The determination of trace elements in crude oil is difficult due to the complex nature of the sample and the various different chemical forms in which the metals can occur. The advantage of graphite furnace atomic absorption spectrometry is that only a minimum of sample pretreatment is required. In this work two techniques have been compared to establish a fast and reliable method for lead determination in crude oil. In the first one the crude oil samples were weighed directly onto solid sampling (SS) platforms and introduced into the graphite tube for analysis. In the second one the samples were prepared as oil-in-water emulsions and analyzed in a filter furnace (FF). Twenty μL of a mixture of 0.5 mg L− 1 Pd + 0.3 mg L− 1 Mg + Triton X-100 has been used as the modifier, and calibration against aqueous solutions has been used for both methods. The sensitivity obtained with the FF was more than a factor of two better than that with SS; however, as a larger sample mass could be introduced in the latter case, so that the limits of detection for both techniques were 0.004 mg kg− 1. Seven crude oil samples were analyzed using the two procedures, and all results were in agreement at a 95% confidence level using a paired Student's t-test. For validation purposes, three crude oil samples have been mineralized using an open-vessel acid digestion, and the results were in agreement with those found with direct sampling and with emulsion sampling using FF according to ANOVA test. Both methods are simple, fast and reliable, being appropriated for routine analysis; however, the direct method using SS technology should be preferred because of its simplicity, speed and commercial availability.  相似文献

The impact of pollutants in an organism can be observed by changes in functional complexity at different levels. Bivalve gills are suitable for histopathological analysis because of their structure and function. This study aimed at examining the morphology of Mytella falcata gill filaments from three sites in the Santos estuary (São Paulo, Brazil) with different levels of environmental degradation to identify possible changes in gill structure and discuss the significance of these alterations. For this purpose, histological, histochemical and ultrastructural techniques were used. The filaments of animals from site A (less impacted site) were intact, while in sites B and C, pathological changes were observed, such as: detachment of the epithelium in the intermediate zone, morphological changes of this epithelium, inflammatory process, increase in the number of mucous cells and cell turnover processes. These results suggest that the related changes are an attempt to prevent the entrance of pollutants through gill filaments into the entire organism and that cell turnover is the final way to compensate cell injury.  相似文献

The quantification of trace elements in used lubricating oil is useful for evaluating the wearing of specific components of engines. In the present work, inductively coupled plasma optical emission spectrometry (ICPOES) was used for determining five refractory elements (Ni, Mo, Cr, V and Ti) in lubricating-oil samples. The methodology was developed while aiming at the introduction of such organic samples into the ICP as emulsions. Several nebulization systems were tested with clear advantage for Meinhard K3 coupled with a cyclonic spray chamber. The carbon deposition on the injector tip as well as the plasma background was minimized through careful optimization of the Ar and O2 gas mixture flows into the plasma. The optimization of instrumental and experimental parameters allowed quantification using calibration curves prepared with analyte inorganic standards. An internal standard (Sc) was used to correct the matrix effects and signal fluctuations. The limits of detection (3Sb/m), in the ng g(-1) range were obtained for all five elements. The methodology was validated through an analysis of standard reference materials (SRM 1084a, 1085a and 1085b). Good analyte recoveries (from 92.6 to 104.7%) were achieved. Comparison studies against established ICPOES methodologies (sample acid decomposition or sample direct dilution in an organic solvent) have shown that the proposed methodology present clear advantages in terms of simplicity of sample preparation, overall analysis time, and the use of inorganic standards for calibration instead of expensive metallorganic standards.  相似文献

A versatile preconcentration system for trace element determination by ICPMS was developed. It is composed of a commercial flow injection analysis system (FIAS) retrofitted with a home-made control unit containing three solenoid valves and working concomitantly with the FIAS, permitting selection and segmentation of sample, reagent, washing solution and elution flow. The knotted reactor used had a length of 200 cm and was made from 0.05 cm i.d. PTFE tubing. The method applies ammonium 1-pyrrolidinedithiocarbamate (APDC) as complexant, allowing the preconcentration and quantitative multi-element determination of Cu, Ni, V, Co, Nb, Mo, In, Sb and Bi. Ethanol and 4-methyl-2-pentanone were tested as eluents. Recovery tests using complex matrices and spike concentrations of 200 ng L(-1) showed typical values in the range of 90% to 110%. Relative standard deviations were < 7% for elution with ethanol and < 5% with methyl isobutyl ketone. For simulated freshwater samples using 4-methyl-2-pentanone as an eluent, a sample loading rate of 5.0 ml min(-1), and a preconcentration time of 60 s, detection limits (ng L(-1)) were in the range of 0.02 (Bi) to 30 (Cu). Under these conditions, analytical frequency was about 15 samples per hour. The feasibility of the method was demonstrated by the succesful analysis of wastewater and seawater certified reference materials.  相似文献

