A two-component reaction–diffusion system is considered. The question of stabilizing to zero one of the components of the solution via an internal control acting on a small subdomain and preserving nonnegativity of both components is investigated. Intracellular signal transduction is often regulated by transient protein phosphorylation in response to external stimuli. Insulin signaling is dependent on specific protein phosphorylation events, and analysis of insulin receptor substrate-1 (IRS-1) phosphorylation reveals a complex interplay between tyrosine, serine, and threonine phosphorylation. The phosphospecific antibody-based quantification approach for analyzing changes in site-specific phosphorylation of IRS-1 is difficult due to the dearth of phospho-antibodies compared with the large number of known IRS-1 phosphorylation sites. We previously published a method detailing a peak area-based mass spectrometry approach, using precursor ions for peptides, to quantify the relative abundance of site-specific phosphorylation in the absence or presence of insulin. We now present an improvement wherein site-specific phosphorylation is quantified by determining the peak area of fragment ions respective to the phospho-site of interest. This provides the advantage of being able to quantify co-eluting isobaric phosphopeptides (differentially phosphorylated versions of the same peptide), allowing for a more comprehensive analysis of protein phosphorylation. Quantifying human IRS-1 phosphorylation sites at Ser303, Ser323, Ser330, Ser348, Ser527, and Ser531 shows that this method is linear (n = 3; r2 = 0.85 ± 0.05, 0.96 ± 0.01, 0.96 ± 0.02, 0.86 ± 0.07, 0.90 ± 0.03, 0.91 ± 0.04, respectively) over an approximate 10-fold range of concentrations and reproducible (n = 4; coefficient of variation = 0.12, 0.14, 0.29, 0.30, 0.12, 0.06, respectively). This application of label-free, fragment ion-based quantification to assess relative phosphorylation changes of specific proteins will prove useful for understanding how various cell stimuli regulate protein function by phosphorylation.  相似文献

Low energy electron diffraction, grazing incidence X-ray diffraction and photoemission were used to decipher the detailed structural arrangement and chemical composition of the surface region of a transition metal carbide, VC0.8(1 1 0). In agreement with previous scanning tunneling microscopy (STM) studies, we find that the surface reconstructs with a ridge-and-valley grating structure along the direction resulting from {0 0 1} faceting for the (3 × 1) and the (4 × 1) phases.Both superstructures terminate on the vacuum side with a nearly stoïchiometric VC region due to C segregation, in contrast with the conclusions drawn from this previous STM study. However, the present experiments clearly show that these phases are metastable, and slow cooling results in a (1 × 1) surface, which is highly C depleted, similarly to the (1 0 0) face.  相似文献

This paper presents a study on the deformation of anisotropic fibrous porous media subjected to moistening by water in the liquid phase. The deformation of the medium is studied by applying the concept of effective stress. Given the structure of the medium, the displacement of the solid matrix is not taken into account with respect to the displacement of the liquid phase. The transport equations are derived from the model proposed by Narasimhan. The transport coefficients and the relation between the variation in apparent density and effective stress are obtained by test measurements. Block copolymers based on polyethylene (PE) and ethylene butadiene rubber (EBR) were obtained by successive controlled coordinative chain transfer polymerization (CCTP) of a mixture of ethylene and butadiene (80/20) and pure ethylene. EBR-b-PE diblock copolymers were synthesized using the {Me2Si(C13H8)2Nd(BH4)2Li(THF)}2 complex in combination with n-butyl,n-octyl magnesium (BOMAG) used as both the alkylating and chain transfer agent (CTA). Triblock and multiblock copolymers featuring highly semi-crystalline PE hard segments and soft EBR segments were further obtained by the development of a bimetallic CTA, the pentanediyl-1,5-di(magnesium bromide) (PDMB). These new block copolymers undergo crystallization-driven organization into lamellar structures and exhibit a variety of mechanical properties, including excellent extensibility and elastic recovery in the case of triblock and multiblock copolymers.  相似文献

This study is dedicated to building and analyzing the spatial spread of Toxoplasma gondii through a heterogeneous predator–prey system. The spatial domain is made of N patches hosting various population species, some of them being prey, others being predators. Predators offer strong heterogeneities with respect to local sustainable resources yielding variable growth rates, from exponential decay to logistic regulation. T. gondii life cycle goes through several stages, starting in the environment where oocysts are released from cat feces, reaching prey within which asexual reproduction yields cysts and then predators wherein sexual reproduction takes place. The resulting model system is complex to handle. We consider some relevant toy models with three patches, two resident predator species and Lotka–Volterra functional responses to predation. We provide the existence and local stability of a persistent stationary state for the underlying predator–prey model systems. The reproduction number R 0 is computed in the quasistationary case; it simplifies when slow–fast dynamics are considered. Numerical experiments illustrate our analysis.  相似文献

We provide existence results for nonnegative solutions to a class of reaction-diffusion systems with cross-diffusion modeling the spread of an epidemic disease. The existence of weak solutions is proved by means of an approximation process, the Faedo-Galerkin method, and a compactness argument. Under additional assumptions a global existence result for classical solutions is derived upon using interpolation results between Banach spaces.  相似文献

The magnetic profile across the interface of a perpendicular exchange coupled [NiO/CoO]3/Pt-Co/Pt(111) system is investigated. The magneto-optic Kerr effect reveals a strong coupling between the antiferromagnetic (AFM) oxide and the ferromagnetic (FM) Pt-Co layer, by an increasing coercivity and a rotation of the easy magnetization axis of the FM layer along the AFM spins. Soft x-ray resonant magnetic reflectivity is used to probe the spatial distribution of the out-of-plane magnetization inside the oxide above its ordering temperature. It extends over 1 nm and exhibits a change of sign.  相似文献
