Systematic experimental investigations of heteropoly compounds, particularly their structure and activity, led to the conclusion that most of their characteristics are governed by the presence of protons and protonic entities. Special attention has been paid to two forms of 12-tungstophosphoric acid: hexahydrate (WPA-6) and dehydrated phase (WPA-0). It was postulated that in WPA-6 dynamic equilibrium of protonic entities exists, and that dehydrated phase is stabilized by protons. To confirm the role of the “free protons” or “proton gas” derived on the basis of thermal, structural and spectroscopic experimental studies, we carried out also ab initio calculations on a number of systems containing ion. We were not able to perform direct calculations on the real systems investigated experimentally since the structure of heteropoly compounds is too complex. However, it has been found that ion in WPA-6 definitely is not planar and the results obtained indirectly support the postulated dynamic equilibrium, i.e. possibility of existing of free protons.  相似文献

The H-bond ((h3)J(NC')) and peptide bond ((1)J(NC')) scalar couplings establish connectivity of the electronic structure in the H-bond chains of proteins. The correlated changes of (h3)J(NC') and (1)J(NC') couplings extend over several peptide groups in the chains. Consequently, the electronic structure of the H-bond chains can affect (h3)J(NC') in a manner that is independent of the local H-bond geometry. By taking this into account, and by using a more complete set of H-bond geometry parameters, we have predicted (h3)J(NC') couplings in the H-bond chains with deviations commensurate to the standard deviations of the experimentally determined values. We have created a comprehensive database of (h3)J(NC') and (1)J(NC') couplings by measuring the coupling constants in ubiquitin (alphabeta-fold) intestinal fatty acid binding protein (beta-barrel) and carp parvalbumin (alpha-helical).  相似文献

Summary. Compounds of phosphotungstic acid (WPA) containing the amino acids alanine (WPAAla) or glycine (WPAGly) as counter cations were synthesized and characterized by elemental analysis, thermal analysis, and IR spectroscopy. Cellular toxicity was assessed by the trypan blue exclusion method, and the antiviral activity of WPA and the modified WPA compounds was tested against herpes simplex viruses (HSV) type 1 and type 2. Biological assays indicate that the newly synthesized compounds exhibit no evident cytotoxic effects on Vero cells and negligible antiviral activity against HSV-1 and HSV-2.  相似文献

High-performance liquid chromatography was developed for further separation of double-stranded (ds) RNAs obtained by CF-11 cellulose chromatography from plants infected with satellite associated cucumber mosaic virus. Fractions separated by monolithic polymer column, especially applicable for nucleic acid analyses, were identified electrophoretically and confirmed with a polymerase chain reaction test. Once standardized, the method has revealed clear evidence of satellite presence without precipitation and electrophoresis. According to demonstrated sensitivity, its application in the preliminary diagnostics of field samples is also predictable. Principally, it can be used as a powerful preparative approach resulting in highly pure satellite dsRNA for further analyses.  相似文献

Free sugar radicals. V. Deoxyhydroxylaminosugar derivatives and related compounds We describe several synthetic routes to deoxyhydroxylaminosugar derivatives of the type Glyc-N(OH)-R where Glyc stands for a sugar moiety linked by any of its C-atoms except the anomeric one and R for one of the following substituants: H-atom, acyl, phosphoryl groups, aminoacid or sugar residues. Compounds of the above structure are potentially close analogs, homoisosteres, ? NOH? replacing ? O? , of biochemically important molecules. Under aerobic conditions, solutions of these derivatives contain minute concentrations of the corresponding nitroxide radicals which do not decrease significantly the resolution of the NMR. spectra but render these compounds usable as a new kind of spin labels. Spectroscopic properties (1H-NMR., 13C-NMR., ESR.) of some of these compounds are reported.  相似文献

Deoxy-hydroxylamino-sugar Derivatives and Corresponding Diglycosylnitroxides Radicals A number of sugar aldonitrones, including C,N-diglycosylnitrones, and ketonitrones have been treated with Grignard reagents or cyanide anion leading to the corresponding deoxy-hydroxylamino-sugars. On oxidation (air, H5IO6 or PbO2), these compounds gave the corresponding nitroxide radicals whose ESR. spectra are reported. Analogues of disaccharides, in which the interglycosidic O-bridge is replaced by a hydroxyimino group, have been obtained by reacting a partially blocked sugar bearing a free hemiacetal group either with a deoxy-hydroxylaminosugar or with hydroxylamine, followed by reaction with an aldehydosugar and a reducing agent (NaBH4). These reactions represents the key synthetic steps for the oligosaccharide-type synthesis of deoxy-hydroxyimino-oligosaccharides. Their oxidation yielded the corresponding nitroxide radicals whose ESR. spectra gave information on the conformation about the ‘interglycosidic’ bridge. This type of compounds should constitute useful spin markers for biological studies.  相似文献
