The interaction between cucurbit[8]uril(Q[8]) and oroxin B(ORB) was investigated by UV-visible(UV-Vis) spectroscopy, isothermal titration calorimetry(ITC), mass spectrum(MS) and nuclear magnetic resonance(NMR) spectroscopy. The results showed that ORB formed a 2:1 inclusion complex with Q[8] with a binding constant of 8.266×105 L2·mol-2. ORB had good scavenging ability for 2,2'-azinobis-(3-ethylbenzthiazoline-6-sulphonate)(ABTS) free radicals(IC50=5.74 μmol/L) and the addition of Q[8] did not significantly affect the antioxidant activity of ORB(IC50=5.76 μmol/L). A phase solubility experiment revealed a 1.86-fold increase in the solubility of ORB when c(Q[8])=100 μmol/L. In vitro drug release experiments showed that the release rate for ORB@Q[8] complex was lower than that of ORB in artificial intestinal juice, and higher than that of ORB in artificial gastric juice.  相似文献

A sensitive assay for determining SYUIQ-F5, a novel telomerase inhibitor and anti-tumor drug, in rat tissues and plasma was developed and validated by using liquid chromatography/tandem mass spectrometry (LC?CMS?CMS). After a single step liquid?Cliquid extraction with ethyl acetate-dichloromethane, SYUIQ-F5 and SUCL (internal standard) were subjected to LC?CMS?CMS analysis using positive electro-spray ionization under selected reaction monitoring mode. Chromatographic separation of SYUIQ-F5 and SUCL was achieved on a Zorbax Eclipse Plus C18 column (I.D. 4.6 mm × 150 mm, 3.5 ??m) with a mobile phase consisting of acetonitrile-2 mM ammonium formate (90:10, v/v) at a flow rate of 0.6 mL min?1. The intra- and inter-batch precision of the method were <12.2 and 8.7%, respectively. The intra- and inter-batch accuracies ranged from 100.2 to 107.3%. The lowest limit of quantification for SYUIQ-F5 was 0.5 ng mL?1. The method was applied to a SYUIQ-F5 tissue distribution study after an oral dose of 30 mg kg?1 to rats. SYUIQ-F5 tissue concentrations decreased in the order of small intestine> liver> lung> spleen> stomach> kidneys> heart> brain> muscle> fat> testes> plasma. SYUIQ-F5 could still be detected in most of the tissues at 48-h post-dosing. These results indicated that the LC?CMS?CMS method was sensitive, reliable, and specific to quantify SYUIQ-F5 in different rat tissues.  相似文献

利用254 nm紫外光照射山茱萸水提液在室温下制备了纳米银,并通过UV-Vis光谱检测其在410 nm附近的等离子体共振峰;研究了溶液p H值、料液比以及反应时间对还原反应的影响,确定了纳米银的最优合成条件:p H=7. 0,料液比1∶1,反应时间1 h.通过X射线晶体衍射、透射电子显微镜和激光粒度仪对纳米银的晶体结构、粒径、表面性质和形貌等进行表征发现,在最优反应条件下制得的纳米银为面心立方结构,呈近球形,平均粒径(55. 4±0. 9) nm,分散均匀,表面带负电(-10. 2 m V),具有较高的稳定性.生物活性研究结果表明,制得的纳米银具有良好的抗氧化、抗菌及抗癌活性.当纳米银浓度为62. 5μg/m L时,对DPPH自由基的清除率为70. 0%;对S. aureus和E. coli最低抑菌浓度分别为3. 9和7. 8μg/m L;对结直肠癌细胞HCT116和SW620的IC50值分别为23. 1和35. 1μg/m L.  相似文献

[reaction: see text] UDP-D-galactofuranose (2), which is essential for both cell growth and virulence in many pathogenic microorganisms, is converted from UDP-D-galactopyranose (UDP-Galp, 1) by the flavin adenine dinucleotide (FAD)-dependent enzyme UDP-galactopyranose mutase (UGM). Here, we report the synthesis of UDP-GalOH (13) and show it as an inhibitor for UGM with a binding affinity similar to that of 1. Novel core-shell nanoparticles consisting of poly(methyl methacrylate) (PMMA) cores coated with synthetic polymer and biopolymer (polyethyleneimine, chitosan, and casein) shells were synthesized via direct graft copolymerization of methyl methacry late from hydrophilic polymers in the absence of surfactant. Average hydrodynamic diameters of the nanoparticles ranged from 163 to 263 nm. High-capacity (up to 530 mg/g) immobilizations of enzymes and high-activity retained percentage (E spe) (up to 90%) were achieved.  相似文献

本文提出了一种适于野外条件下的富集天然水中多个痕量元素的方法。研究了四种螫合剂在活性碳上的吸附特性及多种溶剂、酸、碱洗脱螯合剂的特性。研究了20多个元素在不同pH值下的螯合剂——活性碳的静态和动态吸附性能。采用直径10mm,长80mm柱可在现场富集天然水中不同形态的痕量元素。加入回收率为90~108%。天然水中大量的Na、K、Ca、Mg等元素不被吸附。可用多种分析方法测定。  相似文献
