The interaction of ethidium bromide (2,7-diamino-10-ethyl-9-phenylphenanthridinium bromide; EB) with double stranded (ds) calf thymus DNA and thermally denatured single stranded (ss) DNA was studied in solution and at the electrode surface by means of transfer voltammetry using a carbon paste electrode (CPE) as working electrode in 0.2 M acetate buffer, pH 5.0. As a result of intercalation of this dye between the base pairs of dsDNA, the characteristic peak of dsDNA, due to the oxidation of guanine residues, decreased and after a particular concentration of EB a new peak at +0.81 V appeared, probably due to the formation of a complex between dsDNA and EB. The non-intercalated EB gives another peak, but at an increased concentration of the dye. A similar behaviour was observed during the interaction of the dye with ssDNA.Furthermore, the interaction of EB with ds, ss and supercoiled (sc) DNA was studied at the hanging mercury drop electrode (HMDE) surface by means of alternating current voltammetry in 0.3 M NaCl and 50 mM sodium phosphate buffer (pH 8.5) as supporting electrolyte. dsDNA yields a smaller peak at −1.42 V (peak III) compared to the one yielded by ssDNA, since the latter is a relaxed and more accessible form. By addition of EB into the buffer solution an increase of peak III was observed in the dsDNA form as well as in ssDNA resulting from their interaction with EB. Furthermore, the appearance of peak III in covalently closed circular scDNA after exposure to increasing concentrations of EB is a result of the introduction of ‘free ends’ in DNA affecting its structural integrity.  相似文献

This work reports the simultaneous determination of Cd(II), Pb(II) and Zn(II) at the low μg l−1 concentration levels by square wave anodic stripping voltammetry (SWASV) on a bismuth-film electrode (BFE) plated in situ. The metal ions and bismuth were simultaneously deposited by reduction at −1.4 V on a rotating glassy carbon disk electrode. Then, the preconcentrated metals were oxidised by scanning the potential of the electrode from −1.4 to 0 V using a square-wave waveform. The stripping current arising from the oxidation of each metal was related to the concentration of each metal in the sample. The parameters for the simultaneous determination of the three metals were investigated with the view to apply this type of voltammetric sensor to real samples containing low concentrations of metals. Using the selected conditions, the limits of detection were 0.2 μg l−1 for Cd and for Pb and 0.7 μg l−1 for Zn at a preconcentration time of 10 min. Finally, BFE's were successfully applied to the determination of Pb and Zn in tapwater and human hair and the results were in satisfactory statistical agreement with atomic absorption spectroscopy (AAS).  相似文献

Summary The spontaneous adsorption of the Co2+ complex with 1,10-phenanthroline on HMDE was utilized for the development of a voltammetric analytical method employing phase selective alternating current, with first harmonic AC1 in phase, =0°, voltammetry. The dependence of the analytical voltammetric response on physical and chemical parameters in various supporting electrolytes was investigated in detail. Optimum conditions for Co2+ determination were established. The method is very sensitive and selective. The determination limit is 205 ng/l Co2+ (adsorption time, tads=20 min). The precision and accuracy were evaluated: 1.84 g/l Co2+, RSD=3.24%, RE=–0.54%; for 205 ng/l Co2+, RSD=4.85%, RE=–3.90%. The results showed good reproducibility and reliabiality. The method was applied to natural waters, wines, mussel tissue and aquatic algae certified reference material.  相似文献

Mercury film plated on a glassy carbon electrode modified with multiwalled carbon nanotubes (MF/MWNTs/GCE) was used for the analysis of single (ss) and double stranded (ds) DNA, as well as for Jurkat genomic DNAs methylated to different degrees. The results indicated that the DNA molecules adsorbed quite strongly on the MF/MWNTs/GCE surface allowing ex situ adsorption and produce well developed peaks (due to cytosine and adenine) by using adsorptive stripping (Ads, ex situ) square wave voltammetry (SWV). Also, SWV of Jurkat DNA mixtures methylated to different degrees revealed a linear decrease of the peak height with increasing methylation indicating an increase of structural rigidity.  相似文献

