Over the past few years, a large number of studies have been prepared that describe the analysis of peptides and proteins using capillary electrophoresis (CE) and laser-induced fluorescence (LIF). These studies have focused on two general goals: (i) development of automatic, selective and quick separation and detection of mixtures of peptides or proteins; (ii) generation of new methods of quantitation for very low concentrations (nm and subnanomolar) of peptides. These two goals are attained with the use of covalent labelling reactions using a variety of dyes that can be readily excited by the radiation from a commonly available laser or via the use of noncovalent labelling (immunoassay using a labelled antibody or antigen or noncovalent dye interactions). In this review article, we summarize the works which were performed for protein and peptide analysis via CE-LIF.  相似文献

Molecular imprinted polymers (MIP) were prepared by the copolymerization of styrene (S) or methyl methacrylate (MMA) and methacrylic acid (MAA) using ethylene glycol dimethacrylate (EGDMA) as the crosslinker with molar ratios of [monomer]/[crosslinker] and [MAA]/[template] of 3:7 (to obtain a rigid structure) and 1:6 (to optimise hydrogen interactions), respectively. The polymerizations occurred in presence of the template molecule (MIP) - GlcNcouma - an amphiphilic monosaccharide. The same materials, non-imprinted polymers (NIP), were also prepared in absence of the template. These MIPs were characterized and used as SPE supports for selective enrichment. The results showed the correlation between retention efficiency and the porogen character of the polymerization solvent.  相似文献

Amino acids are studied extensively using capillary electrophoresis. In a previous article, we reviewed applications reported in the period 1999-early 2001 (Prata, C., Bonnafous, P., Fraysse, N., Treilhou, M., Poinsot, V., Couderc, F., Electrophoresis 2001, 22, 4129-4138). In this article we follow on with this review for the period end of 2001-beginning of 2003. We will report the developments of detection methods, separations of enantiomers, the new medical applications, and amino acids in food and plants. This review shows that CE is more and more important for the amino acid analysis.  相似文献

We report the minimum amounts of polysaccharides (6.5 ng) that can be studied using capillary electrophoresis-laser-induced fluorescence (CE-LIF) and describe the separation of the various monosaccharides susceptible to occur in Sinorhizobium cell wall. On-gel hydrolysis of the polysaccharides is described. It allowed the easy and rapid determination of the monosaccharide composition of the excised lipopolysaccharides. The composition obtained fits with gas chromatography-mass spectrometry (GC-MS) results. This study underlines the simplicity of using CE-LIF, which remains an entirely "water medium" method, unlike GC-MS which is, in addition, less sensitive (at least 100-fold) than CE-LIF.  相似文献

We report the first observation of a weak ferromagnetic state of Cr in Cr/Ru(0001) superlattices, based on magnetic hysteresis and corroborated by x-ray magnetic circular dichroism at the CrL(2,3) edges. In situ reflection high-energy electron diffraction, x-ray diffraction, and Cr K-edge polarized x-ray absorption investigations have shown that the Cr layers thinner than 8 angstroms adopt a slightly distorted hcp structure, accompanied by a large atomic volume expansion of up to 14% compared to the bcc packing volume. The expanded hcp structure clearly induces the observed ferromagnetism, in agreement with theory.  相似文献

Increasingly stringent regulations and the need to tackle rising fuel prices have placed great emphasis on the design of aeronautical gas turbines, which are unfortunately more and more prone to combustion instabilities. In the particular field of annular combustion chambers, these instabilities often take the form of azimuthal modes. To predict these modes, one must compute the full combustion chamber, which remained out of reach until very recently and the development of massively parallel computers. In this article, full annular Large Eddy Simulations (LES) of two helicopter combustors, which differ only on the swirlers' design, are performed. In both computations, LES captures self-established rotating azimuthal modes. However, the two cases exhibit different thermo-acoustic responses and the resulting limit-cycles are different. With the first design, a self-excited strong instability develops, leading to pulsating flames and local flashback. In the second case, the flames are much less affected by the azimuthal mode and remain stable, allowing an acceptable operation. This work describes numerical simulations of a hot jet in cross‐flow with applications to anti‐ice systems of aircraft engine nacelles. Numerical results are compared with experimental measurements obtained at ONERA to evaluate the performances of LES in this industrial context. The combination of complex geometries requiring unstructured meshes and high Reynolds number does not allow the resolution of boundary layers so that wall models must be employed. In this framework, the relative influence of subgrid‐scale modelling and conjugate heat transfer in LESs of aerothermal flows is evaluated. After a general overview of the transverse jet simulation results, a LES coupled with a heat transfer solver in the walls is used to show that thermal boundary conditions at the wall have more influence on the results than subgrid scale models. Coupling fluid flow and heat transfer in solids simulations is the only method to specify their respective thermal boundary conditions. Copyright © 2011 John Wiley & Sons, Ltd.  相似文献

Characteristic formulations for boundary conditions have demonstrated their effectiveness to handle inlets and outlets, especially to avoid acoustic wave reflections. At walls, however, most authors use simple Dirichlet or Neumann boundary conditions, where the normal velocity (or pressure gradient) is set to zero. This paper demonstrates that there are significant differences between characteristic and Dirichlet methods at a wall and that simulations are more stable when using walls modelled with a characteristic wave decomposition. The derivation of characteristic methods yields an additional boundary term in the continuity equation, which explains their increased stability. This term also allows to handle the two acoustic waves going towards and away from the wall in a consistent manner. Those observations are confirmed by stability matrix analysis and one‐ and two‐dimensional simulations of acoustic modes in cavities. Copyright © 2009 John Wiley & Sons, Ltd.  相似文献

This paper describes a compressible Large Eddy Simulation (LES) used to investigate cyclic variations for nonreacting flow in an optical single cylinder engine setup. The simulated operating point is part of a large experimental database designed to validate LES for cycle-to-cycle prediction, and constitutes a first step towards the realization of fired operating points. The computational domain covers almost the whole experimental setup (intake and exhaust plenums, intake and exhaust ducts, cylinder) to account for acoustic phenomena. The assessment of the computation is performed in two regions of the domain: the intake and exhaust duct predictions are compared to the results of a Helmholtz solver and the experiment (pressure transducers and Particle Image Velocimetry (PIV)) while the in-cylinder dynamics are compared to PIV measurements. The ability of the developed methodology to capture the correct level of cycle-to-cycle variations is demonstrated considering in-cylinder pressure and velocity fields predictions. Cycle-to-cycle variations in velocity are highlighted and localized using a proper orthogonal decomposition analysis.  相似文献
