用硅烷化偶联剂把DNA直接共价固定在载玻片表面,将辣根过氧化物酶标记的探针与之进行核酸杂交,杂交后用增强的化学发光检测。方法的检出限为75pg。研究了DNA分子固定在玻璃载体表面的各种条件,并建立了在玻璃载体表面进行核酸杂交的体系,为研究光纤DNA生物传感器打下了基础。 相似文献

以辣根过氧化物酶(HRP)和吖啶酯(AE)为化学发光标记试剂分别标记神经生长因子(NGF)单克隆抗体,经分离纯化制成标记抗体(HRP-Ab,AE-Ab),采用化学发光免疫分析法(CLIA)对NGF进行检测,其检出限为0.5ng/mL,线性范围为2~128ng/mL.10例样本分别用CLIA和RIA进行检测,其结果无显著性差异. 相似文献

CdS nanoparticles were formed on the surface of silica microspheres by the improved layer‐by‐layer self‐assembled technique. High‐resolution electron microscope (HRTEM) image and energy dispersive x‐ray analysis (EDX) confirmed formation of a quasi‐continuous CdS nanoparticles film on the silica microspheres. The results of UV‐vis and fluorescence spectra display that the spherical silica surface has a great effect on the photoluminescence of the loaded CdS nanoparticles. In contrast to the CdS nanoparticles powder, the composite can exhibit the emission ascribed to the band gap transition when the CdS nanoparticles film is relatively thick. This phenomenon is probably due to an enhancement of the crystallinity of CdS nanoparticles induced by the silica spheres. 相似文献

In this study, we developed a simple and selective spin column extraction technology utilizing hydrophilic molecularly imprinted polymers as the sorbents for extracting nitrophenol pollutants in water samples (the East Lake, the Yangtze River, and wastewater). The whole procedure was achieved by centrifugation of the spin column, and multiple samples were simultaneously processed with a low volume of solvent and without evaporation. Under the optimized condition, recoveries of nitrophenol compounds on the spin column packed with hydrophilic molecularly imprinted polymers ranged from 87.3 to 92.9% and an excellent purification effect was obtained. Compared with activated carbon, multi‐walled carbon nanotubes, LC‐C18 sorbents, hydrophilic molecularly imprinted polymers exhibited a highly selective recognition ability for nitrophenol compounds and satisfactory sample extraction efficiency. Subsequently, the spin column extraction coupled with high‐performance liquid chromatography was established, which was found to be linear in the range of 2–1000 ng/mL for 2,4‐dinitropehnol and 2‐nitrophenol, and 6–1000 ng/mL for 4‐nitrophenol with correlation coefficients greater than 0.998. The detection limits ranged from 0.3–0.5 ng/mL. It is shown that the proposed method can be used for the determination of trace nitrophenol pollutants in complex samples, which is not only beneficial for water quality analysis but also for environmental risk assessment. 相似文献

An efficient and effective microwave-assisted Heck cross-coupling of different terminal olefins with various 3-iodo-benzopyrones including sterically hindered, electron-rich, electron-neutral, and electron-deficient is developed. It proceeded faster and generally gave good to excellent yields under microwave irradiation, phosphine-free, and air condition. A novel flow-injection chemiluminescence-based method has been developed for determination of superoxide dismutase (SOD) activity. An in-vitro superoxide anion generation xanthine/xanthine oxidase stable source was established on line with FIA/CL-detection apparatus, for measuring SOD activity. This method can detect SOD in the linear range of 0.002–2.00 U mL–1 with a detection limit of 0.001 U mL–1. Another method for detection of superoxide anion is based on the luminol–FeCl3 chemiluminescence (CL) reaction. This method was used to evaluate superoxide release and SOD activity in rats treated with the traditional Chinese herb Pulsatilla chinensis, which resulted in high clearance of hepatitis B virus (HBV) after treatment of a hepatitis B patient. Interestingly, we found that treatment with Pulsatilla chinensis can specifically increase superoxide release by liver tissues and, at the same time, slightly increase extracellular SOD (ECSOD) activity in plasma; in particular it can markedly increase MnSOD activity in mitochondria in liver tissue. This work revealed a possible mechanism whereby Pulsatilla chinensis prevents possible infection (for example HBV) by specifically increasing superoxide release in the liver and increasing MnSOD activity to minimize superoxide-mediated toxicity. 相似文献

In this article, polymer network liquid crystal (PNLC) grating/Fresnel lens is fabricated by holography. The exposure light pattern for the grating is obtained by interfering two planar wave fronts, while the Fresnel pattern is achieved by interfering a planar wave front and a spherical wave front. Owing to the alignment effect and anchoring power of polymer network, the holographic PNLC grating achieves improved diffraction efficiency, and remarkably reduced operation voltage (reduced by 80%) compared with holographic polymer-dispersed-liquid-crystal and holographic polymer-stabilised blue-phase liquid-crystal gratings, while maintaining submillisecond response. Moreover, it achieves high spatial frequency with a 2-μm grating period, thanks to the holographic fabrication. The holographic PNLC Fresnel lens also exhibits attractive electro-optical properties. 相似文献

It is believed that next-generation passive optical networks (PONs) are required to provide flexible and various services to users in a cost-effective way. To address this issue, for the first time, this paper proposes and demonstrates a novel wavelength-division-multiplexed PON (WDM-PON) architecture to simultaneously support three types of services: 1) wireless access traffic, 2) optical virtual passive network (VPN) communications, and 3) conventional wired services. In the optical line terminal (OLT), we use two cascaded Mach-Zehnder modulators (MZMs) on each wavelength channel to generate an optical carrier, and produce the wireless and the downstream traffic using the orthogonal modulation technique. In each optical network unit (ONU), the obtained optical carrier is modulated by a single MZM to provide the VPN and upstream communications. Consequently, the light sources in the ONUs are saved and the system cost is reduced. The feasibility of our proposal is experimentally and numerically verified. 相似文献
