Mutation detection in the drug-resistant hepatitis B virus polymerase gene using nanostructured reverse micelles.

The emergence of drug-resistant hepatitis B virus (HBV) has been reported in patients with prolonged administration of lamivudine, which is a potent drug for the prevention of HBV infection. Lamivudine-resistant HBV has several types of mutations at the YMDD motif of its DNA polymerase. We successfully demonstrated that monitoring the hybridization behavior in nanostructured reverse micelles enables us to detect single nucleotide polymorphisms (SNPs). With the aid of reverse micelles, a model 40-mer oligonucleotide containing a single-base substitution was clearly distinguished from the normal, complementary oligonucleotide. In addition, we extended this technique to a high-throughput analysis. The results obtained with a 96-well micro-plate reader indicated the possibility of SNPs detection toward multiple samples of patients.     

InGaAs/InAlAs Five Layer Asymmetric Coupled Quantum Well (FACQW) for Ultra-Wideband Ultrafast Optical Modulators

An InGaAs/InAlAs five-layer asymmetric coupled quantum well (FACQW) is expected to show very large electrorefractive index change. n in a wideband transparency region. Band structures of the FACQW are analyzed with Luttinger-Kohn Hamiltonian. The electrorefractive characteristics of the FACQW are discussed.     

Breakage of λ-DNA by inorganic tin and organotin compounds as environmental pollutants

In proportion to the environmental pollution problems caused by organotin compounds, the genotoxicities of tin compounds in the environments have become of interest so as to estimate their safety in recent years. In this work, isolated λ-DNA (double-strand DNA) was incubated with inorganic tin(II) and tin(IV) and five organotin compounds [n-butyltin trichloride, di(n-butyltin) dichloride, methyltin trichloride, dimethyltin dichloride and trimethyltin chloride] in reaction systems both with and without hydrogen peroxide (H₂O₂) content. The tin compounds tested in this study did not induce DNA breakage in the absence of hydrogen peroxide. Divalent inorganic tin (SnCl₂) and tetravalent inorganic tin (SnCl₄) caused DNA breakage in the presence of hydrogen peroxide (10 mM), and the DNA damage activity of inorganic tin was much more potent in divalent inorganic tin (SnCl₂) than in tetravalent inorganic tin (SnCl₄). Divalent inorganic tin (SnCl₂) induced DNA breakage in a concentration-dependent fashion at concentrations greater than 0.1 mM of SnCl₂ in the presence of hydrogen peroxide (10 mM). DNA breakage was not caused by n-butyltin compounds and methyltin compounds either in the presence or in the absence of hydrogen peroxide.     

Characteristic curved structure derived from collagen-containing tubular giant vesicles under static magnetic field

Collagen-containing tubular giant vesicles (Col–tGVs) under a high static magnetic field were found to constitute characteristic curved structures (e.g. circular, 8-figure, and hairpin-loop), which were sustained by polymerization of collagen units inside the tGV. We ascribed the formation of these structures to the elasticity of the tGV and to the competing diamagnetic anisotropies between collagen and the tGV. These curved structures can be well expressed by the equation of elastica.     

Synthesis of pyridinium ylide complexes of methylcobaloxime and crystal structure determination of [benzoyl(1-pyridinio)methanide]bis(dimethylglyoximato)methylcobalt benzene solvate

Summary Pyridinium ylide complexes of methylcobaloxime were synthesized by the treatment of an ylide with Co(Hdmg)₂ Me(SMe₂). The crystal structure of one of the complexes, [Co(Hdmg)₂Me C₅H₅NCHCOPh]C₆H₆ has been determined by x-ray diffraction techniques. The crystals are monoclinic, space group P2₁/c, witha = 10.456(5),b = 11.079(4),c = 24.58(1) Å, = 99.58(6), V = 2808 ų, Z = 4. The Co-C (ylide) bond distance is 2.18 Å and Co-C(methyl) 2.04 Å. C(ylide)-Co-C(methyl) bond angle is 174.9°. The crystal, i.r. and¹H n.m.r. data suggest that thetrans-influence of the ylide ligands is larger than that of py, Melm, OH₂ or PPh₃.     

NMR and MO Studies on the Molecular Structure of a Fluoranthene-Benzene Complex

Summary. Fluoranthene (FA) forms a 1:1 van der Waals complex with benzene in cyclohexane. The ¹H NMR spectrum of this complex shows that the FA moiety in the complex state has five kinds of hydrogen atoms and that the ¹H NMR peaks assigned to the protons attached to the naphthalene skeleton are largely shifted to higher magnetic field on complex formation with benzene. These observations indicate that the complex takes the structure of C_S symmetry, in which the benzene molecule mainly interacts with the electronic system localized on the naphthalene moiety of FA. The present ab initio calculations reproduce well the ¹H NMR spectral shifts mentioned above and the experimentally predicted C_S structure of the complex. According to the PPP calculations for the electronic absorption spectral changes on the complex formation, the FA-benzene complex is considered to take a sandwich type structure.     

Structure of aszonalenin,a new metabolite of aspergillus zonatus

Aszonalenin ($\text{1}$) has been isolated from $\text{Aspergillus zonatus}$ together with LL-S490β ($\text{2}$) and the structure of $\text{1}$ has been established by spectroscopic evidences and chemical transformation.     

Carbon dioxide fixation by microalgae photosynthesis using actual flue gas discharged from a boiler

To mitigate CO₂ discharged from thermal power plants, studies on CO₂ fixation by the photosynthesis of microalgae using actual exhaust gas have been carried out. The results are as follows.

Inter- and intramolecular activation of aromatic C-H bonds by diphosphine and hydrido-bridged dinuclear iridium complexes

Reactions of [(Cp*Ir)2(mu-dmpm)(mu-H)2]2+ (1) with NaOtBu in aromatic solvent at room temperature give [(Cp*Ir)(H)(mu-dmpm)(mu-H)(Cp*Ir)(Ar)]+ [Ar = Ph (3), p-Tol (4a), m-Tol (4b), 2-furanyl (5a), 3-furanyl (5b)] via intermolecular aromatic C-H activation. Treatment of [(Cp*Ir)2(mu-dppm)(mu-H)2]2+ (2) with base (Et2NH) results in intramolecular C-H activation of the phenyl group in the dppm ligand to give [(Cp*Ir)(H){mu-PPh(C6H4)CH2PPh2}(mu-H)(Cp*Ir)]+ (6). The structures of 3, 5a, and 6 have been determined by X-ray diffraction methods.
