The purpose of this study was to synthesize series of methylated chitosaccharide derivatives, possessing various degree of methylation, and to determine their structure activity relationship (SAR) with regard to their antibacterial effect against Staphylococcus aureus. Chitosan polymer and chitooligomers were used as starting materials and were methylated by reaction with methyl iodide. Depending on the reaction conditions the degree of N-quaternization ranged from 0% to 74%, with varying degree of N,N-dimethylation, N-monomethylation and O-methylation. More selective N-quaternization could be obtained with protection group strategy. At pH 5.5 the chitosaccharide polymers and their methylated derivatives were active against S. aureus with minimal inhibitory concentration (MIC) ranging from 16 to 512 μg/mL. At pH 7.2 the non-quaternized derivatives were inactive but their highly N-quaternized derivatives showed MIC as low as 8 μg/mL. The chitooligomers, as well as their derivatives, were inactive at both pH’s. The SAR studies revealed that N-quaternization was mainly responsible for the antibacterial effects at pH 7.2, whereas it did not contribute to the antibacterial activity under acidic conditions.  相似文献

Anisotropic optical absorption properties of single-walled carbon nanotubes (SWNTs) are determined from a vertically aligned SWNT film for 0.5-6 eV. Absorption peaks at 4.5 and 5.25 eV are found to exhibit remarkable polarization dependence and have relevance to optical properties of graphite. A method for determining a nematic order parameter for an aligned SWNT film based on the collinear absorption peak at 4.5 eV is presented, followed by the determination of the optical absorption cross section.  相似文献

The activity of cholesterol 7 alpha-hydroxylase in rat liver microsomes was investigated under conditions favourable for phosphorylation-dephosphorylation. The enzyme activity was similar in the presence or absence of sodium fluoride during preparation. Preincubation with ATP and magnesium did not affect the enzyme activity. Cholesterol 7 alpha-hydroxylase was inhibited by alkaline phosphatase, but this inhibition was similar also after inactivation of the phosphatase. Under similar conditions, rat hepatic hydroxymethylglutaryl CoA reductase activity was clearly modulated in agreement with phosphorylation-dephosphorylation. The absence of such a modulation of cholesterol 7 alpha-hydroxylase argues against involvement of phosphorylation-dephosphorylation in the regulation of this enzyme.  相似文献

Boiling heat transfer measurements on a tube designed to yield the peripheral variation of heat transfer coefficient without interfering with the nucleation site density are presented. A variation of up to 25% around the tube is found with a maximum at the base. High speed cine photography was used to estimate the variation of mean bubble layer thickness and mean velocities with angle. The quantitative composition of white spirit vapours in workplace air may differ considerably from the solvent being used, although all components are the same. By calculating the hygienic effect from the threshold limit value (NGV) of each component, a more reliable estimate is obtained of the occupational hazard than by using the NGV for white spirit. In this method the analyses were performed by on-column injection onto a temperature-programmed capillary column. Retention indices based on n-paraffins and isooctane were calculated using spline functions. Index tables were established for different hydrocarbon mixtures. The validity of the retention indices was found to be satisfactory, depending on the stability of the column and the possibility of optimizing the indices when replacing one column by another of the same type and dimensions. Comparisons were made with alternative methods for determining the concentration of white spirit vapours in air samples. A polar column was used to check the total content of aromatics.  相似文献

The alignment of nanocoils by an electric field was observed in situ inside a focused ion beam microscope. This alignment occurred at relatively low fields, before the onset of electron field emission. The emission behavior of individual nanocoils was also studied using a field emission probe system. The robust coils reached high emission currents, with more than 10 A coming from individual coils at fields stronger than 4 V/m. The emission behavior, however, was not wholly consistent with conventional field emission theory. Analysis of the data showed non-linear Fowler–Nordheim behavior. This phenomenon is usually attributed to thermal effects, but numerical calculations show that cannot be the case. The data plotted linearly on a Schottky plot, which should not be possible due to Schottky temperature dependence and resistive heating during emission. PACS 81.07.-b  相似文献
