A rapid and accurate HPLC method has been developed for simultaneous determination of pioglitazone and glimepiride. Chromatographic separation of the two pharmaceuticals was performed on a Cosmosil C18 column (150 mm × 4.6 mm, 5 m) with a 45:35:20 (v/v) mixture of 0.01 m triammonium citrate (pH adjusted to 6.95 with orthophosphoric acid), acetonitrile, and methanol as mobile phase, at a flow rate of 1.0 mL min–1, and detection at 228 nm. Separation was complete in less than 10 min. The method was validated for linearity, accuracy, precision, limit of quantitation, and robustness [1, 2]. Linearity, accuracy, and precision were found to be acceptable over the ranges 2.50–30.00 g mL–1 for pioglitazone and 0.10–10.00 g mL–1 for glimepiride.  相似文献

Associated with every finite projective Hjelmslev plane is an invariant pair (t,r): t is the number of neighbours of a given point on a given line passing through it and r is the order of the underlying projective plane. The Drake-Lenz method [2],[3] of using auxiliary matrices for the constructions of projective Hjelmslev planes has become standard by now. This paper is intended to give some new constructions of projective Hjelmslev planes with invariant pairs (t,3) by making use of the generalization and improvement of the Drake-Lenz theorem [3] obtained by the author in [6] and [7]. The results of this paper add 8 new values to the list ([5], example 3.7(ii)) of invariant pairs (t,3) with t 1,000 for projective Hjelmslev planes.  相似文献

A sensitive, simple, and accurate high-performance liquid chromatographic method has been developed for determination of valdecoxib and the internal standard rofecoxib in human plasma. Protein was precipitated from plasma samples by addition of perchloric acid (HClO4); the drug was then extracted with diethyl ether. Separation was performed on a Cosmosil C18 column (150 mm × 4.6 mm i.d., 5 m particles) with ammonium acetate buffer-acetonitrile, 60:40 (v/v), containing 0.1% TEA, pH 6.5, as mobile phase. Detection and quantification were performed by UV-visible detection at 239 nm. Detection and quantification limits were 3 and 5 ng mL–1, respectively. The linear concentration range for valdecoxib was 5–400 ng mL–1. The validated RP HPLC method was used for determination of the pharmacokinetic data for the drug in humans.  相似文献

A simple, rapid and precise reverse-phase high performance liquid chromatographic method has been developed for the quantitative determination of eugenol from the extract of dried powder of Cinnamomum tamala leaves and its polyherbal formulation. Chromatographic analysis was carried out on Zorbax C18 column (150 mm × 4.6 mm, 5 μm) with a mobile phase of mixture of water, acetonitrile and methanol in the volume ratio of 50:40:10, at a flow rate of 1.0 mL min-1. Quantitation was performed using a UV-visible detector at 210 nm. Good linearity was obtained over the ranges of 0.20–3.0 μg mL-1 for eugenol.  相似文献

When a Padé-type function is used as a trial function, it is necessary to investigate the location of singularities and the zeros of the optimized function. It is shown that an unacceptable wave function can sometimes yield numerically acceptable values because of the nature of the integration procedure. Organic nanoparticles of a fluorinated tetraphenylporphyrin (TBTPP) were produced by rapid expansion of supercritical CO(2) solutions into both air (RESS) and an aqueous receiving solution containing a stabilizing agent (RESOLV). The effect of processing conditions on both particle size and form was investigated. The size of the porphyrin nanoparticles produced via RESS increased in a well-behaved manner from 40 to 80 nm as the preexpansion temperature increased from 40 to 100 degrees C, independent of porphyrin concentration, degree of saturation, and preexpansion pressure. RESOLV of TBTPP + CO(2) solutions was investigated both for minimizing particle growth in the free jet and for the prevention of particle agglomeration. Anionic, nonionic, and polymeric stabilizing agents for the aqueous receiving solution were considered. Expansion into a 0.05 wt % SDS solution produced nanorods 50-100 nm in diameter with an aspect ratio of 3-5. RESOLV in a 0.025 wt % Pluronic F68 solution produced well-dispersed, individual, spherical nanoparticles averaging 23 +/- 10 to 32 +/- 10 nm in diameter, independent of the rapid expansion processing conditions selected. Furthermore, the resulting nanoparticle suspensions were stable, with particle sizes remaining unchanged after several months. However, some particle agglomeration occurred at higher (i.e., 1 wt % TBTPP in CO(2)) concentrations. Contact-angle measurements on solid TBTPP compacts with the tested receiving solutions indicate that a moderate wetting agent such as Pluronic F68 is most effective for preserving the size and form of the porphyrin nanoparticles produced by RESOLV. Finally, the fact that nanoparticles are produced from RESS of TBTPP, in contrast with other organics for which microparticles are produced, can be explained in terms of the high melting point of TBTPP (388 degrees C), which results in a solid-state diffusion coefficient of TBTPP low enough so that particle coalescence is significantly reduced in the free jet.  相似文献
