We have developed a rapid and sensitive method for immunomagnetic separation (IMS) of Salmonella along with their real time detection via PCR. Silica-coated magnetic nanoparticles were functionalized with carboxy groups to which anti-Salmonella antibody raised against heat-inactivated whole cells of Salmonella were covalently attached. The immuno-captured target cells were detected in beverages like milk and lemon juice by multiplex PCR and real time PCR with a detection limit of 104 cfu.mL?1 and 103 cfu.mL?1, respectively. We demonstrate that IMS can be used for selective concentration of target bacteria from beverages for subsequent use in PCR detection. PCR also enables differentiation of Salmonella typhi and Salmonella paratyphi A using a set of four specific primers. In addition, IMS—PCR can be used as a screening tool in the food and beverage industry for the detection of Salmonella within 3–4 h which compares favorably to the time of several days that is needed in case of conventional detection based on culture and biochemical methods.
The method uses silica coated magnetic nanoparticles immobilized with anti-Salmonella antibody for immunomagnetic separation of Salmonella from beverages followed by detection by multiplex PCR (mPCR) and real time PCR (qPCR). This methodology contributes to rapid screening and accurate detection of Salmonella contaminations in beverages.  相似文献

A series of benzenesulfonamide-appended [1,2,3]-triazole hybrids was synthesized by using [3 + 2] cycloaddition of primary, secondary, and tertiary sulfonamide azides with various phenoxymethylacetylenes under click reaction conditions. After structural characterization, the compounds were subjected to in-silico absorption, distribution, metabolism, excretion and toxicity (ADMET) screening to evaluate their drug-likeness and other pharmacokinetic parameters. Furthermore, their in vitro antiplasmodial potential was assessed against Plasmodium falciparum (3D7) strain, and some of the synthesized compounds displayed promising antimalarial potency. On cytotoxicity evaluation using MTT cell viability assay, the most active candidate N-(4,6-dimethylpyridin-2-yl)-4-(4-(4-nitrophenoxy)methyl)-1H-[1,2,3]-triazol-1-yl)benzenesulfonamide ( 14 ; IC50 6.2 μg/mL) demonstrated CC50 7.5 μg/mL against human hepatocarcinoma (HUH-7) cells.  相似文献

In the generation of discrete tones by subsonic impinging jets, there exists a difference of opinion as how the feedback is achieved, i.e., the path of the feedback acoustic waves is whether inside the jet or outside the jet? The only available model (Tam and Ahuja model) for the prediction of an average subsonic jet impingement tone frequency assumes that the upstream part of the feedback loop is closed by an upstream propagating neutral wave of the jet. But, there is no information about the plate geometry in the model. The present study aims at understanding the effect of the plate geometry (size and co-axial hole in the plate) on the self-excitation process of subsonic impinging jets and the path of the acoustic feedback to the nozzle exit. The present results show that there is no effect of plate diameter on the frequency of the self-excitation. A new type of tones is generated for plates with co-axial hole (hole diameter is equal to nozzle exit diameter) for Mach numbers 0.9 and 0.95, in addition to the axisymmetric and helical mode tones observed for plates without co-axial hole. The stability results show that the Strouhal number of the least dispersive upstream propagating neutral waves match with the average Strouhal number of the new tones observed in the present experiments. The present study extends the validity of the model of Tam and Ahuja to a plate with co-axial hole (annular plate) and by doing so, we indirectly confirmed that the major acoustic feedback path to the nozzle exit is inside the jet.  相似文献

