A simple method for the determination of the aminoglycoside antibiotic sisomicin or its 1-N-ethyl derivative, netilmicin in whole blood, using dried blood spots (DBSs) on filter-paper punched discs has been developed. Sisomicin or netilmicin in the DBSs were recovered most effectively in 0.5 M Na2HPO4 using ultrasonication. The eluates from the DBSs were treated by ultrafiltration for deproteinization and subjected to pre-column fluorescent derivatization using o-phthalaldehyde and beta-mercaptopropionic acid in 0.05 M KH2PO4-borate buffer (pH 9.0), followed by determination by reversed-phase high-performance liquid chromatography. The detection limits of sisomicin and netilmicin in the DBSs on punched discs (10.1 microliters of whole blood) were 0.053 and 0.50 micrograms per ml of whole blood, respectively (signal-to-noise ratio greater than or equal to 2). The method permits a simple collection of blood at the microlitre level and should prove particularly useful for monitoring sisomicin and netilmicin in blood at therapeutic levels in geriatric and paediatric patients.  相似文献

Reaction of Pd(TMEDA)(CH(3))(2) [TMEDA = tetramethylethylenediamine] with fluoroalkyl iodides R(F)I affords a series of square planar Pd(II) complexes Pd(TMEDA)(CH(3))(R(F)) [R(F) = CF(2)CF(3) (9), CFHCF(3) (10), CH(2)CF(3) (11)], presumably by oxidative addition followed by reductive elimination of CH(3)I. The solid-state structures of each compound have been determined by single crystal X-ray diffraction studies, allowing the effect of increasing alpha-fluorination on the structural trans-influence of alkyl ligands to be examined. In these compounds there is no significant difference observed in the trans-influence of the three fluorinated alkyl ligands toward the trans-N atom, although a significant cis-influence on the neighboring methyl ligand is apparent. Oxidative addition of the same series of fluoroalkyl ligands to the corresponding Pt(TMEDA)(CH(3))(2) affords octahedral Pt(IV) complexes trans-Pt(TMEDA)(CH(3))(2)(R(F))I [R(F) = CF(2)CF(3) (12), CFHCF(3) (13), CH(2)CF(3) (14)] as the kinetic products. In each case, subsequent isomerization to the corresponding all cis-isomers is observed; in the case of 13, the stereocenter at the alpha-carbon results in two diastereomeric cis-isomers, which are formed at different rates. The molecular structures of 13 and its more stable all cis-isomer 16b have been crystallographically determined. Kinetic studies of the trans-cis isomerization reactions show the mechanism to involve a polar transition state, presumably involving iodide dissociation, followed by rearrangement of the cation, and iodide recombination. High dielectric solvents increase the rate, but solvent coordinating ability has no effect. Dissolved salts (LiI, LiOTf) show normal accelerative salt effects, with no inhibition in the case of added iodide, consistent with the formation of an intimate ion pair intermediate. The kinetic parameters show that the trans-effects of fluoroalkyl ligands in these compounds follow the order expected from the relative sigma-donor properties of the ligands, with CF(2)CF(3) < CFHCF(3) < CH(2)CF(3).  相似文献

To generate command surfaces for all-optical switching, highly ordered polymeric Langmuir-Blodgett-Kuhn (LBK) multilayers were fabricated onto silicon substrates and gold-coated optical glass slides from novel azobenzene-bearing polyamic acid systems. Pronounced Bragg peaks and well-defined Kiessig fringes observed in the X-ray reflectivity measurement for samples on silicon substrates indicate that these films possess a regularly repeated Y-type LBK multilayer structure and ultrasmooth surfaces. Fourier transform infrared (FT-IR) spectra taken by grazing incidence reflection suggest specific orientations of the functional groups in the layers. The excellent film-forming properties of the polyamic acid allow for a smooth buildup of several hundreds of layers of the LBK films onto gold-coated glass slides, which in turn allows for determining the geometrical thickness and the anisotropic refractive indices of the films by using optical waveguide spectroscopy. Interestingly, the probe laser beam induced a distinct fluorescence signal from the films, which remained even after the film underwent a thermal imidization process at 160 degrees C for 8 h in vacuo. LBK films fabricated from these compounds can be successfully applied for all-optically switching the alignment of liquid crystals by irradiation with light of different wavelengths.  相似文献

“Laser-assisted magnetic recording”, in which a recording media is heated by a laser beam while writing data, is attracting attention as a technology that enables a recording density of 1 Tb/in.2. There exists another technology for media in which the recording layer is constructed with many small projections that enable high magnetic coercivity. This is called “patterned media”. For developing hard disk drives using these methods, we developed a simulator that analyzes the optical intensity distribution from the optical head for laser-assisted recording and the temperature profile on the patterned media. The simulator calculates the optical model using the finite-difference time-domain (FDTD) method. The thermal analysis of the three-dimensional model allows fast calculations using the alternating direction implicit (ADI) method. The heat source distribution data for thermal analysis is calculated in order to use the results of optical analysis. The optical and thermal analyses of the laser-assisted recording model were investigated with the simulator.  相似文献

