High magnification microscopic system focusing on transparent liquid/liquid interface formed in thin-layer two-phase microcell.

A thin-layer two-phase microcell was fabricated without an adhesive, which showed no fluorescence and resisted acids. The lower hollow of the microcell was thin (0.18 mm), which was in the range of the working distance of the used objective of high magnification (60x) and high numerical aperture (1.2). A reflection method using probe light was newly employed for focusing the objective on a transparent dodecane/water interface strictly with an inverted microscope. The system was applied to in situ fluorescence microscopic measurements of the dynamics of single DNA molecules (165600 base pairs) at the dodecane/water interface.     

3-H-YDROXYRETINAL AS A CHROMOPHORE OF Drosophila melanogaster VISUAL PIGMENT ANALYZED BY HIGH-PRESSURE LIQUID CHROMATOGRAPHY

Abstract— The chromophore of the visual pigment of the fruit fly Drosophila melanogaster was studied by high-pressure liquid chromatography (HPLC). Oxime derivatives of the chromophore were successfully analyzed with a more polar solvent than has been used for retinaloximes. A preceding monochromatic irradiation resulted in reversible interchanges in the amount among geometric isomers of the oximes that reflected the photointerconversion of rhodopsin and metarhodopsin of the peripheral photoreceptors (Rl-R6). Authentic all- trans -3-h-ydroxyretinal was used to identify the chromophore from metarhodopsin. Results demonstrated that the chromophore of the visual pigment in Drosophila is 3-h-ydroxyretinal as was proposed in larger flies.     

Deuterium relaxation time ofα-d-tryptophan included in cyclodextrin host molecules

Deuterium relaxation times ofd- andl--d-tryptophan included in -cyclodextrin derivatives were directly measured by deuterium NMR spectroscopy. The results showed that the molecular motion of the tryptophan molecule was strongly restricted even in the cavity of unmodified -cyclodextrin and the additional recognition groupings — ammonium and carboxylate — on -cyclodextrin did not affect the molecular motion of tryptophan, though the association constants were significantly enhanced.Deceased on March 22, 1987.     

Dynamic kinetic resolution of amino acid amide catalyzed by D-aminopeptidase and alpha-amino-epsilon-caprolactam racemase

Amino acid amide racemizing activity was discovered in alpha-amino-epsilon-caprolactam (ACL) racemase (EC 5. 1. 1. 15) from Achromobacter obae. The enzymatic synthesis of d-alanine from l-alanine amide has been demonstrated by use of d-aminopeptidase (DAP; EC 3. 4. 11. 19) from Ochrobactrum anthropi C1-38 and ACL racemase. The conversion of 45 mM l-alanine amide was carried out at 30 degrees C for 7 h; l-alanine amide was completely converted to d-alanine, and no l-alanine was detected. The result of successive enzymatic reaction shows that the combination of ACL racemase and DAP can be applied for dynamic kinetic resolution of dl-amino acid amides to yield d-amino acids.     

Unusual enlargment of magnetic interactions in the lowest excited states of nitroxide radical chromium(III) complexes as revealed by magnetic circular dichroism and luminescence

Magnetic circular dichroism and NIR luminescence of nitroxide radical complexes, [Cr(III)(beta-diketonato)(2)(NIT2py or IM2py)]PF(6), demonstrate that the energy gaps between the singlet ((1)L(D)) and triplet ((3)L(D)) spin coupled levels in the lowest excited (2)E(g),(2)T(1g) states are much larger than those in the ground state. This is the first observation of magnetic interactions in the excited states of radical complexes, which could be elucidated in terms of the exchange mechanism.     

Determination of gold in silver, copper, lead, selenium and anode slime by atomic-absorption spectrometry

A simple and sensitive combined solvent-extraction and atomic-absorption spectrometric method has been developed for the determination of gold in silver, copper, lead, selenium and anode slime. Samples are decomposed with hydrochloric and nitric acids, and gold is extracted as the trioctylmethylammonium-gold bromide complex and determined by atomic-absorption spectrometry by direct spraying of the extract into the flame. Optimal conditions for the extraction and determination of gold have been established. As little as 0.5 mug of gold in a sample can be determined. The extraction of gold from hydrochloric or hydrobromic acid solution with trioctylamine or trioctylmethylammonium chloride (or bromide) has also been investigated.     

Cationic carbohydroxylation of alkenes and alkynes using the cation pool method

The reactions of an N-acyliminium ion pool with alkenes and alkynes gave gamma-amino alcohols and beta-amino carbonyl compounds, respectively, after treatment with H(2)O/Et(3)N. The present reaction serves as an efficient method for cationic carbohydroxylation of alkenes and alkynes. When vinyltrimethylsilane was used as an alkene, the reaction was highly diastereoselective and served as an access to an enantiomerically pure alpha-silyl-gamma-amino alcohol. A plug and play microfluidic device

Chip-to-world interface is a major issue in the field of microfluidics and its applications. We developed a plug and play microfluidic device composed of a fluid driving unit and a polymer chip containing microfluidic channels and reservoirs. The one and only connection of the device to the external world is a set of electric control lines for the driving unit. Just putting the reagents and samples onto the reservoirs, the chip can be operated for chemical or biochemical reaction and analysis. We demonstrate here that silicon-based micropumps embedded in the present device allow us to achieve flexible fluidic manipulations with minimum time delay and dead volume.
