Far-red emitting fluorescent labels are highly desirable for spectral multiplexing and deep tissue imaging. Here, we describe the generation of frFAST (far-red Fluorescence Activating and absorption Shifting Tag), a 14-kDa monomeric protein that forms a bright far-red fluorescent assembly with (4-hydroxy-3-methoxy-phenyl)allylidene rhodanine (HPAR-3OM). As HPAR-3OM is essentially non-fluorescent in solution and in cells, frFAST can be imaged with high contrast in presence of free HPAR-3OM, which allowed the rapid and efficient imaging of frFAST fusions in live cells, zebrafish embryo/larvae, and chicken embryos. Beyond enabling the genetic encoding of far-red fluorescence, frFAST allowed the design of a far-red chemogenetic reporter of protein–protein interactions, demonstrating its great potential for the design of innovative far-red emitting biosensors.  相似文献

Percent is a standardized ratio comparison that is often used to describe relative amounts of increase and decrease. In this paper, the nonsymmetric relationship between percent of increase and percent of decrease is explored in several ways. Beginning with a simple graphical representation of the relationship, the functional relationship is developed, with relative decrease expressed as a function of relative increase. Once expressed in the form f(x) = 100x/(100 + x), an immediate connection is made to two other topics commonly found within the study of rates—the work problem and the harmonic mean. Finally, successive percents of increase or decrease are shown to be everyday applications of the geometric mean. This deeper look at a common mathematical concept has uncovered rich structural relationships between topics that are often isolated in the curriculum.  相似文献

The ring‐opening metathesis polymerizations (ROMP), using RuCl2 (PCy3)2CHPh, of a series of peptide‐functionalized norbornene derivatives have been investigated. Incorporation of a PEG‐monomer was found to prevent premature precipitation of polymer strands during the course of polymerization reactions and yield water compatible polymers in high conversions. © 2007 Wiley Periodicals, Inc. To develop a greater understanding of interfacial interactions between a semicrystalline polymer and a glassy polymer, adhesion tests were performed on very thin layers of poly(ethylene oxide) (PEO) sandwiched between two layers of poly(tetramethyl bisphenol A polycarbonate) (TMPC). The tests were designed to provide intimate contact between the surfaces while they were heated above the melting point of the PEO and cooled back to room temperature. A contact mechanics approach, based on the Johnson, Kendall, and Roberts theory, was used to determine values of the energy release rate describing the energetic driving force for crack propagation within the interfacial region. The ability to measure crack propagation at large values of the energy release rate was limited by rupture of the silicone elastomer that was used to provide a sufficiently compliant matrix for the adhesion experiment. By cycling the tensile stress at relatively low loading levels, we were able to measure fatigue crack propagation at values of the energy release rate that did not result in failure of the elastomer. © 2004 Wiley Periodicals, Inc. The dimensiond=4 gluon condensateφ 1 is determined from an analysis of charmonium, taking model dependent estimates ofd=6.8 condensates into account. ForS-waves, there is a compatibility region, where both these and model dependent higher state corrections to ground state dominance are small, enablingφ 1 to be determined from a fit to the data. ForP-waves the estimated higher dimension contributions are large in the region of ground state dominance, and noφ 1 determination is possible. Theφ 1 value obtained from the fit to theS-wave ratios is 3 5 times bigger than that obtained by RRY using the plateau method. The methods are compared for the vector current. We show that the higher state correction used by RRY does not fit the data; and that the plateau method is much more sensitive to thed=6.8 condensates than the ratio used here. When reasonable estimates of both these and the higher state corrections are taken into account, there is compatibility between the two methods.  相似文献
