Five ester prodrugs of 2'3'-dideoxyinosine (DDI) were synthesized for the purpose of improving oral bioavailability. The prodrugs, acetate (C2-DDI), octanoate (C8-DDI), stearate (C18-DDI), benzoate (Bz-DDI), and hemisuccinate (Suc-DDI) were proved to quantitatively regenerate their parent drug by enzymatic hydrolysis. Though the chemical stability of the prodrugs under acidic conditions was not improved, their solubility in water was significantly decreased by esterification, except for Suc-DDI. Bioavailability was evaluated by oral administration to rats. Two hydrophobic prodrugs (C8-DDI and Bz-DDI) showed higher absolute bioavailability (23.5% and 31.0%, respectively) than did DDI (15.2%), though that of C2-DDI (11.5%) and Suc-DDI (4.5%) was poor.  相似文献

A membrane permeation-controlled transdermal delivery system (MC-TDS) of isosorbide dinitrate (ISDN), a model drug, was prepared from polyvinyl alcohol aqueous gel containing the drug, a membrane consisting of ethylene-vinyl acetate copolymer membrane and acrylic adhesive (EV-a). The permeability of ISDN through the EV-a membrane was 2.5 times higher than that through excised hairless rat skin. The ratio of plasma concentration of ISDN after application of MC-TDS on stripped (damaged) skin relative to intact skin was lower than that after application of Frandol tape-S, a marketed ISDN TDS, which suggests that the EV-a membrane might work as a control membrane for overall delivery rate of ISDN to the body. When MC-TDS stored at 30 degrees C for 13.5-48h was applied to the damaged skin, however, the initial plasma concentration of ISDN was very much higher than the expected therapeutic level and was not controlled by the EV-a membrane. The initial high plasma concentration of ISDN after application of the stored MC-TDS on the damaged skin was due to migration of ISDN from the reservoir to the adhesive during storage at 30 degrees C. The migration of drugs into the adhesive might be an important problem in developing efficient MC-TDS.  相似文献

X-ray spectroscopy studies of potassium intercalated metal-free phthalocyanine multilayers adsorbed on Al(110) have been undertaken. Photoelectron spectroscopy measurements show the presence of several charge states of the molecules upon K intercalation, due to a charge transfer from the alkali. In addition, the comparison of valence band photoemission spectra with the density functional theory calculations of the density of states of the H(2)Pc(-) anion indicates a filling of the formerly lowest unoccupied molecular orbital by charge transfer from the alkali. This is further confirmed by x-ray absorption spectroscopy (XAS) studies, which show a decreased density of unoccupied states. XAS measurements in different experimental geometries reveal that the molecules in the pristine film are standing upright on the surface or are only slightly tilted away from the surface normal but upon K intercalation, the molecular orientation is changed in that the tilt angle of the molecules increases.  相似文献

 This work concerns constructive aspects of measure theory. By considering metric completions of Boolean algebras – an approach first suggested by Kolmogorov – one can give a very simple construction of e.g. the Lebesgue measure on the unit interval. The integration spaces of Bishop and Cheng turn out to give examples of such Boolean algebras. We analyse next the notion of Borel subsets. We show that the algebra of such subsets can be characterised in a pointfree and constructive way by an initiality condition. The paper establishes, within constructive mathematics, a full and faithful functor M from the category of locally compact metric spaces and continuous functions into the category of formal topologies (or equivalently locales). The functor preserves finite products, and moreover satisfies f?g if, and only if, M(f)?M(g) for continuous . This makes it possible to transfer results between Bishop's constructive theory of metric spaces and constructive locale theory.  相似文献

Palladium nanoparticles immobilized on amino-functionalized mesocellular foam constitute an efficient catalyst for the aerobic oxidation of primary and secondary alcohols to their corresponding carbonyl compounds in high to excellent yields. An exceptionally high TON of 365?000 was reached for the oxidation of 1-phenylethanol under solvent-free reaction conditions. The catalyst can be recycled many times with retained activity as shown by the identical rate curves of the first and fifth runs.  相似文献

A straightforward, versatile approach to the production of protein gradients on planar and spherical particle surfaces is described. The method is based on the spatially controlled oxidation of thiolated surfaces by Au(III) ions generated via the electrochemical oxidation of a gold electrode in a phosphate-buffered saline solution (10 mM PBS, pH 7.2, 150 mM NaCl). Because the gold electrode is in direct contact with the thiolated surfaces, the released Au(III) ions, which are present as Au(III) chloride complexes, give rise to the formation of a surface gradient of Au(I)-thiolate complexes depending on the local redox potential given by the local Au(III) concentration. As is shown on the basis of the use of X-ray photoelectron spectroscopy and fluorescently labeled proteins, the Au(I)-thiolate complexes can subsequently be functionalized with thiolated proteins, yielding surface density protein gradients on micrometer-sized nonconducting polymer beads as well as linear Au(I)-thiolate gradients on planar silicon surfaces.  相似文献
