DNA-nanotube artificial ion channels   总被引:4,自引:0,他引:4  
There is considerable interest in developing chemical devices that mimic the function of biological ion channels. We recently described such a device, which consisted of a single conically shaped gold nanotube embedded within a polymeric membrane. This device mimicked one of the key functions of voltage-gated ion channels: the ability to strongly rectify the ionic current flowing through it. The data obtained were interpreted using a simple electrostatic model. While the details are still being debated, it is clear that ion-current-rectification in biological ion channels is more complicated and involves physical movement of an ionically charged portion of the channel in response to a change in the transmembrane potential. We report here artificial ion channels that rectify the ion current flowing through them via this "electromechanical" mechanism. These artificial channels are also based on conical gold nanotubes, but with the critical electromechanical response provided by single-stranded DNA molecules attached to the nanotube walls.  相似文献

Protein biosensors based on biofunctionalized conical gold nanotubes   总被引:1,自引:0,他引:1  
There is increasing interest in the concept of using nanopores as the sensing elements in biosensors. The nanopore most often used is the alpha-hemolysin protein channel, and the sensor consists of a single channel embedded within a lipid bilayer membrane. An ionic current is passed through the channel, and analyte species are detected as transient blocks in this current associated with translocation of the analyte through the channel-stochastic sensing. While this is an extremely promising sensing paradigm, it would be advantageous to eliminate the very fragile lipid bilayer membrane and perhaps to replace the biological nanopore with an abiotic equivalent. We describe here a new family of protein biosensors that are based on conically shaped gold nanotubes embedded within a mechanical and chemically robust polymeric membrane. While these sensors also function by passing an ion current through the nanotube, the sensing paradigm is different from the previous devices in that a transient change in the current is not observed. Instead, the protein analyte binds to a biochemical molecular-recognition agent at the mouth of the conical nanotube, resulting in complete blockage of the ion current. Three different molecular-recognition agents, and correspondingly three different protein analytes, were investigated: (i) biotin/streptavidin, (ii) protein-G/immunoglobulin, and (iii) an antibody to the protein ricin with ricin as the analyte.  相似文献

A recently suggested scheme for relating ionisation potentials (IP) to excitation energies (E) has been applied to pyrazine and 2,6-dimethylpyrazine. The results indicate that in these systems the energy gap (ΔE) between the two lowest lying 1n,π*-states is significantly smaller than that (ΔIP) between the corresponding 2n-states of the parent radical cations. The values estimated for ΔE on the basis of ΔIP, measurable by photoelectron spectroscopy, disagree with those calculated theoretically but seem to be supported by the experimental evidence available. Since ΔE = ΔIP would normally be expected, the relationship ΔE < ΔIP actually present in these systems is discussed and related to the different shape of the n-MO's involved in the ionization and excitation processes.  相似文献

The electronic absorption spectra of 22 arylferrocenes and their arylferricenium salts with pentacyanopropenide (PCNP) were recorded. The attemptedHammett correlation of max of the d-d transition bands of arylferrocenes revealed that this band is substituent dependent only for strong electron-withdrawing substituents. Excellent correlation of max of the d-d transition bands with + constants was found in the case of arylferricenium cations. Good correlations of max was also found with theE 1/2 oxidation potentials measured by cyclic voltametry.

Zusammenfassung Elektronenabsorptions-Spektren von 22 Arylferrocenen und ihren Arylferricenium-Pentacyanopropenid-Salzen wurden gemessen. VersuchteHammett-Korrelationen des längstwelligen d-d-Überganges (max) zeigten, daß diese Bande nur von stark elektronenanziehende Substituenten abhängig ist. Im Fall der Arylferricenium-Kationen wurde eine ausgezeichnete Korrelation zwischen max der d-d-Bande und den +-Konstanten festgestellt. Eine gute Korrelation der max-Werte wurde auch mit denE 1/2 Oxidationspotentialen gefunden, die durch cyclische Voltametrie meßbar sind.

A hybrid PDMS/glass microfluidic system for evaluation of the efficiency of photodynamic therapy is presented. 5-aminolevulinic acid (ALA) was used as a precursor of photosensitizer. The geometry of the microdevice presented in this paper enables to test different concentrations of the photosensitizer in a single assay. The viability of the A549 cells was determined 24 h after PDT procedure (irradiation with light which induced a photosensitizer accumulated in carcinoma cells, λ = 625 nm). The presented results confirmed the possibility to perform the photodynamic therapy process in vitro in microscale and the possibility to assess its effectiveness. Moreover, because two identical microstructures on a single chip were performed, the microchip can be used for examination simultaneously various cell lines (carcinoma and normal) or various photosensitizers.  相似文献

