The tetramer destabilization of transthyretin into monomers and its fibrillation are phenomena leading to amyloid deposition. Heparan sulfate proteoglycan (HSPG) has been found in all amyloid deposits. A chromatographic approach was developed to compare binding parameters between wild‐type transthyretin (wtTTR) and an amyloidogenic transthyretin (sTTR). Results showed a greater affinity of sTTR for HSPG at pH 7.4 compared with wtTTR owing to the monomeric form of sTTR. Analysis of the thermodynamic parameters showed that van der Waals interactions were involved at the complex interface for both transthyretin forms. For sTTR, results from the plot representing the number of protons exchanged vs pH showed that the binding mechanism was pH‐dependent with a critical value at a pH 6.5. This observation was due to the protonation of a histidine residue as an imidazolium cation, which was not accessible when TTR was in its tetrameric structure. At pH >6.5, dehydration at the binding interface and several contacts between nonpolar groups of sTTR and HSPG were also coupled to binding for an optimal hydrogen‐bond network. At pH <6.5, the protonation of the His residue from sTTR monomer when pH decreased broke the hydrogen‐bond network, leading to its destabilization and thus producing slight conformational changes in the sTTR monomer structure. In this paper, we introduce a new concept of semi-preemptive scheduling and we show how it can be used to derive a maximum-flow-based lower bound for the P|rj|Lmax which dominates the well-known preemptive lower bound. Ladjama Daif A series of chiral cyclosulfamides have been synthesized by alkaline cyclisation starting from N‐benzoylamino acids (Ala, Val, Leu, Phe) derivatives and chlorosulfonyl isocyanate. A simplified and regioselective deprotection of the cyclic compounds (cyclosulfamides) containing the tert‐butyloxycarbonyl group (Boc) has been achieved in good yield by fusion under reduced pressure.  相似文献

A simplified solid phase extraction method, eliminating a preliminary protein precipitation has been developed for the determination of celecoxib in rat plasma. The technique included a solid phase extraction of the serum samples on a [poly (divinylbenzene-co-N-vinylpyrrolidone)] sorbent. After conditioning, the cartridge was loaded with 0.5 mL of acidified serum containing internal standard. Elution was made with 1 mL of a mixture of acetonitrile and methanol (1/1, v/v). After evaporation of the eluate to dryness and reconstitution with methanol, the samples were analyzed on an octadecyl bonded phase with several mobile phases containing acetonitrile and a phosphate buffer. Detection was carried out using a Photodiode Array Detector. Full validation of the proposed method was provided (linearity range: 0.01–2 mg. L–1, average extraction efficiency: 92.4%; average intra-day variability: 4.6% with an accuracy of 94.8%; average interday variability: 5% with an accuracy of 95.3%, limit of detection: 0.005 mg. L–1, limit of quantification: 0.002 mg. L–1). Up to today, several techniques have been used to maintain cells in culture for studying many aspects of cell biology and physiology. More often, cell culture is dependent on proper anchorage of cells to the growth surface. Poly-l-lysine is commonly used as adhesive molecule. In this study, we present, as an alternative to poly-l-lysine, new polymer film substrates, realized by electropolymerization of different monomers on fluorine-doped tin oxide (FTO) surfaces since electropolymerization is a good method to coat selectively metallic or semiconducting electrodes with polymer films. So, the adhesion, proliferation and morphology of rat neuronal cell lines were investigated on polymer treated surfaces. Several amine-based biocompatible polymers were tested: polyethyleneimine (PEI), polypropyleneimine (PPI), polypyrrole (PPy) and poly(p-phenylenediamine) (PPPD). These polymer films were coated on FTO surfaces by electrochemical oxidation. After 8 h in a culture medium, a high percentage of cells was found to be attached to PEI and PPI compared to the other polymers and to the reference surfaces (glass and FTO uncovered). After 24 and 72 h in the culture medium, cells were found to proliferate faster on PEI and PPI than on other polymers and reference surfaces. Consequently, cells have a greater fold expansion on PEI and PPI than on PPPD, PPy or glass and FTO uncoated. From these results, we deduce that PEI and PPI can be useful as coating surface to cultivate neuronal cells.  相似文献

New hybrid organic-inorganic gels have been obtained by reaction of 1,4-butanediol, on tetramethoxysilane Si(OMe)4 dissolved in CCl4. This reaction does not require water and leads to the formation of polymeric transparent materials.Infrared, 29Si and 13C NMR spectroscopy shows that interchange reactions between OMe groups of alkoxide and -O-(CH2)4-O of 1,4-butanediol occurred, leading to the monolithic transparent gels in which both organic (Si-O-(CH2)4-O-Si) and inorganic (Si-O-Si) bridges are formed.  相似文献

A series of random polyesteramides (PEAs) within a range of molar composition from 90/10 to 10/90 were synthesized by a direct melt polycondensation of lactic acid and β-alanine. Their structures were fully characterized by NMR spectroscopy. The resulting copolymers are amorphous; they are thermally stable to temperatures up to 254°C, and present increasing glass transition temperatures at increasing amide content. The copolymers were also characterized by FTIR and viscosimetry measurements.  相似文献

The different contributions of the interfacial capacitance are identified in the case of passive materials or thin protective coatings deposited on the electrode surface. The method is based on a graphical analysis of the EIS results to determine both the passive-film capacitance in the high-frequency domain and the double-layer capacitance in the low-frequency domain. The proposed analysis is shown to be independent of the physicochemical origins of the frequency dispersion of the interfacial capacitances which results, from an analysis point of view of the experimental results, in the use of a constant-phase element However, for a correct evaluation of the thin-film properties such as its thickness, the high-frequency data must be corrected for the double-layer contribution. In particular, it is shown that if the double-layer capacitance gives a frequency-dispersed response, it is necessary to correct the high-frequency part for the double-layer constant-phase elements. This is first demonstrated on synthetic data and then used for the determination of the thickness of thin oxide film formed on Al in neutral pH solution.  相似文献

A series of new polyazomethines containing furan moieties was synthesized by polycondensation of bifuranic diamine monomers with commercially available aromatic dialdehydes viz., terephthaldehyde (TPA), isophthaldehyde (IPA). Inherent viscosities and number average molecular weights of polyazomethines were in the range 0.90–1.56 dL/g and 10460–17850 (SEC, polystyrene standard), respectively indicating formation of medium to reasonably high molecular weight polymers. The resulting polyazomethines were characterized by solubility tests, viscosity measurements, FTIR, NMR, UV spectroscopy, differential scanning calorimetric (DSC), and thermogravimetric analysis (TGA). These furan-based polyazomethines were essentially amorphous and exhibited glass transition temperatures (Tg) in the 150–190°C range. The temperature at 10% wt loss (T10), determined from TGA of polyazomethines were in the range 300–380°C, indicating their good thermal stability.  相似文献