Optical near-field plasmon-enhancement techniques have made marked contributions to the field of subwavelength optical beam spot generation for photonic devices. However, the metal material used is not sufficiently heat resistant for high-temperature operating requirements, such as laser-assisted magnetic recording storage. Therefore, it is preferable to use a dielectric material as a heat source for this application. To realize a subwavelength optical beam spot size using a dielectric material, we extensively analyze a leaky mode dielectric waveguide with a thin core. Finally, we show a new ridge-type dielectric waveguide using a leaky mode operation for a subwavelength beam spot.  相似文献

Rapid overlay of chemical structures (ROCS) is a method that aligns molecules based on shape and/or chemical similarity. It is often used in 3D ligand-based virtual screening. Given a query consisting of a single conformation of an active molecule ROCS can generate highly enriched hit lists. Typically the chosen query conformation is a minimum energy structure. Can better enrichment be obtained using conformations other than the minimum energy structure? To answer this question a methodology has been developed called CORAL (COnformational analysis, Rocs ALignment). For a given set of molecule conformations it computes optimized conformations for ROCS screening. It does so by clustering all conformations of a chosen molecule set using pairwise ROCS combo scores. The best representative conformation is that which has the highest average overlap with the rest of the conformations in the cluster. It is these best representative conformations that are then used for virtual screening. CORAL was tested by performing virtual screening experiments with the 40 DUD (Directory of Useful Decoys) data sets. Both CORAL and minimum energy queries were used. The recognition capability of each query was quantified as the area under the ROC curve (AUC). Results show that the CORAL AUC values are on average larger than the minimum energy AUC values. This demonstrates that one can indeed obtain better ROCS enrichments with conformations other than the minimum energy structure. As a result, CORAL analysis can be a valuable first step in virtual screening workflows using ROCS.  相似文献

Cytochrome P450 (CYP) 3A4, 2D6, 2C9, 2C19, and 1A2 are the most important drug-metabolizing enzymes in the human liver. Knowledge of which parts of a drug molecule are subject to metabolic reactions catalyzed by these enzymes is crucial for rational drug design to mitigate ADME/toxicity issues. SMARTCyp, a recently developed 2D ligand structure-based method, is able to predict site-specific metabolic reactivity of CYP3A4 and CYP2D6 substrates with an accuracy that rivals the best and more computationally demanding 3D structure-based methods. In this article, the SMARTCyp approach was extended to predict the metabolic hotspots for CYP2C9, CYP2C19, and CYP1A2 substrates. This was accomplished by taking into account the impact of a key substrate-receptor recognition feature of each enzyme as a correction term to the SMARTCyp reactivity. The corrected reactivity was then used to rank order the likely sites of CYP-mediated metabolic reactions. For 60 CYP1A2 substrates, the observed major sites of CYP1A2 catalyzed metabolic reactions were among the top-ranked 1, 2, and 3 positions in 67%, 80%, and 83% of the cases, respectively. The results were similar to those obtained by MetaSite and the reactivity + docking approach. For 70 CYP2C9 substrates, the observed sites of CYP2C9 metabolism were among the top-ranked 1, 2, and 3 positions in 66%, 86%, and 87% of the cases, respectively. These results were better than the corresponding results of StarDrop version 5.0, which were 61%, 73%, and 77%, respectively. For 36 compounds metabolized by CYP2C19, the observed sites of metabolism were found to be among the top-ranked 1, 2, and 3 sites in 78%, 89%, and 94% of the cases, respectively. The computational procedure was implemented as an extension to the program SMARTCyp 2.0. With the extension, the program can now predict the site of metabolism for all five major drug-metabolizing enzymes with an accuracy similar to or better than that achieved by the best 3D structure-based methods. Both the Java source code and the binary executable of the program are freely available to interested users.  相似文献

We present spontaneous emission control of a core-shell CdSe/ZnS nanoparticle array assembled with polymer ultrathin films consisting of polymer nanosheets on a silver grating substrate, which served as a unique and simple photonic cavity. The grating-coupled waveguide modes enabled 10(3) order luminescence enhancement and one-fourth spectral narrowing. The light emission from a CdSe/ZnS nanoparticle array can be controlled by tuning the film thickness of hybrid polymer nanoassemblies, which provides multiple emission performance with good tuning ability from red to green at low-power continuous wave laser excitation (~μW).  相似文献

We use results derived in the framework of the replica approach to study the liquid-glass thermodynamic transition. The main results are derived without using replicas and applied to the study of the Lennard-Jones binary mixture introduced by Kob and Andersen. We find that there is a phase transition due to the entropy crisis. We compute both analytically and numerically the value of the phase transition point T(K) and the specific heat in the low temperature phase.  相似文献
