The emergence of drug-resistant hepatitis B virus (HBV) has been reported in patients with prolonged administration of lamivudine, which is a potent drug for the prevention of HBV infection. Lamivudine-resistant HBV has several types of mutations at the YMDD motif of its DNA polymerase. We successfully demonstrated that monitoring the hybridization behavior in nanostructured reverse micelles enables us to detect single nucleotide polymorphisms (SNPs). With the aid of reverse micelles, a model 40-mer oligonucleotide containing a single-base substitution was clearly distinguished from the normal, complementary oligonucleotide. In addition, we extended this technique to a high-throughput analysis. The results obtained with a 96-well micro-plate reader indicated the possibility of SNPs detection toward multiple samples of patients.  相似文献

A tensammetric method is proposed for the determination of microgram amounts of vanadium, based on catalysis of the oxidation of o-aminophenol with sodium chlorate in acidic solution (pH 2.0). The oxidation product gives a very sensitive tensammetric wave; under optimum conditions, the wave-height is proportional to the concentration of va vanadium. From 0.2 to 3.0 mug of vanadium can be determined in a final volume of 50 ml. Mo(VI), W(VI), Mn(VII), Ce(IV) and large amounts of Al(III) and Fe(III) cause positive errors, and Hg(II) and thiosulphate negative errors. Interference from Fe(III), Al(III) and Cu(II) can be eliminated by solvent with oxine at about pH 8.0.  相似文献

Abstract

In this paper, an analysis of two dimensional smectic layer structure is presented to clarify the layer distortion between two chevrons directed towards opposite directions. From numerical computations, it is found that a parallelogram region appears between two antiparallel chevrons for a sufficiently large molecular tilt and thick sample. The elastic free energy density is found to concentrate near the boundaries of the kinks of the layer structure. In addition the field effect on such a pair of chevrons is presented to show that it is efficient to apply an electric field so as to obtain a quasi-bookshelf structure.  相似文献

Perpendicular magnetic recording tape medium with laminated soft magnetic underlayers was successively formed on a 4.5 μm thin plastic film by the facing targets sputtering method at room temperature. The tape medium showed extremely high SNR values with sharp transitions in playback tests by a drum tester, compared to a linear format commercial tape medium. An areal density of 45.0 Gb/in.2 of perpendicular tape medium was confirmed by a high-resolution contact tester. These results suggest that the perpendicular recording tape medium by the facing targets sputtering method is a promising candidate for a tape cartridge of over 50-TB.  相似文献

The present study was designed to show the applicability of scanning ion conductance microscopy (SICM) for imaging different types of biological samples. For this purpose, we first applied SICM to image collagen fibrils and showed the usefulness of the approach-retract scanning (ARS)/hopping mode for such samples with steep slopes. Comparison of SICM images with those obtained by AFM revealed that the ARS/hopping SICM mode can probe the surface topography of collagen fibrils and chromosomes at nanoscale resolution under liquid conditions. In addition, we successfully imaged cultured HeLa cells, with 15 μm in height by ARS/hopping SICM mode. Because SICM can obtain non-contact (or force-free) images, delicate cellular projections were visualized on the surface of the fixed cell. SICM imaging of live HeLa cells further demonstrated its applicability to study the morphological dynamics associated with biological processes on the time scale of minutes under liquid conditions. We further applied SICM for imaging the luminal surface of the trachea and succeeded in visualizing the surface of both ciliated and non-ciliated cells. These SICM images were comparable with those obtained by scanning electron microscopy. Although the dynamic mode of AFM provides better resolution than the ARS/hopping mode of SICM in some samples, only the latter can obtain contact-free images of samples with steep slopes, rendering it an important tool for observing live cells as well as unfixed or fixed soft samples with complicated shapes. Taken together, we demonstrate that SICM imaging, especially using an ARS/hopping mode, is a useful technique with unique capabilities for imaging the three-dimensional topography of a range of biological samples under physiologically relevant aqueous conditions.  相似文献

As the maximum speed of high speed trains increases, the effect of aeroacoustic noise on the sound level on the ground becomes increasingly important. In this paper, the distribution of dipole sound sources at the bogie section of high speed trains is predicted numerically. The three-dimensional unsteady flow around a train is solved by the large eddy simulation technique. The time history of vortices shows that unstable shear layer separation at the leading edge of the bogie section sheds vortices periodically. These vortices travel downstream while growing to finally impinge upon the trailing edge of the section. The wavelength of sound produced by these vortices is large compared to the representative length of the bogie section, so that the source region can be regarded as acoustically compact. Thus a compact Green's function adapted to the shape can be used to determine the sound. By coupling the instantaneous flow properties with the compact Green's function, the distribution of dipole sources is obtained. The results reveal a strong dipole source at the trailing edge of the bogie section where the shape changes greatly and the variation of flow with time is also great. On the other hand, the bottom of the bogie section where the shape does not change, or the leading edge and boundary layer where the variation of flow with time is small, cannot generate a strong dipole source.  相似文献

A facile chemical process has been developed for the preparation of magnetic FeCo nanoparticles. The FeCo nanoparticles were mono-dispersed, obtained by the safe and ecofriendly method, possessed saturation magnetization up to 187 emu/g, and demonstrated excellent chemical stability. In this work, we have studied how to control Fe/Co ratio by variation of precursor ratio, and how to vary particle size from 9.3 to 12.3 nm by surfactant amount used. The cytotoxicity of as-synthesized nanoparticles was investigated after coating with the poly(methyl methacrylate-co-butyl acrylate) by the emulsion process and the results demonstrated high biocompatibility. Similarly, the same synthesis method was used with the single precursor FeO(OH) or Co3O4. The results showed that this method can also fabricate 10 nm mono-dispersed spherical Fe3O4 particles and self-assembly Co nanoneedles.  相似文献
