Objective: The present study aimed to develop and optimize esomeprazole loaded proniosomes (EZL-PNs) to improve bioavailability and therapeutic efficacy. Method: EZL-PNs formulation was developed by slurry method and optimized by 33 box-Bhekhen statistical design software. Span 60 (surfactant), cholesterol, EZL concentration were taken as independent variables and their effects were evaluated on vesicle size (nm), entrapment efficiency (%, EE) and drug release (%, DR). Furthermore, optimized EZL-PNs (EZL-PNs-opt) formulation was evaluated for ex vivo permeation, pharmacokinetic and ulcer protection activity. Result: The EZL-PNs-opt formulation showed 616 ± 13.21 nm of vesicle size, and 81.21 ± 2.35% of EE. EZL-PNs-opt exhibited negative zeta potential and spherical confirmed scanning electron microscopy. EZL-PNs-opt showed sustained release of EZL (95.07 ± 2.10% in 12 h) than pure EZL dispersion. The ex-vivo gut permeation result exhibited a significantly (p < 0.05) enhanced flux than pure EZL. The in vivo results revealed 4.02-fold enhancement in bioavailability and 61.65% protection in ulcer than pure EZL dispersion (43.82%). Conclusion: Our findings revealed that EZL-PNs formulation could be an alternative delivery system of EZL to enhance oral bioavailability and antiulcer activity.  相似文献

The first total synthesis of naturally occurring sacidumlignans A (1), B (2), and D (4) was executed and the absolute configuration of 2 and 4 was determined. A diastereoselective α- methylation of a lactone was used as the key step for the control of the chiral centers of the central lignan core. An acid mediated dehydrative cyclization of an aldehyde to construct the dihydronaphthalene unit of 2 and the aromatization of the intermediate dihydronaphthalene derivative to synthesize 1 are the key reactions employed in this regard.  相似文献

The main objective of study is assessment of chemo-radiological risk using measured concentrations of uranium in drinking water. For estimation of U concentration, LED Fluorimeter was used. For data accuracy, statistical tools were applied, geographical distribution GIS based software were used. To assess relation between uranium to other parameters, correlation test was performed. On the basis of presence of U in the water, hazard quotient, effective dose and cumulative dose for lifetime were estimated, it ranges from 0.001 to 1.09, 0.01–18.61 μSv year−1 and 1.22–1303 μSv for lifetime correspondingly. The estimated ingested dose is well below than the suggested limit of 0.1 mSv.

The purpose of this research was to mask the intensely bitter taste of aceclofenac (ACF) and to formulate oro dispersible tablet (ODT) of the taste-masked drug. Taste masking was done by complexing aceclofenac with Hydroxypropyl-β-Cyclodextrin (HPβCD) by different methods. Phase solubility studies indicated complex with possible stoichiometry of 1:1 and a stability constant of 221.11 M?1. The complexes were characterized by Fourier transform infrared spectroscopy, X-ray diffraction, and differential scanning calorimetry studies. The characterization studies confirmed inclusion of the ACF within the nonpolar cavity of HPβCD in the neutralization method (NM). Remarkable improvement in the in vitro drug release profiles in pH 6.8 phosphate buffer was observed with all complexes, especially the neutralization. The complexes of ACF–HPβCD (1:1) was compressed into tablet and properties of tablets such as tensile strength, wetting time, in vitro disintegration time, and disintegration in the oral cavity were investigated to elucidate the wetting and disintegration characteristics of tablets. Polyplasdone XL-10 7% wt/wt gave the minimum disintegration time. Tablets of batch F4 containing Avicel 200 and 7% wt/wt Polyplasdone XL-10 showed faster disintegration, within 12 s, than the marketed tablet (128 s). Good correlation between in vitro disintegration with in-house developed method and in the oral cavity was recognized. Taste evaluation of ODT in human volunteers revealed considerable taste masking with the degree of bitterness below threshold value. Thus, results conclusively demonstrated successful masking of taste and rapid disintegration of the formulated tablets in the oral cavity.  相似文献

