A simple, robust and sensitive sequential injection spectrophotometric method for the assay of penicillamine (PA) in pharmaceutical formulations is developed. The method was based on the complex formation when PA is reacted with iron(III) solution in hydrochloric acid media. The deep blue colored PA-iron(III) complex produced is monitored at a maximum wavelength of 600 nm. A five level orthogonal array design coupled to genetic algorithm was employed to obtain the optimum experimental conditions for the determination of PA using peak absorbance as the measure of the system's performance criterion. A linear dynamic range for the determination of PA of 25-300 ppm was obtained with a sampling frequency of 50 h−1 and a relative standard deviation of less than 0.98%. The method was successfully applied to the determination of PA in pharmaceutical formulations.  相似文献

OBJECTIVE: To establish a suitable human model for the study of the genetics of complex diseases. METHODS: We have selected an Omani Arab population to provide the statistical power required to study the genetics of complex diseases with confidence. This model consists of five multigenerational highly inbred pedigrees, descending from a small number of founders just a few generations ago with environmental homogeneity, restricted geographical distribution, detailed records and well-ascertained and -validated pedigrees. Stringent criteria were adopted for defining the phenotypes of hypertension, diabetes mellitus, dyslipidemias and obesity. The SOLAR genetic software package was used to draw the pedigree structure. RESULTS: Outstanding statistical power to detect susceptibility loci was obtained. CONCLUSIONS: This model represents a large homogeneous human family-based population for the study of genetic and environmental factors contributing to complex diseases.  相似文献

A deep understanding of both guided propagating and evanescent waves is very essential for non-destructive evaluation. Guided propagating waves have received considerable attention, but the research on guided evanescent waves is quite limited, especially for functionally graded structures. This paper presents an analytical approach, based on the orthogonal function technique, to solve the evanescent waves in a spherical curved plate with functionally graded material in the radial direction. The proposed approach can obtain all the real, imaginary and complex solutions but without iterative process. The validity of the proposed approach is illustrated by comparison with available results. The convergence of the approach is also discussed through a numerical example. Dispersion characteristics of the guided waves in various graded spherical curved plates are studied and three dimensional dispersion curves are plotted in frequency-complex wavenumber space. The effects of different radius-thickness ratios and graded fields on the dispersion curves of the guided evanescent waves are illustrated. The displacement distributions are also discussed to analyze the characteristics of the guided evanescent waves.  相似文献

A simple, robust and sensitive sequential injection spectrofluorimetric method for the determination of penicillamine (PA) in pharmaceutical formulations is developed. The method is based on the formation of a highly fluorescent derivative when penicillamine is reacted with fluorescamine (FL) in borate buffer of pH 9.3. The derivative produced is monitored at an emission wavelength of 495 nm using an excitation wavelength of 355 nm. The optimum conditions for the determination of PA with FL were: 3 mM FL, pH 9.3, 5 mM methyl-β-cyclodextrin, sample volume of 75 μL and reagent volume of 75 μL. Furthermore, the effect of various media on the fluorescence intensity of the PA–FL derivative was studied and methyl-β-cyclodextrin was found to give the largest enhancement. A linear dynamic range for the determination of PA of 5–80 ppm was obtained with a sampling frequency of 50 h−1 and a relative standard deviation of less than 2.5%. The method was applied to the determination of PA in pharmaceutical formulations with reasonable recoveries ranging from 101.0–103.1%, indicating that no interference is observed from concomitants usually present in dosage forms.  相似文献

An aqueous extract of Hibiscus sabdariffa L. is a common beverage in many parts of the world. Reports on its effect on reproduction are conflicting, with anecdotal evidence that the plant is an aphrodisiac, while others report that it is estrogenic, and adversely affects spermatogenesis in rats. We have studied the effect of different concentrations of aqueous extracts of H. sabdariffa calyces (10%, 15% and 20%) used as drinking water for 10 consecutive weeks, and its anthocyanins (50, 100, 200 mg/kg for 5 days, orally) on the weight and histology of the testis, and on some biochemical constituents in testicular homogenates, in addition to the plasma concentrations of testosterone, luteinizing hormone and estradiol. The possible presence of an estrogenic effect of the extract and anthocyanins on the uteri of immature female rats was also tested. Neither the H. sabdariffa extract nor the anthocyanins significantly altered either testicular weight and histology, or uterus weight. Plasma concentrations of the three hormones studied, the testicular concentrations of protein, reduced glutathione and total cholesterol, and superoxide dismutase activity were all insignificantly affected by either the extract or the anthocyanins, except for a slight, but statistically significant, decrease in testicular protein concentration caused by the 15% aqueous extract when compared with controls. These results suggest that H. sabdariffa exerts no adverse effect on the male reproductive system. Consumption of H. sabdariffa aqueous extract inhibited the growth of the rats compared with the controls.  相似文献

