential of tandem mass spectrometry (MS/MS) in structure elucidation as a stand-alone technique. The glycolipid-taurinate 1 was subjected to MS/MS analyses under a variety of conditions with and without preceding chemical degradation. Acidic methanolysis of 1 and subsequent in-batch derivatization (trideuterioacetylation) yielded methyl 2,3,4,6-tetrakis-O-trideuterioacetyl-glucopyranoside (2), methyl 8,15-bis-trideuterioacetoxy-palmitate (3), and taurine (4) as suitable target compounds for direct mixture analysis.Low energy collision induced dissociation (CID) on selected precursor ions (MS/MS on [M + H – CH3OH]+ and [M + H]+ produced by fast atom bombardment (FAB)) allowed direct identification of 2 and 4, respectively, by comparison with appropriate reference ions. In the case of 3, low energy CID (desorption chemical ionization (DCI) instead of FAB, MS/MS on [M + H]+) permitted deduction of gross molecular structure, but failed to provide positional detail. In sharp contrast,high energy CID of trideuterioacetylated intact 1 (FAB-MS/MS on [M – H] ions of la) clearly revealed a linear 8,15-hydroxylated palmitic acid backbone. Less certain was assignment of 15-O-glucosylation by this approach.  相似文献

The C42-C52 part of ciguatoxin CTX3C (1) was synthesized from tri-O-acetyl d-glucal. The synthetic segment had a tetrahydropyran ring corresponding to the ‘C49-reduced’ L-ring of 1, designed to avoid side reactions due to acid-labile C49 acetal carbon during acidic reductive conditions planned in further synthesis toward 1. The vicinal dimethyl part at C47-C48 was constructed by a stepwise conjugate addition/methylation procedure. The C50-C52 unit was installed by Grignard addition of the C3 unit followed by spirocyclization and reductive cleavage of the spirocyclic acetal. Stereoselective assembly of the C42-C44 part was achieved by Brown’s asymmetric crotylboration.  相似文献

The dynamic theory of linear piezoelectricity is applied to analyze the scattering of time harmonic flexural waves by a through crack in a symmetric piezoelectric laminated plate subjected to electric field loading. An incident wave giving rise to moments symmetric about the crack plane is considered. Piezoelectric layers are added to the upper and lower surfaces. Classical lamination theory is extended to include dynamic piezoelectric effects. Fourier transforms are used to reduce the problem to the solution of a pair of dual integral equations, the solution of which is then expressed in terms of a Fredholm integral equation of the second kind. The dynamic moment intensity factor vs. frequency is computed and the influence of the electric field on the normalized values is displayed graphically.  相似文献

Reversible phosphorylation of proteins represents an important component of cellular signaling pathways. The isolation of phosphoproteins in complex mixtures and the determination of the level of phosphorylation have been and remain a major challenge. It has prompted the development of several strategies, including immobilized metal affinity capture to enrich for phosphorylated peptides. An improved methodology was published (Ficarro, et al., Nature Biotechnology 2002, 20, 301-305) that showed increased selectivity through esterification of amino acid side chain carboxylic groups of enzymatically digested peptides. This method was applied for relative quantitation of phosphopeptides in conjunction with the use of stable isotope labeling. The merits and limits of the approach are discussed and its application to the analysis of the effects of serum starvation on in vitro cultured human lung cells is presented.  相似文献

Low energy collision induced dissociation (CID) spectra indicate that m/z 173 ions formed by the loss of a halogen atom from diethyl chloro- and bromo-succinate under electron impact are mixtures of O-protonated diethyl maleate (>90%) and fumarate (<10 %). Hydrogen migration precedes the C-halogen bond cleavage in these cases. The low energy CID spectrum of the m/z 173 [M? I]+ ion obtained from diethyl iodosuccinate shows that only a small fraction of the [M? I]+ ions are the O-protonated species formed by hydrogen migration. The results of this study demonstrate the potential of low energy CID in the determination of structure (including configuration) of gas phase ions.  相似文献

Permethylated oligosaccharides were analyzed by matrix-assisted laser desorption/ionization mass spectrometry (MALDI/MS) using a reflectron time-of-flight instrument in the post-source decay (PSD) mode. Under these ionization conditions, such derivatives yield intense signals corresponding to sodium or potassium cationized molecular species. Fragments observed in the PSD spectra result exclusively from cleavage of glycosidic bonds, preferentially at N-acetylhexosamine residues. A systematic study was carried out on a series of permethylated oligosaccharides to allow rationalization of the fragmentation processes. Fragments originating from both the reducing and the non-reducing ends of the oligosaccharide yield information on sequence and branching. Moreover, glycosyl residues linked in position 3 of HexNAc units give rise to a highly specific elimination process, which allows unambiguous assignment of (1-3) interglycosidic linkages. Special attention was paid to the structural analysis of oligosaccharides carrying the commonly encountered fucosyl and sialyl end-caps. In the case of sialylated residues, a targeted methodology involving desialylation and specific CD3-labeling of the nascent free hydroxyl groups was developed to mark the initial location of sialic acid residues along the oligosaccharide backbone. As accurate mass determination of fragment ions is essential for their assignment, a simplified protocol for the calibration in the PSD mode is described. This procedure allows the determination of the correction function parameters required to process the data for an instrument that employs post-acceleration detection. MALDI/PSD-MS of permethylated oligosaccharides, by providing structural information at the low picomole level, appears to be a valuable complement, or an alternative, to the techniques currently in use for carbohydrate structural analysis.  相似文献

Convergent synthesis of the IJKLM-ring part (2) of ciguatoxin CTX3C has been achieved from the I-ring and the L-ring parts (4 and 5) in total eight steps in 27% overall yield. The carbanion derived from 4, stabilized by a dimethyldithioacetal S-oxide group, was readily reacted with aldehyde 5 to give an adduct, which was facilely transformed into the corresponding α,ε-dihydroxy ketone 3. The JK-ring formation from 3 under reductive conditions followed by oxidative M-ring cyclization efficiently led to the pentacyclic ether 2. Improved synthesis of 6, a synthetic intermediate for 4, was also established.  相似文献

A new method for achieving the decyanation of disubstituted malononitrile derivatives without using reducing agents has been developed. Treatment of a six-membered malononitrile derivative with NaHMDS followed by methanol afforded the corresponding acetonitrile derivative in high yield. The present method was applicable to the decyanation reactions of a variety of malononitriles including four- and five-membered compounds as well as acyclic ones.  相似文献
