In vitro production of sperm is a desirable idea for fertility preservation in azoospermic men and prepubertal boys suffering from cancer. In this study, a biocompatible porous scaffold based on a triad mixture of silk fibroin (SF), alginate (Alg), and laminin (LM) is developed to facilitate the differentiation of mouse spermatogonia stem cells (SSCs). Following SF extraction, the content is analyzed by SDS-PAGE and stable porous 3D scaffolds are successfully prepared by merely Alg, SF, and a combination of Alg-SF, or Alg-SF-LM through freeze-drying. Then, the biomimetic scaffolds are characterized regarding the structural and biological properties, water absorption capacity, biocompatibility, biodegradability, and mechanical behavior. Neonatal mice testicular cells are seeded on three-dimensional scaffolds and their differentiation efficiency is evaluated using real-time PCR, flow cytometry, immunohistochemistry. Blend matrices showed uniform porous microstructures with interconnected networks, which maintained long-term stability and mechanical properties better than homogenous structures. Molecular analysis of the cells after 21 days of culture showed that the expression of differentiation-related proteins in cells that are developed in composite scaffolds is significantly higher than in other groups. The application of a composite system can lead to the differentiation of SSCs, paving the way for a novel infertility treatment landscape in the future.  相似文献

A carbon past electrode modified with [Mn(H2O)(N3)(NO3)(pyterpy)], ( \textpyterpy = 4¢- ( 4 - \textpyridyl ) - 2,2¢:\text6¢,\text2¢¢- \textterpyridine ) \left( {{\text{pyterpy}} = 4\prime - \left( {4 - {\text{pyridyl}}} \right) - 2,2\prime:{\text{6}}\prime,{\text{2}}\prime\prime - {\text{terpyridine}}} \right) complex have been applied to the electrocatalytic oxidation of nitrite which reduced the overpotential by about 120 mV with obviously increasing the current response. Relative standard deviations for nitrite determination was less than 2.0%, and nitrite can be determined in the ranges of 5.00 × 10−6 to 1.55 × 10−2 mol L−1, with a detection limit of 8 × 10−7 mol L−1. The treatment of the voltammetric data showed that it is a pure diffusion-controlled reaction, which involves one electron in the rate-determining step. The rate constant k′, transfer coefficient α for the catalytic reaction, and diffusion coefficient of nitrite in the solution, D, were found to be 1.4 × 10−2, 0.56× 10−6, and 7.99 × 10−6 cm2 s−1, respectively. The mechanism for the interaction of nitrite with the Mn(II) complex modified carbon past electrode is proposed. This work provides a simple and easy approach to detection of nitrite ion. The modified electrode indicated reproducible behavior, anti-fouling properties, and stability during electrochemical experiments, making it particularly suitable for the analytical purposes.  相似文献

The electrocatalytic oxidation of methanol studied at the surface of nickel disc electrode coated with N,N-bis(salicylidene)phenylenediamine (Salophen) legend in 0.10 M NaOH solution using cyclic voltammetry. The results showed that the Ni-salophen layer formed at the surface of electrode behaves as an efficient electrocatalyst for the oxidation of methanol in alkaline medium. The thermal analysis of Ni-salophen complex studied at temperature ranges 0–800°C in argon atmosphere. Also, FTIR was employed to prove the formation of complex on the modified electrode surface. Moreover, the effects of various parameters such as oven temperature, methanol concentration, NaOH concentration and media temperature on electro-oxidation of methanol were investigated. The kinetic parameters such as activation energy and Tafel slope were calculated for methanol oxidation on the modified electrode surface. Nitride semiconductors and their alloys recently have versatile applications as high-power and high-efficiency electro optical devices duo to their high thermal stability, direct transition and wide bang-gap. Nanostructure light emitting diodes of these materials have an emission spectrum from infrared to ultraviolet. In single-layer graphene sheets non-local interband exchange leads to a renormalized Fermi-surface effective mass which vanishes in the low carrier-density limit. We report on a comparative study of Fermi surface effective mass renormalization in single-layer and AB-stacked bilayer graphene. We explain why the mass does not approach zero in the bilayer case, although its value is still strongly suppressed.  相似文献

Cyclocarbopalladation/cross-coupling cascade intramolecular Heck–Suzuki–Miyaura reactions is applied for the first time by palladium immobilized on pyridine-imidazolium ionic liquid supported magnetic iron oxide nanoparticle catalyst (denoted Pd@Py-IL-SPION) for the last step to synthesize trisubstituted arylidene–isoquinolinones derivatives having Combretastatin skeleton. The reaction is performed via propargylamide intermediates prepared by Ugi 4-CR reactions, which undergoes intramolecular Heck–Suzuki–Miyaura domino reaction to produce the desired trisubstituted arylidene-isoquinolinones. The method shows full regio- and stereoselectivity derives from the particular Pd-catalyzed syn-insertion of triple bond.  相似文献

Electrocatalytic oxidation of methanol on a glassy carbon electrode coated with Ni(II)-(1,2-phenylendiamine)2 (GC/NiOPD), conditioned by the potential recycling in a potential range of 100–650 mV (vs. SCE) is studied by cyclic voltammetry in an alkaline medium (0.10 M NaOH). The results show that the NiOPD layer formed at the surface of the electrode behaves as an efficient electrocatalyst for the oxidation of methanol in the alkaline medium via the Ni(III) species with a cross exchange reaction occurring throughout the layer at a low concentration of methanol. The effects of various parameters such as potential scan rates, methanol concentration and NiOPD surface concentration on the electro-oxidation of methanol are also investigated.  相似文献

In this study, anti-reflection (AR) \(\hbox {SiO}_{2}/ \hbox {ZrO}_{2}\) thin films with 3-layers were designed and fabricated by the essential Macleod software and physical vapor deposition, respectively. In order to improve the optical and physical properties of the prepared samples, laser shock peening (LSP) technique was applied. For this purpose, an Argon Fluoride Excimer laser \((\lambda =193 \,\text {nm})\) with 110 and 240 mJ energies and 1 Hz frequency at different pulses was used. The effect of LSP method in improving transmissions and laser damage thresholds of the prepared samples was proved by using UV–Vis–IR spectroscopy in the wavelength range of 400–1200 nm and international standard ISO11254 at 1064 nm. In addition, scanning electron microscopy was used to check the effect of applying LSP.  相似文献
