Semiconducting polythiophenes display electroactive properties which make them excellent candidates for applications as electroactive materials. Ability to undergo doping and to switch between different oxidation states allow tuning the chemical and physical properties of polythiophenes. Furthermore, the ability to integrate polythiophenes into copolymers, hybrid composites with metals and inorganic materials make them useful in electronic and biomedical applications. The capability to vary the properties of the final material with low production cost and high processability into thin films makes polythiophenes desirable materials for many applications. © 2017 Wiley Periodicals, Inc. Detection of anions in complex aqueous media is a fundamental challenge with practical utility that can be addressed by supramolecular chemistry. Biomolecular hosts such as proteins can be used and adapted as an alternative to synthetic hosts. Here, we report how the mutagenesis of the β-bulge residues (D137 and W138) in mNeonGreen, a bright, monomeric fluorescent protein, unlocks and tunes the anion preference at physiological pH for sulfate, resulting in the turn-off sensor SulfOFF-1. This unprecedented sensing arises from an enhancement in the kinetics of binding, largely driven by position 138. In line with these data, molecular dynamics (MD) simulations capture how the coordinated entry and gating of sulfate into the β-barrel is eliminated upon mutagenesis to facilitate binding and fluorescence quenching.  相似文献

Poly{2,6‐bis(thiophene‐2‐yl)‐4,8‐bis(5‐dodecylthiophen‐2‐yl)benzo[1,2‐b :4,5‐b' ]dithiophene} [poly(Th‐bDTBDT‐Th)] was successfully synthesized through Stille coupling polymerization. The addition of the thiophene spacer groups between the benzodithiophene units resulted in improved performance in optoelectronic devices. This was attributed to the reduced lamellae stacking distance in thin film with prominent π–π stacking peak indicating close assembly of poly(Th‐bDTBDT‐Th). Spacing between the benzodithiophene units in poly(Th‐bDTBDT‐Th) helped the close packing of dodecyl chains and generated improved π stacking interaction. For poly(Th‐bDTBDT‐Th), the measured average field effect mobility was 2.32 × 10?3 cm2 V?1 s?1 and average hole mobility in vertical direction was 2.92 × 10?5 cm2 V?1 s?1. Heparan sulfate (HS) contains variably repeating disaccharide units organized into high- and low-sulfated domains. This rich structural diversity enables HS to interact with many proteins and regulate key signaling pathways. Efforts to understand structure-function relationships and harness the therapeutic potential of HS are hindered by the inability to synthesize an extensive library of well-defined HS structures. We herein report a rational and expedient approach to access a library of 27 oligosaccharides from natural aminoglycosides as HS mimetics in 7–12 steps. This strategy significantly reduces the number of steps as compared to the traditional synthesis of HS oligosaccharides from monosaccharide building blocks. Combined with computational insight, we identify a new class of four trisaccharide compounds derived from the aminoglycoside tobramycin that mimic natural HS and have a strong binding to heparanase but a low affinity for off-target platelet factor-4 protein.  相似文献

(S)-1,1'-Binaphth-2-ol (BINOL) in combination with ZnEt(2), Ti(O(i)Pr)(4), and biscyclohexylamine was found to catalyze the highly enantioselective (83-95% ee) addition of various 1,3-diynes to aldehydes of diverse structures. This method provides a convenient pathway to generate a number of optically active dienediynes as the acyclic precursors to polycyclic compounds. The chiral dienediynes undergo highly chemoselective Pauson-Khand (PK) cycloaddition in benzaldehyde by using [Rh(cod)Cl](2) as the catalyst in the presence of rac-BINAP. High diastereoselectivity (up to >20:1) has also been achieved with the chiral dienediyne substrates containing a bulky substituent adjacent to the chiral center. In the presence of the Grubbs II catalyst, ring-closing enyne metathesis of the PK cycloaddition products led to the formation of the desired 5,5,7- and 5,5,8-fused tricyclic compounds. Further highly diastereoselective Diels-Alder reaction of a 5,5,7-tricyclic compound with maleic anhydride produced a 5,5,7,6-polycyclic product. The asymmetric synthesis of polycyclic compounds from optically active dienediynes has established a novel and efficient synthetic route to the structural framework of many biologically significant molecules.  相似文献