A simple, sensitive and rapid reversed-phase high-performance liquid chromatography (RP-HPLC) method is proposed for the analysis of some environmentally important phenols in water. The use of coumarin-6-sulphonyl chloride (C6SCl) as a fluorescence-labeling reagent has been investigated. The compound reacts with phenols within 20 min under mild conditions (ambient temperature, pH 9.0) to give sulphonates that can be separated by RP-HPLC employing fluorescence detection at lambda(ex) = 360 and lambda(em) = 460 nm. The optimum conditions for fluorescence, derivatization and chromatographic separation have been established and detection limits in the range 0.1-0.9 microg l(-1) were obtained for the studied compounds. The calibration curves were linear for the range 6-200 microg l(-1) for phenol, 3-200 microg l(-1) for 2-chlorophenol, 4-chlorophenol and 2,3,5-trichlorophenol and for the range of 3-100 microg l(-1) for 2,3-dichlorophenol and 3,5-dichlorophenol. The practical applicability of the method to environmental samples was demonstrated by analyzing drinking and industrial water samples spiked with the phenolic compounds.  相似文献

A highly efficient protein digestion device has been fabricated using commercially available immobilized trypsin on agarose beads, packed into a silica capillary and connected either directly to an electrospray mass spectrometer via a 'microtight T' connector, from which aqueous acetic acid (0.2%) was pumped, or via a monolithic column connected to the mass spectrometer ion source. Six proteins with molecular mass ranging from 2848 to 77703 Da were digested completely using this system. In the second set of experiments a short monolithic separation column was placed after the immobilized trypsin capillary and partial separation of the generated peptides was obtained. The detection limits were increased from the micromol to pmol range by utilization of this separation column. Gradient elution, using a binary HPLC pump and a flow splitter, was used to optimize the peptide separation. This provided significantly enhanced resolution of the tryptic peptides but increased the analysis time to 30 minutes.  相似文献

基于DTO分子( 1A1)的氢同位素效应,得到修正的Born-Oppenheimer(B-O)理论下多体展式分析势能函数.用准经典的Monte-Carlo轨迹法研究了D+ TO(0 ,0)和T+ DO(0 ,0)的分子反应动力学过程.结果表明:在碰撞能量较低时( < 209.2 kJ. mol - 1 ),D+ TO(0 ,0)和T+DO(0,0)反应主要生成O+ DT,并且该反应是无阈能的;有少量的交换反应产物DO (TO)生成,并伴有极少量的络合物产生.碰撞能大于209.2kJ . mol - 1后,逐渐出现分子被完全碰散成D,T ,O 原子的情形.反应 D+TO(0 ,0) →OD + T和 T +DO(0 ,0) →OT + D是无热反应但是有阈能存在.由于D和T原子的同位素效应,置换产物轨线存在非一致性.  相似文献

 An image-processing method is proposed to obtain the distribution of the removal efficiency of particles on a plate by an air jet. This method can be used to measure particle removal from a flat surface by processing the image of the reflected light from the surface. Factors affecting the particle removal efficiency such as air pressure, distance between the nozzle and the impinging surface and the impinging angle are discussed. The effect of detection chip geometry on chemiluminescence (CL) signal intensity of tris(1,10-phenanthroline)-ruthenium(II) peroxydisulphate system for analysis of chlorpheniramine maleate (CPM) in pharmaceutical formulations was investigated. It was observed that the design of the detection chip is very crucial and can play an important role in enhancing the CL signal intensity in this system. The CL signal intensity was enhanced 250% when a teardrop micromixer chip was used, compared to the commonly used serpentine chip geometry. The study was conducted using a multi-chip device. In this device, chip 1 was used to prepare and pump the reagent mixture, whereas chip 3 was used for pumping the sample. The two chips were connected to the teardrop chip (2) via silica capillary where detection took place. Non-linear regression curve fitting of the calibration data revealed that the calibration curves are best described by third order polynomial equation with excellent correlation coefficients (R2 = 0.9998) for the concentration range 7.69 × 10−8 to 5.12 × 10−5 mol L−1. A linear response is also observed over the range 7.69 × 10−8 to 1.28 × 10−5 mol L−1 (R2 = 0.9996) and the detection limit was found to be 5.49 × 10−8 mol L−1. The device was successfully used for the analysis of CPM in tablets and a multi-component cough syrup. Results were reproducible with relative standard deviation (RSD) of 0.6-1.1%.  相似文献
