Summary The activities of diamine oxidases (DAO) from submaxillary glands, parotid glands and kidneys, from human serum during the pregnancy and from pig serum and of DAO from pea seedlings and the activities of urease and histidine ammonia-lyase have been determined by measuring NH4 + released from the corresponding substrates. Ammonia has been determined with the aid of glutamate dehydrogenase reaction in the coupled optic test. The specifity and sensitivity of the method, by which still 3×10–9 Mol of NH4 + could be measured, is excellent, the performance simple and rapid. The sensitivity could be enhanced by fluorometric determination of NAD+ in strong alkali solution. A method for the preparation of tissues for the spectrophotometric test of DAO is described. The importance of DAO determination in different diseases is briefly discussed.
Zusammenfassung Die Aktivität von Diaminoxydasen aus Speicheldrüsen und Nieren, aus menschlichem Serum während der Schwangerschaft und Schweineserum und aus Erbsenkeimlingen sowie die Aktivität der Urease und Histidase wurden anhand der NH3-Abspaltung aus ihren entsprechenden Substraten bestimmt. Das entstehende Ammoniak wurde dabei mit Hilfe der Glutamatdehydrogenasereaktion im gekoppelten optischen Test gemessen. Spezifität und Empfindlichkeit der Methode, mit der noch 3 · 10–9 Mol NH4 + bestimmt werden können, sind ausgezeichnet, die Durchführung einfach und rasch. Die Empfindlichkeit konnte außerdem noch durch die fluorimetrische Bestimmung von NAD+ gesteigert werden. Für die Bestimmung der Diaminoxydasen im Gewebe wird eine Methode der Aufarbeitung des Gewebes angegeben, die praktisch keinen Verlust an Gesamtaktivität gegenüber dem Ausgangshomogenat aufweist. Die Bedeutung der Diaminoxydasebestimmung für die Klinik wird kurz diskutiert.

Summary Several procedures for the isolation of histamine from tissues and body fluids were examined. For the fluorometric assay of histamine concentrations higher than 1 g/g tissue or ml blood the extraction into n-butanol as well as the cation-exchange chromatography on Dowex 50W-X8 proved suitable. The determination of histamine in whole blood of dogs gave valid results only after chromatography on Dowex 50, in human plasma and gastric juice only after the combined application of both purification methods. Histamine was identified by thin-layer chromatography on cellulose, by inactivation in the presence of purified diamine oxidase from pig kidney and purified histamine methyltransferase from guinea-pig brain, by the biological activities and by the fluorescence spectra of the condensation product between histamine and o-phthaldialdehyde. Histamine concentrations in tissues and body fluids of man and some laboratory animals are reported.
Fluorimetrische Bestimmung von Histamin in Geweben und Körperflüssigkeiten: Wahl eines geeigneten Reinigungsverfahrens und Identifizierung im ng-Bereich
Zusammenfassung Verschiedene Isolierungsverfahren für Histamin zur fluorimetrischen Bestimmung wurden geprüft. Für Histamingehalte von mehr als 1 g/g Gewebe oder Milliliter Vollblut eignet sich sowohl die Butanolextraktion als auch die Ionenaustausch-Chromatographie an Dowex 50W-X8, für Vollblut des Hundes nur das letztere Verfahren, für Plasma und Magensaft des Menschen nur die Kombination der beiden Isolierungsmethoden. Histamin wurde identifiziert durch Dünnschicht-Chromatographie auf Cellulose, Umsatz durch angereicherte Diaminoxydase und Histaminmethyltransferase, durch die Fluorescenzspektren nach Kondensation mit o-Phthaldialdehyd und durch seine biologische Aktivität. Die Histamingehalte in Geweben und Körperflüssigkeiten des Menschen und einiger Versuchstiere werden mitgeteilt.

A novel synthesis of thiazolo[2,3-b]quinazolines 4(a–e), pyrido[2′,3′:4,5]thiazolo[2,3-b]quinazolines {5(a–e), 6(a–e), and 7(a–e)}, pyrano[2′,3′:4,5]thiazolo[2,3-b]quinazolines 8(a–e), and benzo[4,5]thiazolo[2,3-b]quinazoloine9(a–e) derivatives starting from 2-(Bis-methylsulfanyl-methylene)-5,5-dimethyl-cyclohexane-1,3-dione 2 as efficient α,α dioxoketen dithioacetal is reported and the synthetic approaches of these types of compounds will provide an innovative molecular framework to the designing of new active heterocyclic compounds. In our study, we also present optimization of the synthetic method along with a biological evaluation of these newly synthesized compounds as antioxidants and antibacterial agents against the bacterial strains, like S. aureus, E. coli, and P. aeruginosa. Among all the evaluated compounds, it was found that some showed significant antioxidant activity at 10 μg/mL while the others exhibited better antibacterial activity at 100 μg/mL. The results of this study showed that compound 6(c) possessed remarkable antibacterial activity, whereas compound 9(c) exhibited the highest efficacy as an antioxidant. The structures of the new synthetic compounds were elucidated by elemental analysis, IR, 1H-NMR, and 13C-NMR.  相似文献