The thermal decomposition of the complexes Zn(form)2⋅2phen (I), Zn(ac)2⋅2phen (II), Zn(prop)2⋅2phen (III), Zn(but)2⋅2phen (IV), where phen=phenazone, form=formiate, ac=acetate, prop=propionate, but=butyrate has been studied in air by TG/DTG and DTA methods. The possible mechanism of the thermal decomposition was proposed. The final product of thermal decomposition was ZnO. IR data show unidentate coordination of carboxylate group to Zn(II) ion. The complexes were tested against various strains of microorganisms and their efficiency decrease in the sequence yeasts >bacteria>filamentous fungi.  相似文献

(Liquid + liquid) phase equilibria (LLE) of binary mixtures containing hyperbranched polymer Boltorn® H2004 and n-alkanes (n-hexane, n-heptane, n-octane, and n-decane) were studied over the temperature range from about (260 up to 360) K. The polymer is partially miscible with n-alkanes and the solubility decreases with an increase of the chain length of the solvent. Corresponding LLE phase diagrams including spinodal and binodal (liquid + liquid) coexistence curves were calculated in terms of the statistical mechanics – based on the lattice-cluster theory, based only on the upper critical solution temperature, and the polymer chain architecture. The results show semi-qualitative agreement of predicted and experimental equilibrium compositions and temperatures. Boltorn® H2004 reveals complete miscibility in the liquid phase with alcohols (C1–C8), aromatic hydrocarbons (benzene, toluene, and thiophene), and ethers (methyl tetra-butyl ether, ethyl tetra-butyl ether, and tetrahydrofurane).  相似文献

This work reports on the solvatochromic properties of a simple heterocyclic betaine pyridinium, 2-(1-pyridinio)benzimidazolate (SBPa), having promising potentialities in non-linear optics. From advanced PCM-TDDFT calculations, the solvatochromism of SBPa was found to be unusual, involving two different electronic states for absorption (S(0)→ S(2)) and emission (S(1)→S'(0)). To account for this behavior, we developed an innovative physical treatment which consists in a non-linear fit of the solvatochromic data using the Bilot-Kawski theoretical model and visualizing the least-square coefficient χ(2) on a 2D map as a function of the solute polarizability and gas phase absorption energy. In parallel, Kamlet-Taft correlations were undertaken to select a propitious set of electrostatic solvents usable in this treatment. Protic solvents that lead to specific interactions and nonpolar solvents that favor dimerization processes were excluded. From a choice of aprotic solvents with sufficiently high polarity, 4 dipole moments μ(g)(S(0)) = +9.1 D, μ(e)(S(2)) = -1.5 D, μ(e)(S(1)) = 0 D and μ(g)(S'(0)) = +3.31 D were determined, the 3 former values being in close agreement with TDDFT values, although the solute polarizability values seem underestimated. Anyhow, disregarding this discrepancy, we evaluated the static hyperpolarizability to β(0) = -64 × 10(-30) esu from the solvatochromic data in close agreement with DFT calculations.  相似文献

Despite many advances in therapy, glioblastoma (GB) is still characterized by its poor prognosis. The main reason for this is unsuccessful treatment, which slightly extends the duration of remission; thus, new regimens are needed. One of many types of chemotherapeutics that are being investigated in this field is topoisomerase inhibitors, mainly in combination therapy with other drugs. On the other hand, the search for new anti-cancer substances continues. Neobavaisoflavone (NBIF) is a natural compound isolated from Psoralea corylifolia L., which possesses anti-oxidant, anti-inflammatory, and anti-cancer properties. The aim of this study was to evaluate the effect of NBIF in human U-87 MG glioblastoma cells in comparison to normal human NHA astrocytes, and to examine if it influences the activity of irinotecan, etoposide, and doxorubicin in this in vitro model. We demonstrated that NBIF decreases U-87 MG cells viability in a dose-dependent manner. Furthermore, we found that it inhibits cell growth and causes glutathione (GSH) depletion more intensely in U-87 MG cells than in astrocytes. This study also provides, for the first time, evidence of the potentialization of the doxorubicin effect by NBIF, which was shown by the reduction in the viability in U-87 MG cells.  相似文献

Fe(II)/2-oxoglutarate dependent dioxygenases (ODDs) share a double stranded beta helix (DSBH) fold and utilise a common reactive intermediate, ferryl species, to catalyse oxidative transformations of substrates. Despite the structural similarities, ODDs accept a variety of substrates and facilitate a wide range of reactions, that is hydroxylations, desaturations, (oxa)cyclisations and ring rearrangements. In this review we present and discuss the factors contributing to the observed (regio)selectivities of ODDs. They span from inherent properties of the reactants, that is, substrate molecule and iron cofactor, to the interactions between the substrate and the enzyme's binding cavity; the latter can counterbalance the effect of the former. Based on results of both experimental and computational studies dedicated to ODDs, we also line out the properties of the reactants which promote reaction outcomes other than the “default” hydroxylation. It turns out that the reaction selectivity depends on a delicate balance of interactions between the components of the investigated system.  相似文献