Present work deals with the preparation of TiO2 thin films of different thicknesses by PVD technique using an electron beam and to characterize the films for oxygen gas detection. The films were characterized using optical transmission measurements, XRD and atomic force microscopy. From the spectral data, the extinction, absorption coefficient and refractive index of the films are evaluated and reported. The optical band gap energy varies between 3 and 3.68 eV. The XRD pattern confirms the formation of polycrystalline anatase structure of Titanium with preferred orientation of (110) plane. The AFM images indicate the presence of coarse and fine grains with uniform as well as smooth surfaces over the entire range of the analyzed surface. Response characteristics of TiO2 thin films for oxygen gas detection are studied, which indicate a low response time of 120 seconds and high sensitivity of 16 at the operating temperature 450 °C. We are describing immunochromatographic test strips with smart phone-based fluorescence readout. They are intended for use in the detection of the foodborne bacterial pathogens Salmonella spp. and Escherichia coli O157. Silica nanoparticles (SiNPs) were doped with FITC and Ru(bpy), conjugated to the respective antibodies, and then used in a conventional lateral flow immunoassay (LFIA). Fluorescence was recorded by inserting the nitrocellulose strip into a smart phone-based fluorimeter consisting of a light weight (40 g) optical module containing an LED light source, a fluorescence filter set and a lens attached to the integrated camera of the cell phone in order to acquire high-resolution fluorescence images. The images were analysed by exploiting the quick image processing application of the cell phone and enable the detection of pathogens within few minutes. This LFIA is capable of detecting pathogens in concentrations as low as 105 cfu mL?1 directly from test samples without pre-enrichment. The detection is one order of magnitude better compared to gold nanoparticle-based LFIAs under similar condition. The successful combination of fluorescent nanoparticle-based pathogen detection by LFIAs with a smart phone-based detection platform has resulted in a portable device with improved diagnosis features and having potential application in diagnostics and environmental monitoring. Aryl alcohol oxidase (AAO) is an extracellular flavoenzyme involved in lignin degradation by white rot fungi. Screening of lignolytic and AAO activity from twenty different fungal species were carried out. Among them, seven species showed lignolytic activity and three of them (Pleurotus ostreatus, Pleurotus eous, and Pleurotus platypus) were found to be AAO positive. Maximal AAO activity was observed in batch cultures of P. ostreatus and was found to be induced by aromatic amino acids and aryl alcohols up to a level of 289 U/l. Purification of AAO was carried out by three-phase partitioning (TPP). The 67 kDa enzyme was purified up to 10.19-fold by TPP with an overall recovery of 10.95%. Optimum pH and temperature for P. ostreatus AAO activity was found to be around 6 and 40 °C, respectively. From the LB plot, K m value of AAO for oxidizing veratryl alcohol was determined to be 0.6 mM. Results of the study indicate that P. ostreatus is the best producers of AAO, and they could be employed as promising fungal species for biotechnological applications.  相似文献

In the present investigation, the recently developed, simple, robust, and powerful metaheuristic symbiotic organism search (SOS) algorithm was used for simulation of J-V characteristics and optimizing the internal parameters of the dye-sensitized solar cells (DSSCs) fabricated using electrospun 1-D mesoporous TiO2 nanofibers as photoanode. The efficiency (η =?5.80%) of the DSSC made up of TiO2 nanofibers as photoanode is found to be ~ 21.59% higher compared to the efficiency (η =?4.77%) of the DSSC made up of TiO2 nanoparticles as photoanode. The observed high efficiency can be attributed to high dye loading as well as high electron transport in the mesoporous 1-D TiO2 nanofibers. Further, the validity and advantage of SOS algorithm are verified by simulating J-V characteristics of DSSC with Lambert-W function.  相似文献

The development of hydrogen evolution catalysts based on nonprecious metals is essential for the practical application of water‐splitting devices. Herein, the synthesis of Co9S8?MoS2 hierarchical nanoboxes (HNBs) as efficient catalysts for the hydrogen evolution reaction (HER) is reported. The surface of the hollow cubic structure was organized by CoMoS4 nanosheets formed through the reaction of MoS42? and Co2+ released from the cobalt zeolite imidazole framework (ZIF‐67) templates under reflux in a mixture of water/ethanol. The formation process for the CoMoS4 HNB structures was characterized by TEM images recorded at various reaction temperatures. The amorphous CoMoS4 HNBs were converted through sequential heat treatments into CoSx?MoS2 and Co9S8?MoS2 HNBs. Owing to their unique chemical compositions and structural features, Co9S8?MoS2 HNBs have a high specific surface area (124.6 m2 g?1) and superior electrocatalytic performances for the HER. The Co9S8?MoS2 HNBs exhibit a low overpotential (η10) of 106 mV, a low Tafel slope of 51.8 mV dec?1, and long‐term stability in an acidic medium. The electrocatalytic activity of Co9S8?MoS2 HNBs is superior to that of recently reported values, and these HNBs prove to be promising candidates for the HER.  相似文献

A new superionic conducting transparent phosphate glasses with composition Li(4+x)TixNb1?xP3O12 (0 ≤ x ≤ 1.0) were prepared by rapid melt quenching. As quenched samples were characterized by X-ray powder diffraction, differential scanning calorimetric and Fourier transform infrared spectroscopy studies. These glasses were found to have high thermal stability parameter and Li4NbP3O12 has been found to have high glass forming ability. Electrical properties of the present glasses were studied by impedance and dielectric spectroscopy in the frequency range 10 Hz–3 MHz in the temperature range 323–523 K. Arrhenius behavior has been observed for all the glass in conductivity, dielectric loss and conductivity relaxation and their activation energies are explained and reported.  相似文献