Reaction of a Cyclic Bis(amino)germylene with Germaniumazides: Trapping-Reactions of Unstable Germa-Imines . The cyclic bis(amino)germylene 1 reacts with different germaniumazides of the type Me2Si(NtBu)2Ge(R)N3 (R = Me ( 2 ), tBu ( 3 ), N(SiMe3)2 ( 4 ), R = N3 ( 5 )). With the exception of 4 all azides lose dinitrogen when treated with 1 and the GeII center coordinates the α-nitrogen of the azide group. It seems to be reasonable to assume a transient germaimine (nitride) which is trapped by further reaction with the azide molecules 2 and 5 or by reaction with the solvent pyridine ( 3 ). In the case of 2 the germatetrazole [Me2Si(NtBu)2]GeN4[Ge(NtBu)2SiMe2]2 ( 6 ) is formed, the tetrazole nitrogens being exclusively substituted by germanium atoms (point symmetry of the molecule Cs(m)). When 1 is treated with 5 a tris(germa)amine [Me2Si(NtBu)2Ge(N3)]3N ( 8 ) is formed, which has an azide group attached to each Ge-atom. X-ray analysis reveals that the nine nitrogen atoms of the azide groups are coplanar with the trigonal planar Ge3N moiety (crystallographic symmetry: 3/m). The reaction of 1 with 3 is very surprising: the pyridine in the product Me2Si(NtBu)2Ge(C5H4N)? N(H)Ge(tBu)(NtBu)2SiMe2 ( 7 ) is bonded via an α-carbon atom while the remaining hydrogen has added to the nitride-nitrogen. 6 crystallizes in the monoclinic system space group C2/m, a = 24.306(9), b = 10.933(6), c = 19.420(9) Å, β = 91.81(2)° and Z = 4. 7 crystallizes in the hexagonal system space group P63/m with a = b = 16.73(1), c = 11.006(8) Å, γ = 120° and Z = 2, and 8 crystallizes in the monoclinic system space group P21/n, a = 11.341(6), b = 26.086(9), c = 13.244(7) Å, β = 98. Generic computer simulations using empiric interatomic potentials suggest a new, collective mechanism that could be responsible for mixing at heteroepitaxial interfaces. Even if single adsorbate atoms diffuse by hopping on the substrate surface and do not mix at the terraces, two-dimensional islands formed by nucleation may become unstable above a certain critical size and explode upwards forming clusters of several atomic layers. This process is accompanied by strong distortions of the underlying atomic layers, and on soft materials it can result in surface etching and incorporation of substrate atoms into the islands.  相似文献

Spectroscopic techniques are increasingly used for field laser applications in industry and research. Under field conditions complex gas sensors cannot be considered as stable and therefore drift characterization is a key issue to distinguish between sample data and sensor drift. In this paper the history of von Neumann’s two-sample variance and Allan variance stability investigations in the field of frequency metrology and the relationship to wavelet analysis are reviewed. The concept has been used to characterize accuracy and precision of spectroscopic data in the time domain and practical guidelines for the interpretation of σ/τ plots are presented. Two topics relevant for spectroscopic measurements are discussed: First, the optical fringe effect, which is present in any spectrometer, limits the precision and accuracy of spectroscopic measurements by forming time dependent background structures superimposed to the signal under investigation. The two-sample variance is used to characterize optical etalons and long-term drift using σ/τ plots. Second, the short-term instrument stability characteristic in the presence of atmospheric turbulence is discussed. This is important for laser-based gas monitors measuring the turbulent transport of trace gases between the biosphere and the atmosphere using the eddy-covariance technique. It will be shown how the spectral characteristics of turbulence in the Kolmogorov inertial subrange can be identified in the time domain and how the effect of optical fringes can be separated from atmospheric signals.  相似文献

Some acidic chiral templates were used to prepare open tubular (OT) molecule imprinted polymer (MIP) capillary columns to explore the effects of molecular structures of templates on chiral recognition capabilities and to verify the feasibility of the general MIP preparation protocol introduced in the previous study. The templates are phenyl carboxylic acids and their derivatives. Optimization was carried out for chiral separation of template enantiomers for each MIP column through varying pH and composition of eluent. It was found that the preparation protocol can be successfully applied for the appropriate templates with functional groups fulfilling the three-points interaction rule. The chiral separation performances were quite satisfactory for MIPs of such templates although they are yet inferior to the separation performances of the MIP columns fabricated with the templates of profen drugs (2-arylpropionic acids with a large substituent on the phenyl ring). Subtle variations of the template molecular structures have been found to be critical to enhance chiral recognition ability of the resultant MIP column.  相似文献
