The mesoionic compound, 1,4,5-triazole-2-thiolate, is a versatile ligand, capable of binding metal centres in different ways. The present study shows how it reacts with the bridging platinum(II) complex μ-dichlorobis[(chlorotriethylphosphineplatinum(II)], [Pt2(PEt3)2Cl4], originating a bimetallic complex. Both the sulphur atom and nitrogen 3 behave as basic sites, coordinating different platinum centres. An interesting isomerization process, shown by 31P{1H} n.m.r. spectroscopy, occurs at the S-bonded platinum moiety, resulting in a fluxional behaviour of the complex. It is also shown to target CT-DNA with moderate binding affinity.To the memory of Dr. Wagner M. Teles (1971–2004).  相似文献

Anisotropic NMR parameters, such as residual dipolar couplings (RDCs), residual chemical shift anisotropies (RCSAs) and residual quadrupolar couplings (RQCs or ΔνQ), appear in solution‐state NMR when the molecules under study are subjected to a degree of order. The tunable alignment by reversible compression/relaxation of gels (PMMA and p‐HEMA) is an easy, user‐friendly, and very affordable method to measure them. When using this method, a fraction of isotropic NMR signals is observed in the NMR spectra, even at a maximum degree of compression. To explain the origin of these isotropic signals we decided to investigate their physical location inside the NMR tube using deuterium 1D imaging and MRI micro‐imaging experiments. It was observed that after a certain degree of compression the gels start to buckle and they generate pockets of isotropic solvent, which are never eliminated. The amount of buckling depends on the amount of cross‐linker and the length of the gel.  相似文献

The electrochemical reduction of the antidepressant drug fluoxetine was investigated by cyclic, linear sweep, differential pulse and square wave voltammetry using a hanging mercury drop electrode in alkaline buffer solution in water and in a water/acetonitrile mixed solvent. Cyclic voltammograms in aqueous solution showed very strong adsorption of fluoxetine on the electrode with formation of a compact film. The effect of addition of different percentages of acetonitrile on the voltammetric response was evaluated. It is shown that acetonitrile protects the electrode surface, thus preventing the adsorption of fluoxetine as a compact film, although reduction occurs at more negative potentials. Adsorption was used to accumulate the drug onto the electrode surface. The adsorbed species were measured voltammetrically by reduction at -1.3 V in an aqueous 0.05 M Ringer buffer, pH 12, 20% acetonitrile v/v. Linear calibration graphs were obtained in the range 0.52-5.2 M. The quantification of fluoxetine in pharmacological formulations existing in the market was performed using adsorptive square wave cathodic stripping voltammetry. and compared with data from UV spectrophotometry. The method is simple and not time-consuming. A comparative high performance liquid chromatography assay with UV detection was performed. Recovery data for both methods are reported.  相似文献

This work aimed to immobilize the antibody anti-galectin-3 onto polysiloxane–polyvinyl alcohol (POS-PVA) support, to evaluate its capacity to capture the serum antigen galectin-3 and to quantify by ELISA the antigen levels in sera from patients with prostatic adenocarcinoma (PA) and benign prostatic hyperplasia (BPH) and healthy individuals. Also, for comparative effect, the galectin-3 expression in the prostate tissue through immunohistochemistry was evaluated. The optical density (galectin-3 level) values established for the sera from PA and BPH patients were lower compared with those found for the healthy individuals. Galectin-3 immunohistochemically showed a significant increase and reduction of the cytoplasmatic protein expression in BPH and PA, respectively, compared with the normal prostate. These results showed that POS-PVA disks could be used as solid phase to immobilize serum galectins and in immunoassays procedures for the correspondent IgG anti-galectins detection in human sera.  相似文献

The main purpose of this study was to validate and compare Mean Glandular Dose (MGD) values obtained using Monte Carlo simulations with experimental values obtained from Entrance Surface Dose (ESD) and depth dose measurements performed in a Hospital mammography unit. ESD and depth dose were measured using ThermoLuminescent Dosimeters (TLDs), and a tissue equivalent mammography phantom recommended by the American College of Radiology (ACR). Measurements and Monte Carlo simulations were also compared with the MGD calculated using the Automatic Exposure Control (AEC) system of the mammographic unit. In the simulations the Doppler energy broadening effect was also taken into account. The simulated ESD are about 5%–10% higher than the measured ESD values. The deviation between the measured and simulated MGD values in the phantom is of about 15%. The MGD evaluated using the AEC system is smaller both with respect to the Monte Carlo simulation and experimental result by a factor of about 15% and 25% respectively. Moreover the BackScatter Factor (BSF) estimated by Monte Carlo simulations was used for the MGD calculation according to the Wu’s method. Finally the inclusion of the energy broadening effect on MGD calculation produces negligible variations on the simulated results.  相似文献