Amisulpride (AMI) is an atypical antipsychotic having poor aqueous solubility and poor oral bioavailability. Inclusion complex between AMI and gamma cyclodextrin (γ-CD) was prepared by kneading method using 1:1 stoichiometry. Solubility of AMI was enhanced by 3.74 times after inclusion complex formation. Amisulpride–γ-cyclodextrin inclusion complex was characterized by FTIR, DSC and XRD techniques. Further sustained release granules of Amisulpride–γ-cyclodextrin inclusion complex (CDSR) were prepared by treating complex with molten stearic acid. Drug release from CDSR granules was sustained up to 12 h with 100 % stearic acid proportion. The integrity of AMI–γ-CD inclusion complex in lipid phase was assessed by XRD study. Finally orodispersible tablets of CDSR granules (OD-CDSR) were prepared using Ac-Di-Sol and microcrystalline cellulose. Disintegration time was assessed by both pharmacopoeial and modified method. Optimized formulation was rapidly disintegrated within 25 s. Thus solubility enhancement and sustained release of AMI was achieved by orodispersion of CDSR granules for improvement of patient compliance.  相似文献

This article presents a software reliability growth model based on non-homogeneous Poisson process. The main focus of this article is to deliver a method for software reliability modelling incorporating the concept of time-dependent fault introduction and fault removal rate with change point. Also in this article, a cost model with change point has been developed. Based on the cost model optimal release policy with change point has been discussed. Maximum likelihood technique has been applied to estimate the parameters of the model. The proposed model has been validated using some real software failure data. Comparison has been made with models incorporating change point and without change point. The application of the proposed cost model has been shown using some numerical examples.  相似文献

The inclusion complexation behaviour of higher chain fatty acid, stearic acid (SA) with gamma cyclodextrin has been investigated. The inclusion complex was characterized by FT-IR, 1H NMR, 2D NMR, XRD and DSC techniques. The results showed that the SA molecule was entrapped inside the gamma cyclodextrin cavity. Further, inclusion complex was treated with lopinavir at 85 °C and emulsified with hot water at 85 °C. The resulted nanoemulsion was cooled down to form solid lipid nanoparticles (SLNs) stabilized with gamma cyclodextrin. Prepared SLNs were having average particle size of 212.5 ± 4.8 nm, zeta potential of ?19.7 ± 0.66 mV and drug loading of 57.54 ± 0.62 %. The surface characteristics of SLNs were also observed with transmission electron microscopy and atomic force microscopy. Results indicate that inclusion complex of SA and gamma cyclodextrin can be used for SLNs preparation.  相似文献

This article addresses the problem of delay-dependent stability for Markovian jumping stochastic systems with interval time-varying delays and nonlinear perturbations. The delay is assumed to be time-varying and belongs to a given interval. By resorting to Lyapunov–Krasovskii functionals and stochastic stability theory, a new delay interval-dependent stability criterion for the system is obtained. It is shown that the addressed problem can be solved if a set of linear matrix inequalities (LMIs) are feasible. Finally, a numerical example is employed to illustrate the effectiveness and less conservativeness of the developed techniques.  相似文献

We consider the slotted ALOHA protocol on a channel with a capture effect. There are M < users each with an infinite buffer. If in a slot, i packets are transmitted, then the probability of a successful reception of a packet is q i. This model contains the CDMA protocols as special cases. We obtain sufficient rate conditions, which are close to necessary for stability of the system, when the arrival streams are stationary ergodic. Under the same rate conditions, for general regenerative arrival streams, we obtain the rates of convergence to stationarity, finiteness of stationary moments and various functional limit theorems. Our arrival streams contain all the traffic models suggested in the recent literature, including the ones which display long range dependence. We also obtain bounds on the stationary moments of waiting times which can be tight under realistic conditions. Finally, we obtain several results on the transient performance of the system, e.g., first time to overflow and the limits of the overflow process. We also extend the above results to the case of a capture channel exhibiting Markov modulated fading. Most of our results and proofs will be shown to hold also for the slotted ALOHA protocol without capture.  相似文献
