The kinetics of cluster formation in supersonic jets is examined by numerical integration of the master equation system. Some general characteristics of cluster kinetics could be formulated. Excellent agreement between experimental curves of p-cresol (H2O)0, 1, 2, 3 formation as function of H2O pressure and the corresponding calculated curves were obtained assuming successive cluster formation. From the kinetic curves, an unambiguous assignment of cluster size was possible which agreed with mass-resolved REMPI measurements. The fit of the rate coefficients shows the formation of p-cresol (H2O)1 to be faster than p-cresol (H2O)2 and p-cresol (H2O)3. 相似文献

Quality evaluation of Flos lonicerae through a simultaneous determination of seven saponins by HPLC with ELSD 总被引:9,自引:0,他引:9
A new HPLC coupled with evaporative light scattering detection (ELSD) method has been developed for the simultaneous quantitative determination of seven major saponins, namely macranthoidin B (1), macranthoidin A (2), dipsacoside B (3), hederagenin-28-O-beta-D-glucopyranosyl(6-->1)-O-beta-D-glucopyranosyl ester (4), macranthoside B (5), macranthoside A (6), and hederagenin-3-O-alpha-L-arabinopyranosyl(2-->1)-O-alpha-L-rhamnopyranoside (7) in Flos Lonicerae, a commonly used traditional Chinese medicine (TCM) herb. Simultaneous separation of these seven saponins was achieved on a C18 analytical column. The mobile phase consisted of (A) acetonitrile-acetic acid (95:0.5) and (B) 0.5% aqueous acetic acid using a gradient elution of 29%A at 0-10 min, 29-46%A at 10-25 min and 46%A at 25-30 min. The drift tube temperature of ELSD was set at 106 degrees C, and with the nitrogen flow-rate of 2.6 l/min. All calibration curves showed good linear regression (r2>0.9922) within test ranges. This method showed good reproducibility for the quantification of these seven saponins in Flos Lonicerae with intra- and inter-day variations of less than 3.0% and 6.0%, respectively. The validated method was successfully applied to quantify seven saponins in five sources of Flos Lonicerae, which provides a new basis of overall assessment on quality of Flos Lonicerae. 相似文献

A novel potentiometric immunosensor for detection of Japanese B encephalitis vaccine was developed by immobilizing antiserum of Japanese B encephalitis on nano-Au/polymerized o-phenylenediamine (o-PDA) film on the platinum (Pt) electrode. The performance and factors influencing the performance of the resulting immunosensor were studied. The immunosensor showed a specific response to Japanese B encephalitis vaccine in the range 1.1 × 10−8 to 2.0 × 10−6 lgpfu/ml (plaque forming unit) with a detection limit of 6 × 10−9 lgpfu/ml. The correlation coefficient is 0.9986. The incubation time, incubation temperature, pH, reproducibility and stability of the immunosensor were also studied. A mild, efficient and convenient extraction method of using 2-mercaptoethanol contained extractant solution combined with an incubator shaker for determination of mercury species in biological samples by HPLC–ICP-MS has been developed. The effects of the concentration of 2-mercaptoethanol, the composition of the extractant solution and the shaking time on the efficiency of mercury extraction were evaluated. The optimization experiments indicated that the quantitative extraction of mercury species from biological samples could be achieved by using 0.1% (v/v) HCl, 0.1% (v/v) 2-mercapoethanol and 0.15% (m/v) KCl extractant solution in an incubator shaker for shaking overnight (about 12 h) at room temperature. The established method was validated by analysis of various biological certified reference materials, including NRCC DOLT-3 (dogfish liver), IAEA 436 (tuna fish), IAEA MA-B-3/TM (garfish filet), IAEA MA-M-2/TM (mussel tissue), GBW 08193 (bovine liver) and GBW 08572 (prawn). The analytical results of the reference materials were in good agreement with the certified or reference values of both methyl and total mercury, indicating that no distinguishable transformation between mercury species had occurred during the extraction and determination procedures. The limit of detection (LOD) for methyl (CH3Hg+) and inorganic mercury (Hg2+) by the method are both as 0.2 μg L−1. The relative standard deviation (R.S.D.s) for CH3Hg+ and Hg2+ are 3.0% and 5.8%, respectively. The advantages of the developed extraction method are that (1) it is easy to operate in HPLC–ICP-MS for mercury species determination since the extracted solution can be directly injected into the HPLC column without pH adjustment and (2) the memory effect of mercury in the ICP-MS measurement system can be reduced. 相似文献

Abstract— The analytical formulas previously developed for estimating the spectral irradiance reaching the ground in the ultraviolet are extended into the visible and infrared(350–3000 nm). This approach has two distinct features: (1) all physical inputs for calculating the direct irradiance are given in analytical form, and (2) the diffuse spectral irradiance (skylight) is calculated using dimensionless ratios which relate it to the direct irradiance. In common with other approaches, the global spectral irradiance for arbitrary ground reflectivity is calculated from the sum of the direct and diffuse spectral irradiances and a divisor which depends upon the ground and air spectral reflectivities. The global spectral irradiance on a tilted surface may also be calculated in terms of the above quantities and two angles. As in the case of the ultraviolet, the formulas presented are intended for photobiological applications. 相似文献

Synthetic gp120331-335 glycopeptide fragments carrying hybrid and high-mannose type N-linked glycans were evaluated for binding to broadly neutralizing antibody 2G12 using surface plasmon resonance technology. None of the hybrid-type constructs demonstrated binding to 2G12. In the high-mannose series, the "Cys dimer" construct, presenting two undecasaccharide glycans, showed significantly higher binding than the Cys-protected monomer. The binding of the dimeric structure was further investigated in competition with recombinant gp120. The data suggest that gp120 and its designed synthetic epitope construct bind to the same site on 2G12. 相似文献
