Summary Anti-HIV enantiomeric 1H, 3H-thiazolo[3, 4-a] benzimidazoles have been stereospecifically analyzed by elution on a column of cellulose tris-(4-methyl-phenylbenzoate)ester adsorbed on macroporous silica (ChiralcelR OJ).The enantiomeric resolution of the compounds examined is linked to a complex and competitive contribution of different factors.  相似文献

Summary A high-performance liquid chromatographic method is reported for the resolution of the enantiomers of a series of fused -lactams (2,7-diaza-3-oxo[3.3.0]octan-6-ones) with probable anti-HIV and anticancer activity. Resolution was achieved on a Chiralcel® OD column, cellulose tris-(3,5-dimethylphenyl carbamate) adsorbed on macroporous silica gel; mixtures ofn-hexane and isopropyl alcohol in different proportions were used as the mobile phase. Interferonopathies are rare genetic conditions defined by systemic inflammatory episodes caused by innate immune system activation in the absence of pathogens. Currently, no targeted drugs are authorized for clinical use in these diseases. In this work, we studied the contribution of sulforaphane (SFN), a cruciferous-derived bioactive molecule, in the modulation of interferon-driven inflammation in an immortalized human hepatocytes (IHH) line and in two healthy volunteers, focusing on STING, a key-component player in interferon pathway, interferon signature modulation, and GSTM1 expression and genotype, which contributes to SFN metabolism and excretion. In vitro, SFN exposure reduced STING expression as well as interferon signature in the presence of the pro-inflammatory stimulus cGAMP (cGAMP 3 h vs. SFN+cGAMP 3 h p value < 0.0001; cGAMP 6 h vs. SFN+cGAMP 6 h p < 0.001, one way ANOVA), restoring STING expression to the level of unstimulated cells. In preliminary experiments on healthy volunteers, no appreciable variations in interferon signature were identified after SFN assumption, while only in one of them, presenting the GSTM1 wild type genotype related to reduced SFN excretion, could a downregulation of STING be recorded. This study confirmed that SFN inhibits STING-mediated inflammation and interferon-stimulated genes expression in vitro. However, only a trend towards the downregulation of STING could be reproduced in vivo. Results obtained have to be confirmed in a larger group of healthy individuals and in patients with type I interferonopathies to define if the assumption of SFN could be useful as supportive therapy.  相似文献

The applications of SERS in therapeutic drug monitoring, or other fields of analytical chemistry, require the availability of sensitive sensors and experimental approaches that can be implemented in affordable ways. In this contribution, we show the production of cost-effective SERS sensors obtained by depositing Lee-Meisel Ag colloids on filter paper either by natural sedimentation or centrifugation. We have characterized the morphological and plasmonic features of the sensors by optical microscopy, SEM, and UV-Vis spectroscopy. Such sensors can be used to quantify by SERS the anti-epileptic drug Perampanel (in the concentration range 1 × 10−4–5 × 10−6 M) by spinning them during the micro-Raman measurements on the top of a custom device obtained from spare part hard disk drives. This approach minimizes laser-induced heating effects and allows averaging over the spatial non-uniformity of the sensor.  相似文献

We demonstrate that effective traveling-wave excitation of high-gain amplifiers requires velocities that are remarkably slower than the velocity of light. Experiments with a femtosecond-laser-pumped molecular hydrogen laser exhibit pronounced enhancement of the intensity if an excitation velocity that is slower than the velocity of light is employed. These results are directly scalable to shorter wavelengths, paving the way for a more effective pump setup for x-ray lasers.  相似文献

We show that a gas-to-liquid phase transition at zero temperature may occur in a coherent gas of bosons in the presence of competing nonlinear effects. This situation can take place in atomic systems like Bose-Einstein condensates in alkali gases with two-body and three-body interactions of opposite signs, as well as in laser beams which propagate through optical media with Kerr (focusing) and higher order (defocusing) nonlinear responses. The liquefaction process takes place in the absence of any quantum effect and can be formulated in the framework of a mean field theory, in terms of the minimization of a thermodynamic potential. We study from a thermodynamic point of view all the stationary solutions of the cubic-quintic nonlinear Schrödinger equation which describes our system. We show that solitonic localized solutions connect the gaseous and liquid phases. Furthermore, we also perform a numerical simulation in the presence of linear gain and three-body recombination where a rich dynamics, including the emergence of self-organization behavior, is found.  相似文献

We report the observation of the Debye–Scherrer diffraction using electron pulses emitted from a fs-laser plasma. Titanium sapphire laser pulses with 1.6 mJ/45 fs at 1 kHz are focused on a moving steel tape at close to normal incidence. The laser plasma generated ejects a large number of electrons in the direction of polarization, with a continuous energy spectrum extending up to 100 keV. Selecting an energy range of these electrons and scattering them on a thin aluminium sample generates a “streaked” diffraction pattern with unique features.  相似文献

We report the generation of diffraction patterns using X-ray pulses from a fs-laser plasma focused by an X-ray capillary lens to a spot size smaller than 100 μm. A thin moving iron tape is irradiated at 10 Hz with 200 mJ/130 fs titanium–sapphire laser pulses. [ Fe]Kα (λ=0.194 nm) radiation emitted from the rear side of the tape is focused to a spot by the capillary lens resulting in an intensity enhancement of about 1600. The ability of this system to effectively allow diffraction from samples of micron size is demonstrated by producing well-illuminated diffractograms from a Si(111) crystal with good signal to noise ratio obtained in only about 10 s. The different path lengths of propagation in the lens induce an angle-encoded delay of the converging X-ray pulse and thus provide the possibility of realizing a novel ultra-fast streak camera. By using a 0.7% prepulse that precedes the main pulse by 5 ns and applying a total pump energy of 100 J or less, the J=0-1 lasing is at least one order of magnitude higher than the non-lasing background. For the 32.6-nm line of Ti, the 25.5-nm line of Fe, and the 23.1-nm line of Ni, gain coefficients of (±) 4.20.4cm-1, (±) 3.90.3cm-1, and (±) 3.60.6cm-1, respectively, were measured for 2.4-cm-long curved targets, resulting in gain–length products of ∼10. Angle-resolved spectra indicate a beam divergence of 3 mrad (FWHM), typically. The space-resolved spectra show that the J=0-1 lasing lines are emitted from an approximately 60-μm-wide (FWHM) plasma region, whereas the nearby continuum emission is produced in a considerably broader plasma region of ∼250 μm. Lasing at 25.5 nm in neon-like iron was observed at a pump power as low as 180 GW (∼9 TW/cm2), with, however, considerable shot-to-shot scatter in the absolute laser output.
