There are many methods available for the detection of nucleotide variations in genetic material. Most of these methods are applied after amplification of the target genome sequence by the polymerase chain reaction (PCR). Many efforts are currently underway to develop techniques that can detect single nucleotide variations in genes either by means of, or without the need for, PCR. Allele-specific PCR (asPCR), which reports nucleotide variations based on either the presence or absence of a PCR-amplified DNA product, has the potential to combine target amplification and analysis in one single step. The principle of asPCR is based on the formation of matched or mismatched primer-target complexes by using allele-specific primer probes. PCR amplification by a DNA polymerase from matched 3'-primer termini proceeds, whereas a mismatch should obviate amplification. Given the recent advancements in real-time PCR, this technique should, in principle, allow single nucleotide variations to be detected online. However, this method is hampered by low selectivity, which necessitates tedious and costly manipulations. Recently, we reported that the selectivity of asPCR can be significantly increased through the employment of chemically modified primer probes. Here we report further significant advances in this area. We describe the synthesis of various primer probes that bear polar 4'-C-modified nucleotide residues at their 3' termini, and their evaluation in real-time asPCR. We found that primer probes bearing a 4'-C-methoxymethylene modification have superior properties in the discrimination of single nucleotide variations by PCR. 相似文献

Biaryl‐bridged cyclic peptides comprise an intriguing class of structurally diverse natural products with significant biological activity. Especially noteworthy are the antibiotics arylomycin and its synthetic analogue G0775, which exhibits potent activity against Gram‐negative bacteria. Herein, we present a simple, flexible, and reliable strategy based on activating‐group‐assisted catalytic oxidative coupling for assembling biaryl‐bridged cyclic peptides from natural amino acids. The synthetic approach was utilized for preparing a number of natural and unnatural biaryl‐bridged cyclic peptides, including arylomycin/G0775 and RP 66453 cyclic cores. 相似文献

Effect of surface steps on boundary layer transition 总被引:1,自引:0,他引:1
An experimental study has been carried out to examine the effect of a sharp-edged step on boundary layer transition. The transition position and disturbance spectra in the boundary layer for different step heights and free-stream velocities were measured by hot-wire anemometry. A correlation between the transition Reynolds number and the relative step height has been established for both backward-facing and forward-facing steps. The transition position is associated with the “N-factor” that defines the integrated growth of instability waves at transition. The boundary layer over a step has an earlier transition position than that on a smooth plate, since the instability waves amplify more rapidly than those on a smooth surface. The transition N-factor for the flow containing a step, calculated using the amplification rates on a smooth plate, will, therefore, be smaller than that on surfaces without a step. The observed reduction of the N-factor occurring with a step has been shown to correlate with the height of the step, thus, providing an empirical tool that can be used to estimate the transition position when steps occur. An appropriate value of N can be determined from knowledge of the step height. 相似文献

A new directionally sensitive velocity measurement system based upon a single oscillating hot-wire probe has been developed.
The vibration of the hot-wire was effected by a piezoelectric oscillator that could be driven at different amplitudes and
frequencies. The sign of the local flow velocity was determined by the phase shift between the motion and the output from
the hot-wire.
The system was used to measure mean and fluctuating velocities profiles of the separated flow behind a backward facing step.

Driven by scientific progress and economic stimulus, medical diagnostics will move to a stage in which straightforward medical diagnoses are independent of physician visits and large centralized laboratories. The future of basic diagnostic medicine will lie in the hands of private individuals. We have taken significant strides towards achieving this goal by developing an autoassembly assay for disease biomarker detection which obviates the need for washing steps and is run on a handheld sensing platform. By coupling magnetic nanotechnology with an array of magnetically responsive nanosensors, we demonstrate a rapid, multiplex immunoassay that eliminates the need for trained technicians to run molecular diagnostic tests. Furthermore, the platform is battery-powered and ultraportable, allowing the assay to be run anywhere in the world by any individual. 相似文献

Significant enhancement of both the rate and the chemoselectivity of iron‐catalyzed oxidative coupling of phenols can be achieved in fluorinated solvents, such as 1,1,1,3,3,3‐hexafluoropropan‐2‐ol (HFIP), 2,2,2‐trifluoroethanol (TFE), and 1‐phenyl‐2,2,2‐trifluoroethanol. The generality of this effect was examined for the cross‐coupling of phenols with arenes and polycyclic aromatic hydrocarbons (PAHs) and of phenol with β‐dicarbonyl compounds. The new conditions were utilized in the synthesis of 2′′′‐dehydroxycalodenin B in only four synthetic steps. 相似文献

Efficient and highly selective catalytic conditions for the aerobic autoxidation of methylarenes to benzaldehydes, based on N-hydroxyphthalimide (NHPI) and cobalt(II) acetate in 1,1,1,3,3,3-hexafluoropropan-2-ol (HFIP), were developed. The sustainable conditions enable a multigram scale preparation of benzaldehyde derivatives in high efficiency and with excellent chemoselectivity (up to 99 % conversion and 98 % selectivity). 相似文献