The hydrazino complex {methoxo[4-phenylbutane-2,4-dione(p-nitrobenzoyl)hydrazonato(2-)]oxovanadium(V)}, VO(p-NO2bhbzac)OCH3, (1), has been prepared by the direct reaction of bis(benzoylacetonato) oxovanadium(IV), VO(bza)2, with p-NO2-C6H4C(O)NHNH2, p-NO2bh, in CH3OH. The resulting compound contains benzoylacetone-(p-NO2)benzoyl hydrazone as tridentate Schiff base-type ligand and OCH3 group as Lewis base, both ligated to vanadium. The crystals are orthorhombic, with Z = 8, space group Pbca, a = 11.699(5) Å, b = 14.035(5) Å, c = 22.564(5) Å, R1 = 0.0756 and wR2 = 0.1302. The crystal structure demonstrated the square-pyramidal geometry of the VOoxo(ONO)O coordination sphere with the oxo ligand at the apical position. The electronic absorption spectra revealed a ligand-to-metal charge-transfer (LMCT) band in the near UV region at max = 23,700 cm–1 (B = 5640 dm3 mol–1 cm–1) in CH3CN, max = 23,420 cm–1 (B = 5550 dm3 mol–1 cm–1) in DMSO, and max near 26,950 (sh) cm–1 (B = 10,550 dm3 mol–1 cm–1) in CH2Cl2. The FT-IR spectra of (1) show the characteristic strong (V = O) stretching vibration at 993 cm–1 and support the view that the oxovanadium complex is pentacoordinated and monomeric.  相似文献

Summary Experimental Factors Estimation in Nephelometric Procedures The nephelometric determinations of low concentrations of sulphate ions are usually influenced by a variety of experimental factors such as presence of stabilizing agents, acids, interferences, equilibration and stirring time etc. The present study makes an attempt to estimate the weight of 6 input factors at different levels of sulphate concentrations by the use of the economic experimental design known as random balance method. It is found that some of the input factors (presence of stabilizing media, acidity) are of higher rank than others (equilibration time or barium ions concentration). Using these results it is easy to find optimal conditions for micro-determination of sulphates by nephelometry.  相似文献

A selective, sensitive and reliable voltam- metric method for the simultaneous determination of MoS2-4 and MoO2-4 has been developed. The reduction of HgMoS4 at –0.43 V and the reduction of Mo as complex of MoO2-oxine2 at –0.63 V in acetate buffer at pH 4.9 are utilized for the simultaneous determination by differential pulse cathodic stripping voltametry (DPCSV). Cadmium and lead form complexes with MoS2-4, but do not interfere with the determination. The limit of detection is 0.59 g/l Mo for MoS2-4 and 1.8 g/l Mo for MoO2-4. The R.S.D at a concentration level of 20 g/l is 4.7% for MoO2-4 and 3.6% for MoS2-4. The method is applied to spiked blood plasma samples for the determination of free tetrathiomolybdates and molybdates. Additionally, labile tetrathiomolybdates and molybdates can be determined in spiked blood plasma after treatment of the sample with 0.15 mol/lKCN and ultrafiltration.  相似文献

A sequential injection analysis (SIA) manifold was incorporated with a monolithic strong anion-exchanger disk for on-line drug-protein interaction studies. The antibiotic ciprofloxacin (CF) was selected as a model drug compound. The separation principle was based on the strong retention of bovine serum albumin (BSA) on the monolithic strong anion-exchanger and the liberation/release of the free form of the drug. Elution of the retained BSA was easily achieved by delivering a different mobile phase via the SIA manifold. The type of functional group of the monolithic support, the breakthrough volume and the injected volumes of CF and BSA were studied and optimized. The influence of the variation of incubation time was studied in on-line binding assays. Scatchard plot was employed to obtain the number of binding sites and the equilibrium binding constants. For the off-line study of the CF-BSA binding, two binding classes were determined with constants of (3.16+/-0.21)x10(6)M(-1) and (1.27+/-0.48)x10(4)M(-1) and 6.1+/-1.3 and 17.8+/-3.9 binding sites per class, respectively. In non-equilibrium binding experiments the binding rate constant was k(1)=785 M(-1)min(-1). All measurements were monitored with fluorescence (lambda(ext)=300 nm, lambda(em)=460 nm) and spectrophotometric detection (lambda=280 nm). To evaluate the accuracy of the developed method the obtained results were compared versus ultrafiltration experiments and were found in good agreement.  相似文献