A rapid, selective, and low-cost chiral capillary electrophoretic method was developed for the simultaneous analysis of hydroxychloroquine (HCQ) and its three chiral metabolites: desethylchloroquine (DCQ), desethylhydroxychloroquine (DHCQ), and bisdesethylchloroquine (BDCQ) in the microsomal fraction of liver homogenates. After liquid-liquid extraction using toluene as extracting solvent, the drug and metabolites were resolved on a fused-silica capillary (50 microm ID, 50 cm total length, and 42 cm effective length), using 100 mmol/L of Tris/phosphate buffer, pH 9.0 containing 1% w/v sulfated-beta-CD and 30 mg/mL hydroxypropyl-beta-CD. The mature testicular vesicles of the species Pseudonannolene tocaiensis have three portions: a peripheral portion; a central portion that is typically secretory and between these two an intermediary portion, a lumen filled with spermatozoa. Both secretory and peripheral portions present the same type of cells. The ultrastructural analysis of the peripheral portion suggests it is involved in hormone synthesis. The secretory portion probably contributes to the production of spermatic fluid, compensating for the absence of male accessory glands in this species.  相似文献

We have previously reported that the palladium-catalyzed cross-coupling reaction of air-stable potassium alkenyltrifluoroborates with aryl halides and triflates proceeds readily with good yields. Recent progress in outlining the scope and limitations of such reactions is described herein. The palladium-catalyzed cross-coupling reaction of potassium alkenyltrifluoroborates with aryl and heteroaryl halides and triflates proceeds readily with moderate to excellent yields. The alkenyl cross-coupling reaction can generally be effected using 2 mol % of PdCl2(dppf).CH2Cl2 as catalyst in i-PrOH-H2O in the presence of t-BuNH2 as the base. A variety of functional groups are tolerated in both partners, and the process is stereospecific with regard to the alkenyltrifluoroborate starting material.  相似文献

Polymerization‐induced phase separation from an all‐monomeric system by direct copolymerization offers the formation of heterogeneous polymeric structures without reliance on polymer blends, block copolymers, or interpenetrating polymer networks. This study examines the potential for the formation of compositional heterogeneity in copolymer networks obtained by free‐radical photopolymerizations of initially homogeneous mixtures of bisphenol A glycidyl dimethacrylate and isodecyl methacrylate as the comonomer ratios and polymerization conditions are varied. Comonomer proportions that control thermodynamic stability prior to (as determined by cloud point measurements) and during [as determined by turbidity measurements coupled with near‐infrared (IR) spectroscopy] polymerization were shown to be a more influential factor on phase separation than irradiance‐imposed kinetic control of the photopolymerization process. Through photorheometry coupled with near‐IR and ultraviolet–visible (UV–Vis), the onset of phase separation was shown to occur at very low conversions and always prior to gelation (as estimated by the crossover of G′/G″). © 2014 Wiley Periodicals, Inc. Fish gills are constituted by primary filaments and secondary lamellae, which are formed, basically, by three different cell types: pillar cells, respiratory cells and erythrocytes that circulate in the lamellae interior. This respiratory organ is very appreciated in environmental studies since it is constantly in contact with the water, accomplishing exchanges; besides this, it presents a high adaptation capacity. In this work, some fishes were reared in polluted waters treated by the flotation method (treated group) and compared with others that were reared in clean waters (control group). The ultramorphological, histological and histochemical analyses were used. The fishes from the treated group presented morphological alterations when compared with the control groups, such as lamellar fusion, aneurism and detachment of the respiratory epithelium. These alterations demonstrate the several adaptations, as morphologic as physiologic, that fishes can present when exposed to an inappropriate environment. The techniques employed are good tools to detect morphological biomarkers in gills of Oreochromis niloticus submitted to polluted waters.  相似文献
